The biological activity of the Ireland strain of Choristoneura fumiferana (Clem) nucleopolyhedrovirus (CfMNPV) propagated in different hosts was determined to provide the basis upon which genetically modified CfMNPV, or other naturally occurring isolates, should be compared. Occlusion bodies (OB) derived from CF-203 cells were significantly larger and more pathogenic than those propagated in vivo when tested against the fifth larval instar of C fumiferana (Clem) and C occidentalis Freeman. The dose-responses (LD50 and LD95, expressed as occlusion bodies per larva) of C fumiferana larvae to in vitro-propagated OBs were 274 and 5785, respectively. The values of LD50 and LD95 to C occidentalis larvae were 19 and 118, respectively. There were no significant differences in pathogenicity or size when OBs propagated in C fumiferana larvae were tested against either insect species, nor were there significant differences for OBs propagated in C occidentalis larvae. The LD50 and LD95 of in vivo-produced OBs to C fumiferana were 925 and 61988, respectively. The LD50 and LD95 to C occidentalis were 50 and 453, respectively. OBs propagated in vitro had a mean volume of 13.13 microm3, whereas those propagated in vivo ranged from 0.84 to 1.41 microm3. The median survival time-responses (ST50) of fifth-instar C fumiferana or C occidentalis larvae to OBs propagated in vivo were not significantly different from those propagated in vitro at the dosage levels tested. Values of ST50 of C fumiferana larvae to in vitro- and in vivo-produced OBs at dosages causing less than 50% mortality rangedfrom 9.6 to 9.8 days post-inoculation (dpi), whereas a LD95 dose resulted in ST50 values ranging from 7.3 to 7.7 days. ST50 values of C occidentalis larvae at dosages causing less than 50% mortality ranged from 9.8 to 10.2 dpi, whereas a LD95 dose resulted in ST50 values ranging from 9.5 to 9.8 dpi. The median feeding cessation time-response (FT50) of fifth-instar C fumiferana larvae to OBs propagated in vitro (5.7 days) was not significantly different from the FT50 of those propagated in vivo in either insect species (5.3 and 5.7 days) at the dosage level tested (LD95). No significant differences in FT50 values were observed between OBs propagated in either larval host. The FT50 of C occidentalis larvae to OBs propagated in vitro (7.7 days) was not significantly different from that to those propagated in vivo in C occidentalis larvae (7.6days), but somewhat different (7.2 days) from that to those propagated in C fumiferana larvae. Results indicate that CfMNPV can be propagated in vivo in either C fumiferana or C occidentalis larvae (or sequentially through both) without alteration in infectivity, although the use of the CF-203 cell line yields the most biologically active OBs.  相似文献

Fish are always susceptible to a wide variety of deadly pathogens which cause a huge loss in aquaculture industries. In this investigation, we have demonstrated the in vivo probiotic efficiency of Bacillus sp. MVF1 (GenBank Acc. No. KP256503) in Labeo rohita challenged with pathogenic strain of Aeromonas hydrophila (MTCC 1739). To check the probiotic potential of the selected bacterial strain, fish were divided into four groups: control, D1, D2 and D3. A total of 100 days (70 days probiotic feeding + 71th day sampling and 28 days challenged test + 29th day sampling) of feeding trial was conducted. To establish the probiotic potential of Bacillus sp. MVF1, certain haematological parameters (haemoglobin, total erythrocyte and leucocyte count), serum biochemical parameters (total protein, albumin and globulin), immune parameters (serum lysozyme and total IgM levels) and hepatic stress profile (malondialdehyde production, superoxide dismutase and catalase activity) have been measured. Our results demonstrated that red blood cell number, white blood cell number and haemoglobin content were much higher in D2 group fish compared to other groups and control fish. Similarly, total protein contents, albumin concentration, globulin concentration, lysozyme activity and IgM production were also recorded to be highest in D2 group fish. This finding clearly indicated the probiotic potential of Bacillus sp. MVF1 in L. rohita. Furthermore, our results also demonstrated that 1 × 107 CFU g?1 feed (D2) provides better immunity compared to 1 × 105 (D1) and 1 × 109 (D3). Due to beneficial effects, the bacterium Bacillus sp. MVF1 might be useful in aquaculture industries to reduce the disease susceptibility.  相似文献

基于江苏省南通市2000~2004年的旬气象资料,用FAO推荐的Penman-Monteith公式计算了参考作物蒸发蒸腾量,研究了参考作物蒸发蒸腾量与最高气温、最低气温、平均气温、相对湿度、日照时数、风速和气压等气象因素间的关系,建立了参考作物蒸发蒸腾量的响应模型.结果表明,参考作物蒸发蒸腾量与"温度因子"的关系最强,其次为"湿度和日照因子","风速因子"也有一定的影响,"气压因子"影响作用则稍弱;建立的气象因子响应模型模拟精度较高,可以简化参考作物蒸发蒸腾量计算.  相似文献

以蓝莓为原料,考察了复合酶添加量、复合酶配比、酶解温度、酶解时间4个因素对蓝莓出汁率的影响。在单因素试验的基础上,采用Box-Behnken响应面设计对复合酶酶解蓝莓汁的工艺参数进行进一步的优化。结果表明,复合酶法制取蓝莓汁的最佳工艺参数为:复合酶添加量0.20%,复合酶(果胶酶∶纤维素酶)配比3∶1(m∶m),酶解温度47℃,酶解时间84 min,该工艺条件下蓝莓出汁率达78.38%,与模型预测值78.54%相比,其相对误差约为0.20%,表明在蓝莓汁复合酶法制取过程中采用响应面试验设计对工艺参数进行优化是可行的。  相似文献

为探究当归微波真空干燥的最佳工艺参数,以甘肃定西“岷归1号”为试验材料。运用HPLC法测定不同干燥温度、切片厚度、真空度条件下当归中藁本内酯(LA)、阿魏酸(FA)、洋川芎内酯H(SH)、洋川芎内酯I(SI)、绿原酸(CA)、丁烯基苯酞(BA)、抗氧化性(DPPH)、总酚(TPC)、总黄酮(TFC)、多糖(Pc)的含量,利用AHP-熵权法计算各有效活性成分的复合权重和综合评分;以综合评分为响应值,通过Box Behnken响应面法进行优化设计,确定当归微波真空干燥的最佳工艺参数。结果表明:AHP-熵权法计算得出LA、FA、SH、SI、CA、BA、DPPH、TPC、TFC、Pc、RR的Qj值分别为0.159、0.192、0.120、0.288、0.033、0.063、0.059、0.041、0.024、0.013、0.009;Box Behnken响应面法得到的当归微波真空干燥的最优工艺条件为干燥温度50 ℃、切片厚度5 mm、真空度-0.070 MPa,此时综合评分为84.95分;此外,微观电镜结果也表明在此工艺条件下,样品质构热稳定性最好且内部出现均匀规则的蜂窝状孔隙结构。因此AHP-熵权法可用于当归微波真空干燥工艺中多目标的优化。  相似文献

Objective: To determine the frequency of the systemic inflammatory response syndrome (SIRS) in canine pyometra and to evaluate the relationship between C‐reactive protein (CRP), tumor necrosis factor α (TNFα), interleukin‐6 (IL‐6), and SIRS. Design: Prospective clinical study. Setting: Veterinary teaching hospital. Animals: Fifty‐three clinical cases of canine pyometra and 19 healthy control bitches. Interventions: Upon admission to the veterinary hospital, history and physical examination findings, including previously defined clinical SIRS parameters, were documented. Blood samples were obtained for hematology and biochemical tests and for CRP, TNFα, and IL‐6 analysis. The diagnosis of pyometra was confirmed by histopathology of the uterus after ovariohysterectomy. After surgery, clinical SIRS parameters, length of hospitalization, and mortality were recorded. Measurements and main results: Pyometra dogs were grouped as SIRS positive (30/53; 57%) or SIRS negative (23/53; 43%). Logistic regression showed that CRP was the only parameter that significantly related to SIRS apart from the clinical criteria that define this syndrome. The mortality rate was low (2/53; 3.8%), and conclusions regarding association with SIRS could not be drawn. A positive SIRS status, high plasma CRP concentration, and high body temperature were variables that related to increased morbidity reflected by the length of hospitalization. Conclusions: SIRS was seen in 57% of canine pyometra cases and a positive SIRS status showed a positive association with prolonged hospitalization. The mortality rate was low (3.3%) among SIRS positive dogs, indicating that progression to multiple organ dysfunction syndrome (MODS) rarely occurs in surgically treated cases of pyometra. CRP was associated with SIRS and with prolonged hospitalization. Further studies of plasma CRP may be warranted in canine intensive care cases susceptible to development of SIRS and MODS.  相似文献

To measure the contribution of root respiration (Rr) to total soil respiration (Rt) in arid cotton fields, eighteen plots, nine for girdling and nine control, were built in an arid cotton field in the Aksu National Experimental Station of Oasis Farmland Ecosystem, Xinjiang of China. Given the difference of soil respiration between girdled plots and non-girdled control plots, the components of soil respiration, root respiration (Rr) and respiration originating from decomposition (Rd) were divided. The temperature sensitivities of R r and R d were analyzed, respectively. The results showed that the average contribution of R r to R t in arid cotton field was about 32% during the study period. The temperature-response curve of R r differed from that of Rd . The dynamic variation of R d was more related to the change of soil temperature as compared to Rr . Rr and Rd had different responses to the variation of environment, and thus new models capable of differentiating between Rr and Rd are needed for evaluating the different factors controlling these two components of soil respiration in arid cotton field.  相似文献

AIM: To investigate the effect of rosiglitazone, a peroxisome proliferators-activated receptor γ(PPARγ) agonist, on the expression of PPARγ, the activation of NF-κB and intestine injury in the rats undergoing orthotopic autologous liver transplantation(OALT).METHODS: Sprague-Dawley male rats were randomly divided into 4 groups:control group, sham group, OALT group and rosiglitazone(0.3 mg/kg, iv) pretreatment(ROS+OALT) group. The OALT model was established, and the intestinal tissues were collected 8 h after the liver reperfusion. The intestinal tissue sections were stained to visualize the damage. The expression of PPARγ and NF-κB in the tissues, the concentrations of diamine oxidase(DAO) and fatty acid-binding protein 2(FABP2) in the serum and the concentration of TNF-α and IL-6 in the tissues were measured.RESULTS: Compared with sham group, the intestinal mucosa of the rats showed obvious pathological injury after liver reperfusion in OALT group and ROS group, the Chiu,s scores of intestinal mucosa was significantly higher, and the serum concentrations of DAO and FABP2 increased(P<0.05). After rosiglitazone pretreatment, the injury of intestinal mucosa of the rats was alleviated, the Chiu,s scores was lower and the serum concentrations of DAO and FABP2 decreased(P<0.05), the PPARγ expression was obviously up-regulated in the intestinal tissues, the nuclear translocation of NF-κB was reduced and the concentrations of IL-6 and TNF-α were decreased.CONCLUSION: During perioperative period of OALT in rats, the inflammatory responses are obvious. Furthermore, obvious intestinal injury occurs. PPARγ agonist rosiglitazone obviously up-regulates PPARγ expression and inhibits the inflammation in the intestines, thus protecting against intestinal injury in rats undergoing OALT.  相似文献

【目的】筛选对双斑长跗萤叶甲Monolepta hieroglyphica (Motschulsky)成虫有生物活性的植物挥发物。【方法】通过气相-质谱联用仪(GC-MS)对棉花和玉米挥发物进行鉴定及定量分析,采用触角电位技术 (EAG)测定双斑长跗萤叶甲雌、雄成虫对10种挥发物的触角电位反应。【结果】双斑长跗萤叶甲雄虫对α-长叶蒎烯、α-红没药醇、α-蒎烯、月桂烯、3-蒈烯、1,3丁二烯、棕榈酸反应较为明显;雌虫对10种挥发物部分浓度均有明显反应。【结论】双斑长跗萤叶甲雄虫对3-蒈烯反应最敏感,雌虫对α-红没药醇反应最敏感。  相似文献
