Fen peatlands are specific wetland ecosystems containing high soil organic carbon (SOC). There is a general lack of knowledge about the microbial communities that abound in these systems. We examined the microbial activity and community structure in two fen soils differing in SOC content sampled from the Ljubljana Marsh under different seasonal conditions. Substrate-induced respiration and dehydrogenase activity were used as indicators of total microbial activity. Both methods indicated higher microbial activities in the fen soil with the higher SOC content on all dates of sampling. To determine whether the differences in microbial activity were associated with differences in the microbial community structures, terminal restriction fragment length polymorphism (T-RFLP) of bacterial 16S rRNA genes was performed. Comparison of the T-RFLP profiles revealed very similar community structures in both fens and in the two seasonal extremes investigated. This suggested a stable community structure in the two fens, which is not affected by the SOC content or seasonal variation. In addition, a bacterial 16S ribosomal RNA gene based clone library was prepared from the fen soil with the higher SOC content. Out of 114 clones analysed, approximately 53% belonged to the Proteobacteria, 23% to the Acidobacteria, 21% to a variety of other taxa, and less than 3% were affiliated with the Firmicutes.  相似文献

Determination of the stresses operating on a forest ecosystem demands the use of several bioindication methods. Air quality indicators were assessed from an inventory of forest decline based on the assessment of tree crowns and epiphytic lichens. Photosynthetic pigments, ascorbic acid and major macronutrients were studied in Norway spruce needles as indicators of physiological and biochemical stress. Analyses were carried out on selected forest plots in polluted areas (Zasavje district, vicinity of thermal power plants) and predominantly unpolluted areas (Triglav National Park, Julian Alps, Slovenia). For some bioindication methods, there was good agreement with measured air quality and climatological parameters. The best agreement was found between total foliar sulphur in needles and epiphytic lichens, especially in more polluted areas. Agreement with forest decline inventories and analyses of some needle stress physiological/biochemical parameters was less convincing. The strength of agreement was further decreased by soil characteristics and climatic parameters, influenced also by biotic parameters and forest stand history and management. It was concluded that there is no simple bioindication method available to evaluate the vitality of a forest.  相似文献

The microbial activity and bacterial community structure were investigated in two types of peat soil in a temperate marsh. The first, a drained grassland fen soil, has a neutral pH with partially degraded peat in the upper oxic soil horizons (16% soil organic carbon). The second, a bog soil, was sampled in a swampy forest and has a very high soil organic carbon content (45%), a low pH (4.5), and has occasional anoxic conditions in the upper soil horizons due to the high water table level. The microbial activity in the two soils was measured as the basal and substrate-induced respiration (SIR). Unexpectedly, the SIR (μl CO2 g−1 dry soil) was higher in the bog than in the fen soil, but lower when CO2 production was expressed per volume of soil. This may be explained by the notable difference in the bulk densities of the two soils. The bacterial communities were assessed by terminal restriction fragment length polymorphism (T-RFLP) profiling of 16S rRNA genes and indicated differences between the two soils. The differences were determined by the soil characteristics rather than the season in which the soil was sampled. The 16S rRNA gene libraries, constructed from the two soils, revealed high proportions of sequences assigned to the Acidobacteria phylum. Each library contained a distinct set of phylogenetic subgroups of this important group of bacteria.  相似文献

The antiozonant ethylene diurea is proven to prevent growth reductions in forest trees induced by ozone. The community of mycorrhizal fungi could be useful indicator of environmental stress. In this study, response of mycorrhizal fungi and fine roots to a 4-year exposure to ambient ozone and treatment with antiozonant was investigated in ozone-sensitive poplar clone under field conditions. The community of ectomycorrhizal fungi and root length colonization with ectomycorrhizal, arbuscular mycorrhizal fungi, and root endophytic fungi was analyzed in antiozonant-treated poplar plants and in poplar plants irrigated with water. In general, plants protected by antiozonant showed higher total number of fine roots, number of ectomycorrhizal types, Shannon–Weaver diversity index, and Species richness index compared to the plants treated with water. The ectomycorrhizal community shifted from contact exploration type in the trees irrigated with water to short-distance exploration type in ethylene diurea-treated trees. Ozone protectant may beneficially affect the belowground community of mycorrhizal fungi colonizing roots of ozone-sensitive poplar clone.  相似文献

Saproxylic organisms are among the most threatened species in Europe and constitute a major conservation problem because they depend on the most important forestry product - dead wood. Diversity of fungal and bryophyte communities occurring on dead beech trees was analyzed in five European countries (Slovenia, Hungary, The Netherlands, Belgium and Denmark) considering tree level species richness (TLSR), country level species richness (CLSR), frequency distributions of species, occurrence of threatened species and relations between TLSR and decay stage, tree size and countries. Altogether 1009 trees were inventoried in 19 beech dominated forest reserves.The number of fungi in the full dataset was approximately three times larger (456 versus 161 species) and the proportion of low frequent species was higher than among bryophytes. The species richness of bryophytes and fungi was significantly different among countries considering both TLSR and CLSR. In addition the diversity patterns deviated considerably between the two groups of organisms. Slovenian sites appeared to be biodiversity hotspots for bryophytes characterized by high TLSR and CLSR and a high fraction of threatened species. Hungarian sites had somewhat lower bryophyte diversity, while the Atlantic region had deteriorated assemblages. Fungal species richness was very high in Denmark, but the Hungarian and Slovenian sites were richer in threatened and low frequency species. Tree size was better able to explain variation in TLSR in both organism groups than decay stage. TLSR was found to vary significantly between countries but the difference was most considerable in the case of bryophytes.The diversity patterns of both organism groups along the investigated geographical gradient appear to be influenced by both climatic and management related factors (forest history, dead wood availability and continuity, habitat fragmentation). There is no doubt that an increase in the abundance of dead wood in European beech forests will benefit diversity of saproxylic fungi and bryophytes, especially if a continuous presence of large diameter logs are secured within individual stands.  相似文献
