Potato common scab caused by the actinobacterium Streptomyces scabiei is characterized by the formation of corky lesions on tubers that reduce their marketability. Management of common scab is very complex and often ineffective under various environmental conditions. Using potato varieties that are more resistant to common scab remains one of the most efficient strategies to control this disease. However, very little is known about the factors associated with resistance to common scab. Somaclone RB9 regenerated from thaxtomin A-habituated potato Russet Burbank calli produced tubers more resistant to common scab than the original variety. Comparison of the RB9 tuber proteome with that of Russet Burbank using label-free quantitative proteomic analysis revealed changes in the accumulation of defence-related proteins from the patatin and lipoxygenase (LOX) families, which are involved in the metabolism of lipids, and of two miraculins of the Kunitz-type protease inhibitors family. The implication of LOX during common scab infection was studied using synchronized minitubers developed from leaf-bud cuttings. S. scabiei infection stimulated the accumulation of LOX in both Russet Burbank and RB9 minitubers, but this accumulation was intensified in RB9 minitubers. Infection also increased LOX activity in Russet Burbank and RB9 minitubers. However, LOX activity measured in noninfected RB9 minitubers was similar to that of infected Russet Burbank minitubers, indicating endogenous activation of LOX activity in RB9 minitubers. We discuss how increased LOX abundance and activity in the somaclone RB9 may contribute to improving tuber defence against common scab.  相似文献

Abstract. Vineyards in Champagne, France are generally situated on slopes where the soils are subject to erosion. Therefore it is important to find a soil‐surface management practice that protects the soil against water erosion. We assessed the potential of mulches or grass covers to stabilize soil aggregates in a calcareous sandy loam from a vineyard in Champagne after 9 years under different management systems. Four different treatments were studied: (i) a bluegrass (Poa pratensis) surface cover between the vine rows (GC) with bare soil under the vines (R); two organic mulches of (ii) coniferous (CB) or (iii) poplar (PB) bark that covered the entire soil surface, and (iv) bare soil between the rows as a control. The bark amendments were applied every 3 years at rates of 61 and 67 t ha?1 for the PB and CB treatments, respectively. The kinetics of soil disaggregation in water fitted a power law (A=K t?D), in which K was the fraction of water‐stable >200 μm aggregates remaining after 1 hour of wet‐sieving. In the 0–5 cm layer, aggregate stability was greater for GC (K=21.7), CB (K=15.2) and PB (K=13.6) than for the control (K=10.5) and R (K=11.8). In the 0–20 cm layer, CB also stabilized soil aggregates (K=14.0–15.0); but PB did not. Structural stability was more strongly related to total organic carbon (R2=0.64, P <0.001) than to microbial biomass carbon (R2=0.54, P<0.001). A bluegrass cover enhanced structural stability in the 0–5 cm and 0–20 cm layers (K=14.2), probably because of intense root development and rhizodeposition enhancing microbially produced metabolites, such as carbohydrates. Establishing grass cover or applying bark mulch are effective agricultural practices that improve soil aggregate stability and thus should reduce soil erosion. The vegetative growth of the vines was greater on the soils amended with bark mulches and less on the grass covered soils compared with the control soil; however, no difference in wine quality was observed among the different treatments.  相似文献

The concentration and partitioning of metals in vegetatedand unvegetated surface sediments were compared among fourwetlands designed for wastewater treatment. In wetlands witha significant depth gradient, the highest concentrations ofmetals were found in deeper unvegetated sediments. When theeffect of depth was removed, surface sediments with andwithout cattails (Typha latifolia L.) had similarconcentrations of total metals (Fe, Mn, Zn and Cu). However,vegetated sediments of all four wetlands had higher organiccontent (p < 0.01). Furthermore, the redox potential wassignificantly lower (p < 0.01) in vegetated sediments for 3out of 4 wetlands. Vegetated and unvegetated sediments werealso compared based on geochemical fractions (reactive,silicate, persistent organic and pyritic). Generally, halfof the total metal concentration in sediments was associatedwith the reactive fraction (metal oxides, monosulfides andadsorbed onto organic matter). Zn was an exception as halfof the total Zn was associated with the persistent organicfraction. The partitioning of metals in shallow vegetatedsediments differed from deep unvegetated sediments at theMonahan and Falconbridge wetlands. In contrast, there was nosignificant difference in the partitioning of metals insurface sediments with or without cattails at the Panel andRiverwalk wetlands. This study suggests that the effect ofcattails on the bulk concentration and partitioning ofmetals may be minimal in surface-flow constructed wetlands.  相似文献

Blends of vegetable proteins were prepared to improve the nutritive value of these proteins. Soybean flour, rapeseed protein concentrate, whole wheat flour, soybean 2S+11S extract, wheat albumin-globulin + glutenin (AG+G) functions, and some of their blends were compared to casein for protein efficiency ratio (PER) and apparent digestibility coefficient (ADC). Autoclaving (1 h) soybean proteins was also studied. Casein and rapeseed protein concentrate gave the highest weight gains and PER. Other protein sources gave lower values for both PER (P<0.05) and weight gain. The digestibility of all vegetable proteins was lower (P<0.05) than that of casein. PER of soybean 2S+11S extract was significantly lower (P<0.05) than that of soybean flour. Autoclaving significantly (P<0.05) improved the PER of both soybean flour and 2S+11S extract. The ADC of autoclaved 2S+11S extract was similar (P<0.05) to that of autoclaved soybean flour and significantly higher (P<0.05) than that of 2S+11S extract. Soybean flour had the lowest (P<0.05) ADC. Heat treatment destroyed antinutritional factors, probably trypsin inhibitors and/or haemagglutinins of soybean. This was accompanied by improvement in the nutritional value of protein. The four blends were chosen on the basis of amino acid composition, chromatography of proteolyzates and nutritive value of each fraction. The PER of wheat albumin-globulin and glutenin (AG+G) blend was similar (P<0.05) to that of wheat flour and lower (P<0.05) than that of all other blends used. Wheat AG+G+rapeseed protein concentrate and wheat AG+G+soybean flour blends gave the highest (P<0.05) PER. Wheat AG+G improved the PER of 2S+11S extract and of soybean flour but decreased the PER of rapeseed protein concentrate. Wheat AG+G and wheat AG+G+rapeseed protein concentrate blends gave an ADC significantly higher (P<0.05) than that of wheat AG+G blend containing soybean flour of 2S+11S extract. However, blending wheat AG+G with either 2S+11S extract of soybean flour improved ADC.  相似文献

Many bird species respond to forestry, even at moderate intensities. In New Brunswick, Canada, the Brown Creeper exhibits a negative, threshold response to harvesting intensity. This study aimed to determine whether (a) the threshold found in Brown Creeper occurrence is lower than eventual thresholds in its nesting requirements, and whether (b) the conservation of this species could be achieved through moderate-intensity harvest systems. Creepers are particularly sensitive to forestry because they nest on snags with peeling bark and they mainly forage on large-diameter trees. In northern hardwood stands, we compared habitat structure at local- (r = 80 m) and neighbourhood-scales (r = 250 m) around nest sites and sites not used by creepers. Over two years, we found 76 nests, 66 of which were paired with unused sites for comparison. At the local scale, densities of trees 30 cm dbh and snags 10 cm dbh, and the probability of presence of potential nest sites were significantly higher near nests than at sites where no creepers were detected. At the neighbourhood scale, the area of untreated mature forest was significantly higher around nests. Variance decomposition indicated that habitat variables at the local scale accounted for the majority of explained variation in nest site selection. We also found significant thresholds in the densities of large trees (127/ha) and snags (56/ha), and in the area of mature forest (10.4 ha). The conservation of breeding populations of Brown Creepers may thus require densities of large trees nearly twice as high as those associated with its probability of presence. Such a target seems to be incompatible even with moderate-intensity harvesting.  相似文献

Soil frost heaving is the result of the formation of ice lenses in the soil caused by a segregation of the soil water. Ice lenses are growing from below and pushed upward. Seedlings heave when they are pushed out of the ground by the ice sheet formed at the surface of the soil. Frost heaving may greatly reduce growth and survival of forest tree seedlings particularly in regions where freezing and thawing are accompanied by high soil moisture. Resistance to frost heaving increases with size of seedling as the ability of a seedling to anchor itself increases. A few methods, such as fertilizing, choice of planting spots, sowing or planting at the proper time, shading, and use of mulches, appear to be effective in controlling frost heaving. Shade from natural plant cover can greatly reduce frost heaving.  相似文献

The objective of this work was to study the effect of the extraction of phenols by methanol/acetone/water and proteolysis (pepsin 1 hour; trypsin 2 hours) on the nutritional characteristics of unheated rapeseed protein as measured by weight gain, protein intake, net protein ratio, apparent digestibility and absorbed protein. The effect of proteolysis of the methanol/acetone/water extracted rapeseed protein, and the effect of mixing the methanol/acetone/water extract back with the extracted rapeseed protein was also studied. Extraction of phenolic compounds from rapeseed flour significantly improved weight gain, protein intake, net protein ratio and absorbed protein value. However, the mixing of phenolic extract with the extracted rapeseed protein did not appear to have a significant effect.Enzymatic hydrolysis (1 hour with pepsin and 2 hours with trypsin) of the raw material significantly improved the weight gain and protein intake. The combined methanol/acetone/water extraction and protein hydrolysis treatments were beneficial on all nutritional quality parameters of rapeseed protein. These results suggest that the protein-bound phenolic compounds, rather than the free phenolic compounds contribute mainly to the decrease in the nutritional values of proteins associated with phenolic compounds in raw flour before extraction.  相似文献
