This prospective study evaluated transdermal amlodipine for the control of hypertension in six cats. Cats were treated with oral amlodipine until blood pressures decreased to <180 mm Hg. They were maintained on this dose for 7 days and then administered identical doses of transdermal amlodipine for 7 days. Oral amlodipine decreased pressure by a median of 73 mm Hg, which subsequently increased by 20 mm Hg after 7 days of transdermal amlodipine. Plasma concentrations of amlodipine were measured after oral and transdermal dosing. Additional studies are needed to determine dosing, pharmacokinetics, and efficacy.  相似文献

Metastasis is a defining feature of malignant tumors and is the most common cause of cancer-related death, yet the genetics of metastasis are poorly understood. We used exome capture coupled with massively parallel sequencing to search for metastasis-related mutations in highly metastatic uveal melanomas of the eye. Inactivating somatic mutations were identified in the gene encoding BRCA1-associated protein 1 (BAP1) on chromosome 3p21.1 in 26 of 31 (84%) metastasizing tumors, including 15 mutations causing premature protein termination and 5 affecting its ubiquitin carboxyl-terminal hydrolase domain. One tumor harbored a frameshift mutation that was germline in origin, thus representing a susceptibility allele. These findings implicate loss of BAP1 in uveal melanoma metastasis and suggest that the BAP1 pathway may be a valuable therapeutic target.  相似文献

A total of 4,681 accessions of Hordeum vulgare landrace material from Ethiopia, East Mediterranean, Near East, Nepal and China were sown in the field and subjected to the natural powdery mildew epidemic in Denmark. Apparently resistant accessions were selected. Selfed progeny from them were retested and reselected in subsequent years at four locations in Denmark. Finally, 16 promising donors of resistance were retained. They were characterized in the field and tested in the seedling stage for reaction to up to 72 different isolates of the powdery mildew fungus. The absence of the corresponding virulences in the Danish airborne powdery mildew population was ascertained in five years. The resistances in the 16 donors are apparently mutually different and from known sources of powdery mildew resistance in barley. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Summary Partial resistance to powdery mildew in spring barley was evaluated in three plot types: large isolation plots, in 1.4 m2 plots in chessboard design with guard plots of spring wheat and in single rows. Percentage leaf area covered by powdery mildew was scored four to six times during the season and partial resistance was characterized by the area under the disease progress curve. Varietal differences were revealed in al three plot designs, differences between the most resistant and susceptible genotypes being of a factor five. Differences between varieties decreased with decreasing plot size. The relationship between single scores of amount of powdery mildew on the upper four leaves and the area under the disease progress curve was high in all plot designs during the first two to three weeks after heading, allowing selection for the trait by one or two scorings. Differential ranking of varieties between different plot designs was observed, and is assumed to be due to increasing plot interference with reduced plot size and reduced distance between plots. A reliable selection for partial resistance could be made in large isolation plots and in 1.4 m2 plots, but hardly in single rows.  相似文献

Summary A population of 198 chromosome-doubled haploid lines of spring barley was scored for segregation in locus ml-o (powdery mildew reaction) on chromosome 4 and in the linked loci s (rachilla hair length) and ddt (reaction to the insecticide DDT) on chromosome 7. They were also tested in a disease-free field trial for the agronomic traits: grain yield, thousand grain weight, lodging, and necrotic leaf spotting. The three mutagen-induced resistance genes ml-o5, ml-o6 (from Carlsberg II) and ml-10 (from Foma) showed no detectable differences with respect to effects on agronomic traits. They all conferred a four per cent reduction in grain yield caused mainly by lower thousand grain weight, and an increase in necrotic leaf spotting. The two original mutants of Carlsberg II had additional mutant genes affecting agronomic traits. Lines with gene S (long hair) had on average a three per cent higher thousand grain weight than those with s. The alleles in locus ddt showed no association with the agronomic traits. It is concluded i) that the associations between the three ml-o alleles and agronomic traits are caused by pleiotropy, ii) that ml-o resistant, high-yielding lines may be selected, and iii) that the association between gene s and thousand grain weight may be due to genetic linkage.Abbreviations DH-lines chromosome-doubled haploid lines  相似文献

Summary Mlo resistance to barley powdery mildew is a relatively new kind of resistance. It was originally described in a powdery mildew resistant barley mutant in 1942 and has been mutagen-induced repeatedly since then. About 1970 it was also recognized in barley landraces collected in Ethiopia in the 1930s. It is unique in that 1) Mlo resistance does not conform to the gene-for-gene system; 2)mlo genes originating from different mutational events map as non-complementing recessive alleles in one locus; 3) all alleles confer the same phenotype, though with small quantitative differences; 4) it is effective against all isolates of the pathogen; and 5) the resistance is caused by rapid formation of large cell wall appositions at the encounter sites preventing penetration by the fungus. Powdery mildew isolates with elevated Mlo aggressiveness have been produced on barley in the laboratory, but have not been found in nature. Mlo resistance is considered very durable. The exploitation of Mlo resistance has been hampered by pleiotropic effects of themlo genes, vix. necrotic leaf spotting and reduced grain yield, but they have been overcome by recent breeding work. During the 1980s Mlo-resistant spring barley varieties have become cultivated extensively in several European countries, in 1990 on about 700,000 ha.  相似文献

Three-dimensional (3D) movement of neuroprosthetic devices can be controlled by the activity of cortical neurons when appropriate algorithms are used to decode intended movement in real time. Previous studies assumed that neurons maintain fixed tuning properties, and the studies used subjects who were unaware of the movements predicted by their recorded units. In this study, subjects had real-time visual feedback of their brain-controlled trajectories. Cell tuning properties changed when used for brain-controlled movements. By using control algorithms that track these changes, subjects made long sequences of 3D movements using far fewer cortical units than expected. Daily practice improved movement accuracy and the directional tuning of these units.  相似文献

A 17-ha area in the central California Sierra was harvested in 1979 to provide three shelterwoods with residual basal areas of 10, 15 and 20 m2/ha. An adjacent uncut stand of 75 m2/ha was used as a control. During the summer of 1980, seedling performance was assessed along with five microenvironmental characteristics. Maximum air temperature was similar among shelterwoods; minimum air temperature varied by about 3°C. Daily potential evaporation in all shelterwoods was similar and doubledfrom June to July and remained constant through September. Evaporation in the control was half of the shelterwoods. Subsurface soil temperature reached a maximum in August of 24°C in the least-dense shelterwood and 16°C in the control. Subsurface soil moisture was 35% for all treatments in May, but in September was 18% in the least-dense shelterwood and 12% in the control. Daily incident light in June ranged from 76% to 54% of maximum in the least-dense and most-dense shelterwood respectively, and 12% of maximum in the control.Survival of Douglas-fir was greater than 93% in all treatments. However, survival of white fir ranged from 63% to 85% as the overstory density increased; these differences were not significant. The period of rapid white fir mortality occurred later in the summer with increasing canopy cover. Height growth of both species was about 2.3 cm in the control to 3.1 cm under the least-dense shelterwood; however, treatment differences were significant only for white fir, Stem diameter growth of about 1.3 mm in treatments 10 and 15 was significantly greater than the growth of 0.3 mm in treatments 20 and the control. In Douglas-fir shoot: root ratios and biomass tended to increase with decreasing shelterwood. In white fir, the reverse trend was observed for shoot:root ratio but biomass production was similar in each shelterwood. Seedling sshowed the least overall growth in the uncut stand where plant moisture stress reached -19 bar (-1.9 MPa) in Douglas-fir and -22 bar (-2.2 MPa) in white fir.Results indicate that a 10 m2/ha shelterwood provides conditions for successful 1st-year survival and growth of container-grown Douglas-fir, but a 15 m2/ha density is preferable for bare-root white fir. Shoot growth initiation does not seem to be delayed under shelterwoods of up to 20 m2/ha.  相似文献

Total hip endoprotheses are a good possibility for treatment degenerative wear and pathologic damage of the hip joint in man as well as in dogs. However, aseptic loosening of the protheses, especially in the area of the shaft, is still a problem in conventional total hip endoprotheses. The purpose of the present study was to use the finite-element-analysis (FEA), to enhance endoprothetic design and to prevent loosening of protheses. In order to simulate the femur of the dog for numerical analysis, a material law for the compacta in the femur of the dog was developed. The elastic properties of the compacta were experimentally determined by using compression tests of bone samples of euthanised dogs. The results show constant denseness and a constant axial elastic modul in the compacta.  相似文献
