This study investigated the potential of linalool as an anaesthetic during transportation of common carp, Cyprinus carpio. The fish were transported at a loading density of ~103 g/L for 3 hr in 12 plastic bags (3 L water and 6 L pure oxygen) divided into four triplicated treatments: control (without linalool), L50 (50 mg linalool/L), L100 (100 mg linalool/L) and L200 (200 mg linalool/L). After 3‐hr transportation, serum physiological responses and water physico‐chemical parameters were compared among the treatments. Results showed that water total and un‐ionized ammonia increased and dissolved oxygen decreased in all treatments after transportation. Water total ammonia and dissolved oxygen levels in the linalool‐treated bags were significantly lower than the control. After transportation, the control fish showed significant elevation in serum cortisol and glucose levels; however, the levels were significantly lower than the linalool‐treated fish. All fish showed similar serum lactate levels, significantly lower than the value obtained before transportation. The control fish showed serum urea levels similar to the level obtained before transportation; however, the linalool‐treated fish had significantly higher serum urea levels compared to the control and before transportation levels. After transportation, all fish had decreased serum chloride and sodium levels compared to the before transport level. Overall, the present results showed that linalool is not beneficial for carp transportation in plastic bags, because it reacts with water oxygen, increases stress in fish, interferes with ammonia excretion and has no benefits in preventing ion loss.  相似文献

Identification of resistance resources of sugar beet germplasm against Polymyxa betae has always been a critical concern among sugar beet breeders. In the present study, two different methods including the DAS-enzyme-linked immunosorbent assay (ELISA) test and a nanobiosensor method based on florescent resonance transfer energy (FRET) were compared in order to achieve resistant germplasm. More specifically, 58 sugar beet germplasm as well as two negative and two positive controls were cultivated in infested soil under greenhouse conditions. The contamination level or in another words sensitivity and specificity observed based on the ELISA readings was inaccurately lower in comparison with the investigated nanobiosensor. Moreover, the nanobiosensor was 70 folds less time consuming compared to the ELISA method, for the immuno-reaction was much faster and no sample treatment steps were required. As a result, the quantum dots-FRET-based nanobiosensor investigated herein could well suit the task of everyday screening of resistance resource and could be efficiently used in breeding programs.  相似文献

Background:

Existence of bacterial host-cell DNA contamination in biopharmaceutical products is a potential risk factor for patients receiving these drugs. Hence, the quantity of contamination must be controlled under the regulatory standards. Although different methods such as hybridization assays have been employed to determine DNA impurities, these methods are labor intensive and rather expensive. In this study, a rapid real-time PCR test was served as a method of choice to quantify the E. coli host- cell DNA contamination in widely used recombinant streptokinase (rSK), and alpha interferon (IFN-α) preparations.

Methods:

A specific primer pair was designed to amplify a sequence inside the E. coli 16S rRNA gene. Serial dilutions of DNA extracted from E. coli host cells, along with DNA extracted from Active Pharmaceutical Ingredients of rSK, and IFN-α samples were subjected to an optimized real-time PCR assay based on SYBR Green chemistry.

Results:

The test enabled us to detect a small quantity of genomic DNA contamination as low as 0.0002 pg in recombinant protein-based drugs. For the first time, this study showed that DNA contamination in rSK and IFN-α preparation manufactured in Pasteur Institute of Iran is much lower than the safety limit suggested by the US FDA.

Conclusion:

Real-time PCR is a reliable test for rapid detection of host-cell DNA contamination, which is a major impurity of therapeutic recombinant proteins to keep manufacturers’ minds on refining drugs, and provides consumers with safer biopharmaceuticals. Gypsum-bonded particleboard (GBPB) panels were made from various mixtures of particles of bagasse (Saccharum officinarum L.) and wheat straw (Triticum aestivum L.), bonded with different ratios of particle/gypsum. This study examined the feasibility of bagasse and wheat straw particles in the production of GBPB. One-layer experimental GBPBs with a density of 1.05 or 1.20 g.cm-3 were manufactured at different ratios of bagasse/wheat straw, i.e., 100%/0%, 93.75%/6.25%, 87.5%/12.5%, 75%/25%, 50%/50%, 25%/75% and 0%/100% using two particle/ gypsum composite ratios, i.e., 1/2.75 and 1/3.25 by weight. Thickness swelling (TS), water absorption (WA), modulus of rupture (MOR), modulus of elasticity (MOE) and internal bond strength (IB) properties of the boards were evaluated and a statistical analysis was performed in order to examine the possible feasibility of these agricultural residues for use in commercial GBPB manufacturing. We determined that WA of panels decreases as the amount of straw increases to 100% and the LR/G (wood/gypsum) ratio decreases to 1/3.25, whereas the TS of panels decreases as the proportion of straw decreases to 0% and the LR/G ratio increases to 1/2.75. The experimental results also show that the MOR and MOE of panels containing 0%, 6.25% and 12.5% wheat straw with a LR/G ratio of 1/2.75 were higher than those of panels made from 25%-100% wheat straw with a LR/G ratio of 1/2.75, as well as those from all other percentages of straw with a LR/G ratio of 1/3.25. On the other hand, the IB of panels containing more than 12.5% straw with LR/G ratios of 1/2.75 and 1/3.25 were lower than those of panels made from 0-12.5% straw also with both LR/G ratios. Panels consisting of 0%, 6.25% and 12.5% wheat straw with LR/G ratios of 1/2.75 and 1/3.25 met the minimum EN standard requirements of mechanical properties for general purposes. All of the panels containing 0-100% wheat straw with a LR/ G ratio of 1/2.75 or 1/3.25 met the required level of TS for 24-h immersion.  相似文献

This experiment was conducted to investigate and compare the efficacy of different feed additives on performance, tibial dyschondroplasia (TD) incidence and tibia characteristics of male broilers fed low-calcium diets. A completely randomized design, with six treatments and five replicates of five chicks per each was used. Experimental treatments were: (i) Basal diet containing recommended level of calcium (0.9%) as control treatment (Ctrl), (ii) low-calcium (0.67%) diet without any additive (LC), (iii) low-calcium diet + probiotic (2 g/kg diet), (iv) low-calcium diet + prebiotic (2 g/kg diet), (v) low-calcium diet + synbiotic [mix of probiotic and prebiotic (each 2 g/kg diet)], (vi) low-calcium diet + organic acid (1.5 g/kg diet). Birds were reared in an open-sided house system under natural tropical condition until 21 days of age. Feeding with low-calcium diet negatively influenced broiler performance (body weight, body weight gain and feed conversion ratio) and tibia characteristics, whereas dietary inclusion of all feed additives had beneficial effects on above-mentioned parameters and helped the birds to overcome problems related to low-calcium diets. Background: Antioxidants such as α-tocopherol (vitamin E) and β-carotene (vitamin A) play an important role in protective effect of repeated brief periods of ischemia, namely ischemic preconditioning (IPC). Values of these antioxidants were investigated and compared after induction of ischemia reperfusion (IR) and kidney IPC in both male and female rats. Methods: Forty eight Wistar rats were divided randomly into six groups of 8: groups A and B (male and female controls, respectively), group C (male IR or IR cases), group D (female IR cases) and groups E and F (male and female IPC cases, respectively). In groups C and D, ischemia was induced by clamping of left renal arteries for 45 min. In groups E and F, rats underwent four cycles of 4 min of arterial clamping and 11 min of de-clamping before final 45 min ischemia induction. Afterward, serum was collected to assess the blood urea nitrogen, creatinine and vitamins E and A values. Renal tissues were obtained for histological assessments. Results: α-tocopherol levels in male and female rats showed a significant increase in IPC compared with IR group (P<0.01) and also in female IPC compared with male IPC group. β-carotene levels had no significant variations. Histological evaluation showed that IR-induced renal injuries were less in female rats. Also, protective effects of IPC were more in female rats (P<0.01). Conclusions: Renal IPC reduced damages in both male and female rats, but tissue injuries in females were decreased much more along with the increase of endogenous vitamin E. A study was conducted to evaluate the effect s of silver nanoparticles for ameliorating negative effects of salinity on germination and growth of Fenugreek seeds. In order to investigate salinity stress on Fenugreek germination indices, an experiment was carried out in Iran from October 2014 to November 2014 at Ferdowsi University of Mashhad, in the biotechnology Laboratory, to create salinity stress, sodium chloride (NaCl) at the levels of 0 (as control), 5, 10, 15 and 20 dS/m and Five levels of silver nanoparticles (0, 10, 20, 30 and 40 μg mL–1) on Fenugreek seed (Trigonella foenum-graecum); were tested Germination characteristics such as total germination (GT), Germination Speed Index (GSI), Shoot and Root of seedling long, Fresh Mass and Dry Mass were measured. Results showed the great effects of silver nanoparticles to improve salinity stress on Fenugreek seed germination. Results showed a significant reduction in germination percent and seedling growth due to the salinity stress while significantly increased with silicon nano-particles application. In without silver nanoparticle low level of salinity (0 dS/m) increased seed germination percentage, while the high levels (5, 10, 15 and 20 dS/m) inhibited the seed germination significantly. The results showed that the effect of AgNPs was significant on germination percentage in P ≤ 0.05. Overall, application of AgNPs was beneficial in improving salinity tolerance in the Fenugreek seedling and its application may stimulate the differences defense mechanisms of plants against salt toxicity.  相似文献
