A Campylobacter jejuni isolate obtained from a turkey liver, designated C101, and a C. jejuni isolate obtained from the feces of a chicken, designated C111, were used to inoculate their respective hosts. Isolate C101 depressed weight gain by 20% when inoculated into newly hatched poults or 4-day-old poults. It also caused death, hepatic necrosis, and generalized hemorrhages in turkey embryos. The chicken-derived isolate, C111, did not reduce weight gain in newly hatched chicks, but it did induce mortality in chicken embryos. The supernatant of the cultures of both C. jejuni isolates also caused mortality in embryos.

A grazing study was performed with the main objective of examining the effect of fenbendazole (FBZ) in a ‘dose and move’ system on nematode infections in calves with special emphasis on Dictyocaulus viviparus.

Three groups of six calves were grazed from May to October 1993. One group (DM7) was treated with FBZ and moved to aftermath (pasture which had only been mown) 7 weeks after turnout. The second group (DM9) was similarly treated and moved 9 weeks after turnout and the third group served as untreated pasture control group (PC) and was moved to aftermath 9 weeks after turnout.

FBZ treatment removed adult lungworms from DM7 and DM9. Tracer calves grazed during the first 7 or the first 9 weeks after turnout acquired mean burdens of 18 and 125 lungworms, respectively. In PC faecal larval counts increased until the end of August. Most of the animals in this group were then suffering from lungworm disease and emergency treatment with ivermectin was given. In both FBZ-treated groups, larvae reappeared in the faeces of some of the calves 4–5 weeks after treatment. Subsequent reinfection resulted in higher mean faecal larval counts in both groups 2 months after treatment, although variation in faecal larval counts was high. In DM7 values tended to be higher than in DM9. These higher larval counts were associated with mild signs of parasitic bronchitis in some calves of DM7, whereas no signs were seen in DM9.

At the end of the experiment, all calves, and also a group of six permanently housed non-infected control calves (HC), were infected experimentally with 5000 D. viviparus larvae to evaluate development of immunity. The worm counts at necropsy showed that all calves on pasture had developed immunity.

Engineering resistance against various diseases and pests is hampered by the lack of suitable genes. To overcome this problem we started a research program aimed at obtaining resistance by transfecting plants with genes encoding monoclonal antibodies against pathogen specific proteins. The idea is that monoclonal antibodies will inhibit the biological activity of molecules that are essential for the pathogenesis. Potato cyst nematodes are chosen as a model and it is thought that monoclonal antibodies are able to block the function of the saliva proteins of this parasite. These proteins are, among others, responsible for the induction of multinucleate transfer cells upon which the nematode feeds. It is well documented that the ability of antibodies to bind molecules is sufficient to inactivate the function of an antigen and in view of the potential of animals to synthesize antibodies to almost any molecular structure, this strategy should be feasible for a wide range of diseases and pests.Antibodies have several desirable features with regard to protein engineering. The antibody (IgG) is a Y-shaped molecule, in which the domains forming the tips of the arms bind to antigen and those forming the stem are responsible for triggering effector functions (Fc fragments) that eliminate the antigen from the animal. Domains carrying the antigen-binding loops (Fv and Fab fragments) can be used separately from the Fc fragments without loss of affinity. The antigen-binding domains can also be endowed with new properties by fusing them to toxins or enzymes. Antibody engineering is also facilitated by the Polymerase Chain Reaction (PCR). A systematic comparison of the nucleotide sequence of more than 100 antibodies revealed that not only the 3′-ends, but also the 5′-ends of the antibody genes are relatively conserved. We were able to design a small set of primers with restriction sites for forced cloning, which allowed the amplification of genes encoding antibodies specific for the saliva proteins ofGlobodera rostochiensis. Complete heavy and light chain genes as well as single chain Fv fragments (scFv), in which the variable parts of the light (VL) and heavy chain (VH) are linked by a peptide, will be transferred to potato plants. A major challenge will be to establish a correct expression of the antibody genes with regard to three dimensional folding, assembly and intracellular location.

Fibrocartilaginous embolization (FCE) of the spinal cord is a common disease in large breed dogs. There are only a few reports about this entity in small breed dogs and it has never been reported in chondrodystrophic breed. For definitive diagnosis histopathologic examination is necessary. Magnetic resonance imaging (MRI) as a potential diagnostic tool for intravitam diagnosis of FCE has been mentioned before, but results have not been reported so far. This report describes the neurological findings and MRI results in three small breed dogs, including a Pekingese dog, with FCE of the spinal cord. The disease was suspected in two animals based upon clinical and MRI-appearance and confirmed in the third by histopathological examination. In all three cases, similar focal intramedullary lesions, consisting of hyperintensive signals on T2-weighted images, were detected. Based on these findings, high-field MRI may be used as an antemortem tool for the diagnosis of FCE. It is also shown that FCE can occur in chondrodystrophic dogs.

In determining isolation effects in fragmented populations, the landscape matrix is not often considered. Usually simple distance measures are used to quantify degree of isolation. We tested the effect of the matrix on the presence of red squirrels in 354 wooded patches in the Brussels Region, by comparing several isolation measures. These were 1) distance to the nearest source patch, 2) the Hanski-measure (a combination of distance to and size of all possible sources), 3) effective distances calculated from different least cost models using the ArcView grid extension ‘Cost Distance’ (a combination of distance and resistance of the landscape, with different resistances for different landscape types) and 4) some combinations of the Hanski-measure and the effective distances. Size and quality of the target patches were always included in the tests of the predictive power of different isolation measures on squirrel presence/absence. All variables examined (patch size, quality and isolation) significantly influenced squirrel presence. Models using the effective distances gave the best results. Models including the Hanski-measure improved significantly when Euclidean distance was replaced by effective distance, showing that parameterisation of matrix resistance added significant additional explanatory power when modelling squirrel presence. This revised version was published online in July 2006 with corrections to the Cover Date.
