Cloning of plasma membrane H-ATPase gene in Populus euphratica Oliv.

For this paper, the plasma membrane (PM) H -ATPase gene has been cloned from Populus euphratica Oliv. through a homology based strategy. The isolated 3,210 bp cDNA contains a single 2,862 bp open reading frame (ORF) which encodes a putative H -ATPase protein of 953 amino acid residues, with a significant homology to plasma membrane H -ATPase of Primus persica, Phaseolus vulgaris, Sesbania rostrata and Daucus carota. The predicted protein has a molecular weight of 104,553 Da. The copy number analysis revealed multiple copies of the PM H -ATPase in the P. euphratica genome after digestion of their genomic DNA by the restriction enzymes EcoRI, NdeⅠ, FbaⅠand BglⅡ, and Southern blot.

EFFECT OF HIGH LOADS IN THE BRIDGE LOAD SPECTRUM ON FATIGUE CRACK GROWTH OF 16 Mn STEEL

This paper studies an effect of high loads in Rayleigh distribution random load-ing on fatigue crack growth wi th 16 Mn steel compact tenion specimen,The results show that thehigh loads in the load spectrum close to service load of steel bridges accelerate crack growth and re-duce crack growth life,Lives predicted with constant amplitude da/dn data and equivalent loads arequite in agreement with test lives.     

ELLIPSOMETRIC SPECTROSCOPY OF ELECTROCHROMISM OF POLYANILINE FILMS

The electrochromism of polyaniline films, which is prepared by electropolymer-ization from aniline in acidic aqueous solution onto platinum,is investigated in 330-700nm through in-situ ellipsometric technique. The results indicate that the ellipsometric peak of the reduced state(-0. 2V) of the film appears at 390-400nm and the oxidized state(0. 55V) at 500-530nm. The elec-trochromic reaction from the reduced "yellow" state to the oxidized "green" state is suggested via intermediate state whose ellipsometric peak appears at 466-470nm.     

毕节岩溶区坡耕地烟草镉安全评价研究

以贵州省毕节地区岩溶环境烟水配套工程为例,通过实地调查和采样,对烟草地土壤-烟草系统镉进行评价。结果表明,烟草地土壤镉含量0.12~0.67 mg.kg-1,平均0.40 mg.kg-1,已遭轻度-中度污染。水溶态、离子交换态、碳酸盐结合态、腐植酸态、铁锰氧化物结合态、强有机结合态和残渣态镉分别为0.024、0.100、0.033、0.067、0.096、0.019 mg.kg-1和0.016 mg.kg-1,其中离子交换态、铁锰结合态和腐植酸态占66%。全镉与水溶态、离子交换态和铁锰结合态,水溶态与离子交换态和碳酸盐结合态,离子交换态与铁锰氧化物结合态显著正相关,揭示了镉形态间的转化关系及其对烟草的潜在影响。烟株上部烟叶镉含量1.04～1.59 mg.kg-1,茎、根中镉残留量为0.2～1.4 mg.kg-1,可能与源自K326的主推和示范品种对镉的提取效率高有关。因此,建议采用吸收镉低的烟草品种和合理的差异性水肥管理措施,固定土壤中活性态镉,降低土壤镉向烟株的转移,以保障烟叶品质,同时妥善处理这些农业废弃物以保护环境。     

Application of quantum dot-antibody conjugates for detection of sulfamethazine residue in chicken muscle tissue

A new indirect competitive fluorescence-linked immunosorbent assay (cFLISA) method for the detection of sulfamethazine (SM2) in chicken muscle tissue was demonstrated using quantum dots (QDs) as the fluorescence label coupled with secondary antibody. The sensitivity of the cFLISA was compared with that of the HPLC method. The 50% inhibition value (IC50) and the limit of detection (LOD) of the cFLISA were 7.7 and 1.0 ng/mL, respectively. When SM2 was spiked at levels of 50, 100, and 200 ng/g, recoveries ranged from 80.6% to 117.4%, with a coefficient of variation of 6.9-9.6%. In the incurred sample analysis, the amounts of SM2 quantified by cFLISA were similar to the results obtained by the HPLC method. This study shows that cFLISA could be used as a screening method in practice.

The Avalon explosion: evolution of Ediacara morphospace

Ediacara fossils [575 to 542 million years ago (Ma)] represent Earth's oldest known complex macroscopic life forms, but their morphological history is poorly understood. A comprehensive quantitative analysis of these fossils indicates that the oldest Ediacara assemblage-the Avalon assemblage (575 to 565 Ma)-already encompassed the full range of Ediacara morphospace. A comparable morphospace range was occupied by the subsequent White Sea (560 to 550 Ma) and Nama (550 to 542 Ma) assemblages, although it was populated differently. In contrast, taxonomic richness increased in the White Sea assemblage and declined in the Nama assemblage. These diversity changes, occurring while morphospace range remained relatively constant, led to inverse shifts in morphological variance. The Avalon morphospace expansion mirrors the Cambrian explosion, and both events may reflect similar underlying mechanisms.
