Evidence is presented that a number of mechanisms are involved in the resistance of commercial cultivars of potato to soft rottingErwinia spp. These include phytoalexins, phenolics, cell wall modification, and possibly proteins. The role of elicitor-active oligogalacturonides, which are released from plant cell walls by pectic enzymes, is discussed.  相似文献

A phytotoxic protein that evokes the typical symptoms of Verticillium wilt disease in seedlings of Gossypium hirsutum L. (Upland cotton) was isolated from culture filtrates of Verticillium dahliae. The protein was purified by ammonium sulfate precipitation, Sephadex-G100 fractionation, and native PAGE. The 18.5 kDa protein, designated VD18.5, appears to be a single subunit protein with an isoelectric point between 3 and 5. VD18.5 induces symptoms of leaf dehydration, chlorosis, necrosis and stem discoloration in seedlings of the disease susceptible cotton cultivar Siokra 1–4. The LD50 of VD18.5 on protoplasts of Siokra 1–4 was 18 μg mL−1. VD18.5 had no noticeable effect on Pima S-7, which is a disease resistant cultivar. Phytotoxic activity was partially destroyed at high temperature and was abolished by digestion with proteinase K. Mass spectrometry fingerprinting and protein sequence data from VD18.5 yielded no significant matches when submitted to the Mascot search engine and NCBI non-redundant protein databases, respectively. These results suggest that VD18.5 is a novel protein that may be involved in the development of some of the symptoms associated with Verticillium wilt disease in the cotton plant.  相似文献

Based on recent needs to accurately understand fire regimes and post-fire vegetation resilience at a supra-level for carbon cycle studies, this article focusses on the coupled history of fire and vegetation pattern for 40 years on a fire-prone area in central Corsica (France). This area has been submitted since the beginning of the 20th century to land abandonment and the remaining land management has been largely controlled by frequent fires. Our objectives were to rebuild vegetation and fire maps in order to determine the factors which have driven the spatial and temporal distribution of fires on the area, what were the feed backs on the vegetation dynamics, and the long-term consequences of this inter-relationship. The results show a stable but high frequency of small fires, coupled with forest expansion over the study period. The results particularly illustrate the spatial distribution of fires according to topography and vegetation, leading to a strong contrast between areas never burnt and areas which have been burnt up to 7 times. Fires, when occuring, affect on average 9 to 12% of the S, SE and SW facing slopes (compared to only 2 to 5% for the N facing slopes), spread recurrently over ridge tops, affect all the vegetation types but reburn preferentially shrublands and grasslands. As these fire-proning parameters have also been shown to decrease the regeneration capacity of forests, this study highlights the needs in spatial studies (both in terms of fire spread and vegetation dynamic) to accurately apprehend vegetation dynamic and functionning in fire-prone areas.This revised version was published online in May 2005 with corrections to the Cover Date.  相似文献

The continuous use of plowing for grain production has been the principal cause of soil degradation. This project was formulated on the hypothesis that the intensification of cropping systems by increasing biomass‐C input and its biodiversity under no‐till (NT) drives soil restoration of degraded agro‐ecosystem. The present study conducted at subtropical [Ponta Grossa (PG) site] and tropical regions [Lucas do Rio Verde, MT (LRV) site] in Brazil aimed to (i) assess the impact of the continuous plow‐based conventional tillage (CT) on soil organic carbon (SOC) stock vis‐à‐vis native vegetation (NV) as baseline; (ii) compare SOC balance among CT, NT cropping systems, and NV; and (iii) evaluate the redistribution of SOC stock in soil profile in relation to soil resilience. The continuous CT decreased the SOC stock by 0·58 and 0·67 Mg C ha−1 y−1 in the 0‐ to 20‐cm depth at the PG and LRV sites, respectively, and the rate of SOC sequestration was 0·59 for the PG site and ranged from 0·48 to 1·30 Mg C ha−1 y−1 for the LRV site. The fraction of C input by crop residues converted into SOC stock was ~14·2% at the PG site and ~20·5% at the LRV site. The SOC resilience index ranged from 0·29 to 0·79, and it increased with the increase in the C input among the NT systems and the SOC sequestration rates at the LRV site. These data support the hypothesis that NT cropping systems with high C input have a large potential to reverse the process of soil degradation and SOC decline. Projections show that the cultivation of microalgae will extend to the production of bio-based compounds, such as biofuels, cosmetics, and medicines. This will generate co-products or residues that will need to be valorized to reduce the environmental impact and the cost of the process. This study explored the ability of lipid-extracted Chlorella vulgaris residue as a sole carbon and nitrogen source for growing oleaginous yeasts without any pretreatment. Both wild-type Yarrowia lipolytica W29 and mutant JMY3501 (which was designed to accumulate more lipids without their remobilization or degradation) showed a similar growth rate of 0.28 h−1 at different pH levels (3.5, 5.5, and 7.5). However, the W29 cell growth had the best cell number on microalgal residue at a pH of 7.5, while three times fewer cells were produced at all pH levels when JMY3501 was grown on microalgal residue. The JMY3501 growth curves were similar at pH 3.5, 5.5, and 7.5, while the fatty-acid composition differed significantly, with an accumulation of α-linolenic acid on microalgal residue at a pH of 7.5. Our results demonstrate the potential valorization of Chlorella vulgaris residue for Yarrowia lipolytica growth and the positive effect of a pH of 7.5 on the fatty acid profile.  相似文献

The purpose of this study was to compare the sensitivity of modelled area burned to environmental factors across a range of independently-developed landscape-fire-succession models. The sensitivity of area burned to variation in four factors, namely terrain (flat, undulating and mountainous), fuel pattern (finely and coarsely clumped), climate (observed, warmer & wetter, and warmer & drier) and weather (year-to-year variability) was determined for four existing landscape-fire-succession models (EMBYR, FIRESCAPE, LANDSUM and SEM-LAND) and a new model implemented in the LAMOS modelling shell (LAMOS(DS)). Sensitivity was measured as the variance in area burned explained by each of the four factors, and all of the interactions amongst them, in a standard generalised linear modelling analysis. Modelled area burned was most sensitive to climate and variation in weather, with four models sensitive to each of these factors and three models sensitive to their interaction. Models generally exhibited a trend of increasing area burned from observed, through warmer and wetter, to warmer and drier climates with a 23-fold increase in area burned, on average, from the observed to the warmer, drier climate. Area burned was sensitive to terrain for FIRESCAPE and fuel pattern for EMBYR. These results demonstrate that the models are generally more sensitive to variation in climate and weather as compared with terrain complexity and fuel pattern, although the sensitivity to these latter factors in a small number of models demonstrates the importance of representing key processes. The models that represented fire ignition and spread in a relatively complex fashion were more sensitive to changes in all four factors because they explicitly simulate the processes that link these factors to area burned. The plant pathogenic fungus Corynespora cassiicola causes a severe leaf spot disease on more than 70 host plant species including Hevea brasiliensis . Genetic variability in 32 isolates of C. cassiicola collected from diverse hosts and locations in Sri Lanka and Australia was assessed using restriction fragment length polymorphism (RFLP) analysis of the internal transcribed spacer (ITS) region of ribosomal DNA and random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) analysis of total fungal DNA. Amplified ITS fragments from all 32 C. cassiicola isolates exhibited an identical size, and restriction analysis with seven different restriction endonucleases revealed identity in all of the detected DNA fragments. This finding of high genetic relatedness was further supported by the cloning and DNA sequencing of the ITS2 region from one Sri Lankan and one Australian isolate. However, RAPD-PCR profiles generated by 15 oligonucleotide decamer primers revealed significant polymorphism between groups of organisms. Genetic relationships among the isolates were determined by cluster analysis of the RAPD-PCR data and seven different RAPD groups were identified. Isolates showed strong correlations between the assigned RAPD group and the location and host plant genotype from which the isolate was collected. Correlations were also observed between the RAPD group, growth of the isolate and pathogenicity on different plant hosts.  相似文献

A puppy was evaluated for rapid growth and large stature. Extensive diagnostic evaluation suggested a growth hormone independent disorder. As the sole detected abnormality was elevated leptin concentration, an obesity syndrome causing leptin resistance was speculated to explain the puppy's condition but was not confirmed.
