Rice blast disease caused by the fungus Magnaporthe oryzae is one of the most devastating diseases causing huge losses worldwide. In the present study, major blast resistance genes were investigated in landraces originating from northeastern India. Based on phenotypic evaluation, 288 landraces were classified into three distinct groups: resistant (75), moderately resistant (127) and susceptible (86). The genetic frequencies of the 18 major blast resistance genes were between 6.2% and 27.4%, with only two genotypes possessing a maximum of nine blast resistance genes. The cluster and population structure analysis grouped the landraces into two groups. Through principal coordinate analysis, the scatter plots partitioned the resistant and moderately resistant landraces into different groups. Analysis of molecular variance showed maximum (96%) diversity within populations and least (4%) diversity between populations. Association analysis identified six markers, CRG4_2, RM72, tk59-2, pi21_79-3, RM1233 and RM6648, that are significantly associated with blast disease and explained a phenotypic variance of 1.1–6.5%. The associated genes could be used in marker-assisted rice breeding programmes for gene pyramiding to develop rice varietal resistance against blast disease. The present study represents a valuable blast resistance genetic resource that could be used for identification of new R genes, donors for blast resistance, and genomic studies.  相似文献

A water soluble storage albumin from Inca peanut (IPA) accounted for approximately 25% (w/w) of defatted seed flour weight, representing 31% of the total seed protein. IPA is a 3S storage protein composed of two glycosylated polypeptides, with estimated molecular weights (MW) of 32800 and 34800 Da, respectively. IPA has an estimated sugar content of 4.8% +/- 0.92% (n = 6). IPA is a basic protein (pI of approximately 9.4) and contains all of the essential amino acids in adequate amounts when compared to the FAO/WHO recommended pattern for a human adult. The tryptophan content of IPA is unusually high (44 mg/g of protein), whereas the phenylalanine content is low (9 mg/g of protein). IPA is a highly digestible protein in vitro.  相似文献

Wool fabric was treated with liquid ammonia at -40 °C for 30 and 60 s prior to the application of polypyrrole (PPy). The polymer was deposited on wool fiber using the chemical oxidation method with 0.02 and 0.05 mol/l (Py) monomer concentration and FeCl3 as a catalyst. Functional groups of wool samples were analyzed using FT-IR, and surface morphology was investigated using SEM micrographs. Properties such as water absorbency, surface resistivity, abrasion resistance, weight add-on, and air permeability of coated specimens were explored. The FT-IR outcomes revealed the liquid ammonia pre-treatment changed the amount of amide I (NH), cystic acid, cystic monoxide, and dioxide content of the fiber. SEM micrographs revealed the descaling of wool surface after pre-treatment and smooth coating of polymer. Pre-treatment of wool in liquid ammonia improved absorbency of wool fabric with respect to the treatment duration. The surface resistivity of wool fabric decreased with the increase of monomer concentration and pre-treatment duration. The results of abrasion resistance confirmed that the pre-treated fabric exhibited lower loss of polymer after 200 cycles of abrasion. The weight of the fabric was increased and air permeability decreased when the monomer concentration and liquid ammonia pre-treatment duration was increased.  相似文献

The allopolyploidization event that created cultivated oilseed rape Brassica napus L, followed by intense breeding, reduced its genetic diversity. Resynthesized (RS) B. napus L. obtained by interspecific hybridization between genotypes of B. rapa L. and B. oleracea L. can be a valuable source for broadening genetic diversity in cultivated oilseed rape. In this study, we determined the extent of DNA polymorphism among natural accessions of oilseed rape, resynthesized B. napus, their parental species and double-low quality semi-RS lines carrying the Rfo gene. Using 10 selected primer combinations, 522 polymorphic AFLP markers were scored in the complete set of 100 Brassica sp. To detect relationships between these genotypes, a cluster analysis was performed using the Jaccard’s distance. Resynthesized allopolyploids clustered directly between their diploid parents. Cultivated accessions of oilseed rape created a compact group away from resynthesized allopolyploids as well as semi-RS lines. The natural oilseed rape group, which consists of 49 cultivars and breeding lines of oilseed rape, is characterized by lower genetic diversity than the group of 33 accessions of resynthesized oilseed rape, and the analysis showed that the double-low quality semi-RS lines represent a specific genetic variation of B. napus. The de novo resynthesized B. napus lines and the semi-RS lines of double-low quality generated from them, provide a significant opportunity for enrichment the gene pool of oilseed rape.  相似文献

The combined efficacy of spinosad and chlorpyrifos-methyl was determined against four storage psocid pests belonging to genus Liposcelis. This research was undertaken because of the increasing importance of these psocids in stored grain and the problem of finding grain protectants to control resistant strains. Firstly, mortality and reproduction were determined for adults exposed to wheat freshly treated with either spinosad (0.5 and 1 mg kg(-1)) or chlorpyrifos-methyl (2.5, 5 and 10 mg kg(-1)) or combinations of spinosad and chlorpyrifos-methyl at 30 degrees C and 70% RH. There were significant effects of application rate of spinosad and chlorpyrifos-methyl, both individually and in combination, on adult mortality and progeny reduction of all four psocids. Liposcelis bostrychophila Badonnel and L. decolor (Pearman) responded similarly, with incomplete control of adults and progeny at both doses of spinosad but complete control in all chlorpyrifos-methyl and combined treatments. In L. entomophila (Enderlein) and L. paeta Pearman, however, complete control of adults and progeny was only achieved in the combined treatments, with the exception of spinosad 0.5 mg kg(-1) plus chlorpyrifos-methyl 2.5 mg kg(-1) against L. entomophila. Next, combinations of spinosad (0.5 and 1 mg kg(-1)) and chlorpyrifos-methyl (2.5, 5 and 10 mg kg(-1)) in bioassays after 0, 1.5 and 3 months storage of treated wheat were evaluated. The best treatment was 1 mg kg(-1) of spinosad plus 10 mg kg(-1) of chlorpyrifos-methyl, providing up to 3 months of protection against infestations of all four Liposcelis spp. on wheat.  相似文献

Malignant catarrhal fever (MCF) is a fatal herpesvirus infection of domestic and wild ruminants, with a short and dramatic clinical course characterized primarily by high fever, severe depression, swollen lymph nodes, salivation, diarrhea, dermatitis, neurological disorders, and ocular lesions often leading to blindness. In the present study, fatal clinical cases of sheep associated malignant catarrhal fever (SA-MCF) were identified in cattle in the state of Karnataka. These cases were initially presented with symptoms of diarrhea, respiratory distress, conjunctivitis, and nasal discharges. Laboratory diagnosis confirmed the detection of ovine herpesvirus-2 (OvHV-2) genome in the peripheral blood samples of two ailing animals. The blood samples collected subsequently from sheep of the neighboring areas also showed presence of OvHV-2 genome indicating a nidus of infection in the region. The positive test results were further confirmed by nucleotide sequencing of the OIE approved portion of tegument gene as well as complete ORF8 region of the OvHV-2 genome. Phylogenetic analysis based on the sequence of the latter region indicated close genetic relationship with other OvHV-2 reported elsewhere in the world.  相似文献

Indaziflam is a preemergent herbicide widely used for the control of weeds in pecan (Carya illinoinensis) orchards in the southwestern region of the United States. Given the paucity of data regarding the effect of indaziflam on the biochemical properties of soils supporting pecan production, this study was conducted to evaluate the effects of different application rates of indaziflam on soil microbial activity, diversity, and biochemical processes related to nitrogen (N) cycling. During two consecutive growing seasons (2015 and 2016), soil samples were obtained from experimental mesocosms consisting of soil-filled pots where pecan saplings were grown and treated with indaziflam applied at two different rates (25 and 50 g active ingredient (ai) ha-1, with the higher rate being slightly lower than the recommended field application rate of 73.1 g ai ha-1). Soil samples were collected approximately one week before and one week after herbicide application for determination of soil microbial biomass and diversity, N mineralization, and β-glucosaminidase activity. Soil samples collected from the control mesocosms without herbicide application were treated in the laboratory with two rates of indaziflam (75 and 150 g ai ha-1) to determine the immediate effect on microbial activity. No significant effect of herbicide treatment on soil respiration and microbial biomass was detected. The results showed a slight to moderate decrease in microbial diversity (7% in 2015 and 44% in 2016). However, decreased β-glucosaminidase activity with herbicide treatment was observed in soils from the mesocosms (33%) and soils treated with indaziflam in the laboratory (45%). The mineral N pool was generally dominated by ammonium after indaziflam application, which was consistent with the drastic decrease (75%) in nitrification activity measured in the laboratory experiment. The results of this study indicate that indaziflam, even when applied at higher than recommended rates, has limited effects on soil microbial activity, but may affect N cycling processes.  相似文献

Regional mapping of soils helped to identify and map 24 soil-geomorphic units on the Deoha/Ganga–Ghaghara Interfluve in the Upper Gangetic Plains (Himalayan Foreland Basin). On the basis of luminescence ages soil-geomorphic units can be grouped into five members of a Morphostratigraphic Sequence with ages of ≤ 1.7 ka, 1.8–3.6 ka, 4.6–6.4 ka, 6.8–10 ka and > 10 ka. Except for two units with sandy parent material, all the members with loamy parent materials show systematic increase in the degree of soil development from Member QGMS-I to V. Major pedogenic processes are salinization, alkalinization, illuviation, calcrete development and gleying, and degradation of some micromorphological features is observed in the oldest soils.Regional mapping and dating of soils show that the Interfluve between the Deoha/Ganga–Ghaghara rivers is bounded by longitudinal faults, along the bounding rivers. The major longitudinal faults trend N–S or NNE–SSW in the northern region; turn N–S in the central region and take easterly to SEE direction in the south, giving the Interfluve a curvilinear shape. Strike of a set of six transverse extensional normal faults changes from approximately E–W to NEE–SWW in the southernmost region of the Interfluve. Downthrown sides are to the south and to the east in northern region and western region, respectively.Due to the activity of different segments of various transverse faults during a probably dry sub-humid to semi-arid climatic period of 10–5 ka, terminal fans were deposited on the downthrown blocks. Thus, role of extensional tectonics in an overall compressional regime is significant. Also, tilting of smaller blocks leading to the shifting away of large rivers seems to be an additional control on the distribution of soils and sedimentation on the Interfluve.  相似文献

Tomato root rot caused by Rhizoctonia solani is a major soilborne disease resulting in significant yield loss. The culture filtrates of six isolates of Trichoderma/Hypocrea species were evaluated for in vitro production of hydrolytic enzymes. Results demonstrated that all the six isolates were able to produce chitinase, β-1, 3 glucanase and protease in the range of 76–235 μmol GlcNAc min-1 mg-1 protein, 31.90–37.72 nmol glucose min-1 mg-1 proteins and 63.05–86.22 μmol min-1 mg-1 proteins, respectively. Trichoderma/Hypocrea-based formulation(s) were prepared with chitin (1% v:v) and CMC (0.5% w:v) for root rot management in a greenhouse. Root dip application with bioformulation(s) resulted in a significant reduction of the root rot index. In addition, bioformulations increased plant growth attributing traits significantly relative to untreated control. Accumulation of total phenols, peroxidase, polyphenoloxidase and phenylalanine ammonia lyase increased in chitin-supplemented Trichoderma/Hypocrea formulation-treated plants challenged with R. solani. The results suggest that chitin-fortified bioformulation(s) could be an effective system to control root rot of tomato in an eco-compatible manner.  相似文献
