Soil ecosystem is experiencing stresses due to climate change, and soil inhabitants try to demonstrate their inherent resistance and resilience against those stresses. Application of nanomaterials as agricultural inputs could bring shifts in resistance and resilience patterns of soil microbes and associated enzymes, especially under short-term heat stress. With this background, the impacts of multi-walled carbon nanotube (MWCNT) on the resistance and resilience of soil biological indicators were evaluated. An incubation experiment was conducted with varied MWCNT concentrations (0, 50, 100, 250, and 500 mg kg-1 soil) for 90 d after 24-h heat stress at 48 ±2 ℃ to assess the impacts of MWCNT on soil enzyme activities and microbial populations vis-à-vis their resistance and resilience indices under short-term exposure to heat stress. Enzyme activities were reduced after exposure to heat stress. Resistance indices of enzyme activities were enhanced by MWCNT application on day 1 after heat stress, whereas there was no recovery of enzyme activities after 90-d incubation. Like soil enzyme activities, resistance index values of soil microbial populations followed the similar trend and were improved by MWCNT application. Multi-walled carbon nanotube has the potential to improve resistance indices of soil enzyme activities and microbial populations under heat stress, although they could not recover to their original state during periodical incubation after heat stress. This study helps to understand the relative changes of biological indicators under MWCNT and their ability to withstand heat stress.  相似文献

Asian rice gall midge (Orseolia oryzae) is a major pest across much of south and southeast Asia. This pest is genetically diverse and many gall midge biotypes are known to exist in each country. During the last three decades, host plant resistance has proved to be the most effective mechanism of controlling the Asian rice gall midge. Seven genes conditioning resistance to gall midge larvae have been identified in rice (Oryza sativa) and are being used in cultivar improvement programs. However, some of these genes are rendered ineffective by new gall midge biotypes. Increased understanding of genetics, inheritance, allelic relationships and linkage is necessary to maximise the durability of major gene resistance by the pyramiding of these genes. The two genes, Gm-2 and Gm-6(t), are known to confer resistance against a number of biotypes in India and China, respectively. An F3 population derived from a cross between Duokang #1 (donor of Gm-6(t)) and Phalguna (donor of Gm-2) was screened against Chinese gall midge biotype 4 at Guangdong, China, and Indian gall midge biotype 1 at Raipur, India. At each location, separately,a single gene governed resistance. The parallel segregation of 417 F3progenies for both biotypes at two locations revealed that recombination had occurred between the two genes, establishing that the two genes are not allelic. However, the two genes Gm-2 and Gm-6(t), were found to be linked with a distance of ∼16.3 cM. A number of lines homozygous at one locus and segregating for the other locus were identified and selected. These lines were selfed to obtain lines homozygous for the favourable alleles at both loci (two locus pyramids). This is the first report on use of conventional host-pest interaction method for pyramiding two closely located Gm-resistance loci of dissimilar effects. The implications of deployment of these pyramids within and across country borders, with reference to the prevailing gall midge populations are discussed. Genes normally resident in euchromatic domains are silenced when packaged into heterochromatin, as exemplified in Drosophila melanogaster by position effect variegation (PEV). Loss-of-function mutations resulting in suppression of PEV have identified critical components of heterochromatin, including proteins HP1, HP2, and histone H3 lysine 9 methyltransferase. Here, we demonstrate that this silencing is dependent on the RNA interference machinery, using tandem mini-white arrays and white transgenes in heterochromatin to show loss of silencing as a result of mutations in piwi, aubergine, or spindle-E (homeless), which encode RNAi components. These mutations result in reduction of H3 Lys9 methylation and delocalization of HP1 and HP2, most dramatically in spindle-E mutants.  相似文献

Benign Theileria species of cattle are found in most parts of the world. The major piroplasm surface protein (MPSP), a conserved protein in all Theileria species, has been used as a maker for epidemiological and phylogenetical studies of benign Theileria species. Parasites with Ikeda- or Chitose-type MPSP genes are dominant in Japan, but we report here mixed infection cases of Theileria parasites with an additional MPSP type parasite infecting cattle in Abashiri District, Hokkaido. The MPSP gene sequence found in the additional type was closely related to MPSP genes of Theileria parasites found in Southeast Asian countries, including Thailand (Narathiwat) and Indonesia (Java). Theileria parasites from the blood sample were also distinguishable from the Ikeda or Chitose type parasites by the small subunit (SSU) rRNA gene sequence analysis, and they are grouped into the SSU rRNA types C/D found in Korea, North America, and Spain. The present finding of mixed infections of cattle with three different types of Theileria makes epidemiological feature of bovine theileriosis in Japan more complex. We have designed a set of primers specific to this MPSP type in order to conduct further epidemiological study.  相似文献

Sericulture is an agro-based industry, which heavily depends on mulberry (Morus spp.) for its sustainability, as mulberry leaf is the only available feed for the silkworm Bombyx mori L. Sericulture, in West Bengal and other subtropical regions in India, suffers from lack of adequate quantity of quality mulberry leaf during the colder months, the best season for rearing the high yielding bivoltine silkworm hybrids. In order to develop mulberry varieties, which can sustain normal growth during these colder months to yield better, efforts have been made in different research institutes in India. The present report is part of such an effort in which accessions in germplasm bank were screened for better growth, leaf retention and leaf yield contributing associated traits. Twenty three selected parents were crossed, 3,500 seeds were sown, 2,700 seedlings were transplanted to progeny row trail, selected 210 hybrids were evaluated under primary yield trial and finally nine hybrids along with a control variety were studied in detail for all leaf yield contributing traits under final yield trial. Stability analysis was adopted to identify hybrids, which can yield stably across seasons. The hybrids CT-44 and CT-11 out yielding the control by 17.17 and 7.11% were selected as these hybrids yield 7.93 and 8.15 mt/ha leaf respectively during the colder months (February) for their direct use as cultivars in West Bengal and other subtropical areas of India to sustain bivoltine sericulture to produce quality and gradable silk fibers.  相似文献

The present study was on assessment of the anti-parasitic activities of nickel nanoparticles (Ni NPs) against the larvae of cattle ticks Rhipicephalus (Boophilus) microplus and Hyalomma anatolicum (a.) anatolicum (Acari: Ixodidae), fourth instar larvae of Anopheles subpictus, Culex quinquefasciatus and Culex gelidus (Diptera: Culicidae). The metallic Ni NPs were synthesized by polyol process from Ni-hydrazine as precursor and Tween 80 as both the medium and the stabilizing reagent. The synthesized Ni NPs were characterized by Fourier transform infrared (FTIR) spectroscopy analysis which indicated the presence of Ni NPs. Synthesized Ni NPs showed the X-ray diffraction (XRD) peaks at 42.76°, 53.40°, and 76.44°, identified as 1 1 1, 2 2 0, and 2 0 0 reflections, respectively. Scanning electron microscopy (SEM) analysis of the synthesized Ni NPs clearly showed that the Ni NPs were spherical in shape with an average size of 150 nm. The Ni NPs showed maximum activity against the larvae of R. (B.) microplus, H. a. anatolicum, A. subpictus, C. quinquefasciatus and C. gelidus with LC50 values of 10.17, 10.81, 4.93, 5.56 and 4.94 mg/L; r2 values of 0.990, 0.993, 0.992, 0.950 and 0.988 and the efficacy of Ni-hydrazine complexes showed the LC50 values of 20.35, 22.72, 8.29, 9.69 and 7.83 mg/L; r2 values of 0.988, 0.986, 0.989, 0.944 and 0.978, respectively. The findings revealed that synthesized Ni NPs possess excellent larvicidal parasitic activity. To the best of our knowledge, this is the first report on larvicidal activity of blood feeding parasites using synthesized Ni NPs.  相似文献

A long‐term fertilizer experiment, over 27 years, studied the effect of mineral fertilizers and organic manures on potassium (K) balances and K release properties in maize‐wheat‐cowpea (fodder) cropping system on a Typic Ustochrept. The treatments consisted of control, 100% nitrogen (100% N), 100% nitrogen and phosphorus (100% NP), 50% nitrogen, phosphorus, and potassium (50% NPK), 100% nitrogen, phosphorus, and potassium (100% NPK), 150% nitrogen, phosphorus, and potassium (150% NPK), and 100% NPK+farmyard manure (100% NPK+FYM). Nutrients N, P, and K in 100% NPK treatment were applied at N: 120 kg ha—1, P: 26 kg ha—1, and K: 33 kg ha—1 each to maize and wheat crops and N: 20 kg ha—1, P: 17 kg ha—1, and K: 17 kg ha—1 to cowpea (fodder). In all the fertilizer and manure treatments removal of K in the crop exceeded K additions and the total soil K balance was negative. The neutral 1 N ammonium acetate‐extractable K in the surface soil (0—15 cm) ranged from 0.19 to 0.39 cmol kg—1 in various treatments after 27 crop cycles. The highest and lowest values were obtained in 100% NPK+FYM and 100% NP treatments, respectively. Non‐exchangeable K was also depleted more in the treatments without K fertilization (control, 100% N, and 100% NP). Parabolic diffusion equation could describe the reaction rates in CaCl2 solutions. Release rate constants (b) of non‐exchangeable K for different depth of soil profile showed the variations among the treatments indicating that long‐term cropping with different rates of fertilizers and manures influenced the rate of K release from non‐exchangeable fraction of soil. The b values were lowest in 100% NP and highest in 100% NPK+FYM treatment in the surface soil. In the sub‐surface soil layers (15—30 and 30—45 cm) also the higher release rates were obtained in the treatments supplied with K than without K fertilization indicating that the sub‐soils were also stressed for K in these treatments.  相似文献
