土地开发整理前期地形图测绘调研分析

通过对重庆市彭水县高谷镇共和村土地开发整理测绘项目的实地调研分析,对该项目进行了归纳总结,介绍项目的测绘技术和流程,旨在为今后推进农村土地开发整理测绘提供实践依据。     

广西县级林地变更调查试点技术方法

为探索广西县级林地年度变更调查技术方法,以鹿寨县为例,运用遥感自动检测技术、森林资源管理信息平台等,对地类及管理属性变化图斑进行变更,形成本期林地数据库和林地变化数据库。文章分析了林地变更调查的关键技术方法,指出了存在的问题,并提出相应的建议。     

Information extraction and dynamic evaluation of soil salinization with a remote sensing method in a typical county on the Huang-Huai-Hai Plain of China

Monitoring the dynamics of soil salinization is of great importance for agricultural production. This study selected Yucheng County, a typical county on the Huang-Huai-Hai Plain (HHHP) of China, as the study area and evaluated the spatial and temporal variation of soil salinization. Three methods, consisting of principal component analysis (PCA) transformation, tasseled cap (TC) transformation, and optimal band combination (OBC), were used to extract information from an early Landsat multispectral scanner (MSS) image from 1984, and their advantages were compared. In addition, OBC was used on a thematic mapper (TM) image from 2009. An iteratively self-organizing data analysis algorithm was used together with prior knowledge of likely classifications to interpret the MSS and TM images for data classification. Finally, a transfer matrix method was used to assess the spatial and temporal variability of soil salinization and analyze the driving factors of soil salinization. Compared to PCA transformation and OBC, TC transformation was a more effective method for extracting soil salinization information from the MSS sensor. The results indicate that a soil area of approximately 298 km² was affected by salinity in 1984 in Yucheng County, of which 5.40%, 11.96%, and 12.75% were classified as being subject to slight, moderate, and severe salinization, respectively. In 2009, the saline area was reduced to only 146 km², of which 10.70% and 3.75% were characterized by slight to moderate salinization and no severe salinization, respectively. The saline land decreased at an average rate of 6 km² per year. This decrease was probably a result of lower groundwater depth, increased organic fertilizer or crop straw in soil, changed land use type, and increased vegetation coverage.     

胡萝卜根中主要类胡萝卜素含量相关QTL的精细定位

以白色的野生胡萝卜‘松滋野生’(Ws)和橘色的栽培胡萝卜品种‘Amsterdam’(Af)为亲本构建的回交重组自交系(BIL)为试材,基于低倍重测序技术开发SNP标记,构建了由1 976个Bin标记组成,包含29 435个SNP标记的遗传图谱。图谱总距离834.28 c M,平均图距0.42 c M。通过对胡萝卜肉质根中类胡萝卜素含量相关QTL分析,在连锁群LG04和LG08中检测到调控α–胡萝卜素、β–胡萝卜素、ζ–胡萝卜素、叶黄素、玉米黄质和总类胡萝卜素含量的主效QTL(M-QTL)2、2、3、2、2和2个,表型贡献率为11.47%~19.18%;另检测到调控α–胡萝卜素、β–胡萝卜素、ζ–胡萝卜素、玉米黄质和总类胡萝卜素含量的上位性QTL(E-QTL)1、1、2、1和1个,表型贡献率为2.50%~3.66%。在M-QTL显著区间内共检索到36个有功能注释的预测基因,其中Dck018297为ζ–胡萝卜素脱氢酶2基因,与调控β–胡萝卜素合成和总类胡萝卜素含量有关;Dck008006为乙烯响应因子2.2的同源基因,与调控α–胡萝卜素、ζ–胡萝卜素合成有关;Dck029898为转录因子b HLH135的同源基因,与调控玉米黄质合成有关。     

Genetic analysis and QTL mapping of growth period traits and plant height traits in soybean recombinant inbred lines from Dongnong 47 × PI 317334-B

High and stable yield is the main goal of soybean genetic improvement. In this study, association analysis was used to detect the quantitative trait loci (QTL) for the plant height, and soybean growth period using 182 SSR markers in the RIL population of 136 F_4:8 lines, which developed from a cross between photoperiod-insensitive cultivar ‘Dongnong 47’ and photoperiod-sensitive variety PI317334–B. The results showed that 33 QTLs related to soybean growth period and plant height traits were detected by compound interval mapping, and were located on 12 linkage groups including N, C1, C2, J, D1a, B2, E, G, A2, O, L, I, with the contribution rate of 7.85–33.84%. These QTL loci and linkage markers related to soybean photoperiod sensitivity, would be helpful to identify key genes that control soybean photoperiod sensitivity, and provide an important basis for the breeding of new photoperiod-insensitive soybean varieties based on molecular design breeding.     

棉花单核苷酸多态性标记研究进展

单核苷酸多态性标记已在农作物研究中得到广泛应用并取得重大进展。为了便利棉花SNP(Single nucleotide polymorphism)标记的研究和应用,介绍了利用基因芯片、简化基因组测序、重测序等在棉花中开发SNP标记的方法 ,综述了SNP标记在棉花遗传图谱构建、数量位点的定位和分子标记辅助育种、基因组测序以及系统进化等研究中的应用。并对异源四倍体棉花中SNP标记开发时,同源序列位点和部分同源序列位点上的SNP标记辨别问题进行了系统探讨,对其快捷的开发、检测方式和在数量基因定位中的应用前景进行了展望。     

SSR标记技术在马铃薯遗传育种研究中的应用

基于PCR基础上发展起来的SSR分子标记技术具有共显性遗传、多态性高、重复性好、稳定性好、操作简单等优点，本文简述了SSR分子标记技术的基本原理与特点，重点对马铃薯SSR引物的开发及其在马铃薯品种鉴定、遗传图谱构建与数量性状基因定位、分子标记辅助选择育种的研究现状进行了分析，并对其应用前景进行了展望。     

实例教学法在建筑电气施工图识图教学中的运用--以人为本，学以致用

文章针对识图教学过程中运用实例教学的重要性进行阐述,并针对实例教学法在建筑电气施工识图教学中的应用加以探讨,以提供理论参考。     

Basic characterization of avian β‐defensin genes in the Japanese quail,Coturnix japonica

In this study, we identified a cluster of 14 avian β‐defensins (AvBD; approximately 66 kbp) in the Japanese quail, Coturnix japonica. Except for AvBD12 (CjAvBD12) and ‐13, the CjAvBDs coding sequences exhibited greater than 78.0% similarity to the respective orthologous chicken AvBD genes (GgAvBD). The putative amino acid sequence encoded by each CjAvBD contained six cysteine residues and the GXC (X1‐2) motif considered essential for the β‐defensin family. Each CjAvBDs also formed a sub‐group with the respective orthologous genes of various bird species in a phylogenetic tree analysis. Synteny between the CjAvBD cluster and GgAvBD cluster was confirmed. The CjAvBD cluster was mapped on the long‐arm end of chromosome 3 by linkage analysis based on single nucleotide polymorphisms (SNPs) of CjAvBD1 and CjAvBD12 (approximately 46kbp), as well as GgAvBD cluster. We also confirmed that CjAvBD1, ‐4, ‐5, ‐9, and ‐10 are transcribed in 20 tissues, including immune and digestive tissues. However, our experimental data indicated that the CjAvBD cluster lacks the AvBD3 and ‐7 loci, whereas the CjAvBD101α, ‐101β, and ‐101θ loci arose from gene duplication of the AvBD6 orthologous locus in the CjAvBD cluster after differentiation between Coturnix ‐ Gallus.
