反向遗传技术在牛轮状病毒研究中的应用

本文综述了牛轮状病毒的生物学特性、反向遗传学系统的建立及其研究进展,并对反向遗传技术应用于牛轮状病毒进行展望。

Pathogen Analysis of Goat's Respiratory Diseases

To investigate the pathogens of goat's respiratory disease from a goat farm in Guangxi province, the epidemiological investigation, clinic observation, pathological examination, bacterium isolation and identification, biochemical test, PCR and animal experiment test were conducted.The preventive and control measures were taken on the basis of pathogen epidemiological characteristics and drug sensitive test results.The results showed that Mycoplasma, influenza virus and parasite were negative by isolation and culture or PCR, however, two strains of gram-negative bacteria named MS1 and MS2 were isolated from lung.MS1 was identified as Serratia marcescens by biochemical test and 16S rRNA sequencing, which shared 99% nucleotide homology with other Serratia marcescens in GenBank.And MS2 was identified as E.coil by the same methods that shared 99% nucleotide homology with other E.coli in GenBank.Animal experiment showed that two strains could cause death in mice.The drug sensitive tests showed two strains were highly sensitive to spectinomycin, amikacin, kanamicin and neomycin.Kanamycin and dexamethasone were used to treat the sick goats, and achieved good results.     

波叶大黄白粉病病原菌的鉴定

正波叶大黄Rheum rhabarbarum L.隶属于蓼科Polygonaceae大黄属Rheum,是一种重要的药用植物。白粉病多发生在波叶大黄的叶部,会影响其光合作用进而降低其药用价值。据笔者统计,目前关于蓼白粉菌Erysiphe polygoni DC.在世界范围内的报道有很多~([1~9]),但其中关于蓼科Polygonaceae波叶大黄白粉病病原菌的报道较少,目前仅有1个     

甜玉米‘闽甜208’的生育特性与群体结构分析

多年生育特性观测表明,‘闽甜208’甜玉米春播采青生育期86-90d,秋播采青生育期73-76d,生育动态呈现苗期弱,中间旺长稳定期短,后期早衰快的生育特性,栽培上要注重精细播种,各项田间管理要及时到位。群体结构试验表明,‘闽甜208’叶面积系数（LAI）及叶片净同率（NAR）较高,峰值持续时间较长,大喇叭口至抽雄期LAI对NAR影响最大,通过密植保证一定的总粒数,增加抽雄后群体叶面积及干物质积累,提高成粒率,是高产的关键措施,适宜种植密度为4.95万-5.40万株·hm-2。     

极小种群野生植物八蕊单室茱萸的种群状况研究

八蕊单室茱萸为云南省特有的极小种群野生植物和濒危物种。通过查阅相关文献并结合访问调查,确定八蕊单室茱萸可能分布的区域,开展种群资源调查。结果显示:八蕊单室茱萸目前仅在普洱市和西双版纳州有10个天然种群,其种群规模已低于最小可存活种群,属极小种群野生植物和濒危物种,亟需开展保护工作。生境破坏和生境片段化是导致八蕊单室茱单室茱萸濒危的人为因素。提出收集种质资源、营建和管护近地保护种群、建立迁地和回归种群等保育建议。     

2013年国外家兔遗传育种研究进展

我国是养兔大国，但家兔遗传育种基础研究薄弱。文章对2013年国外家兔遗传育种最新研究进展进行综述，以期为国内家兔育种者提供参考。     

Transgenic expression of a sorghum gene (SbLRR2) encoding a simple extracellular leucine-rich protein enhances resistance against necrotrophic pathogens in Arabidopsis

Plant leucine-rich repeat (LRR) domain-containing proteins are known to play important roles in signaling transduction and defense responses. In sorghum, SbLRR2 is pathogen-inducible gene encoding a simple extracellular LRR protein. Here, we demonstrated an earlier and stronger expression of SbLRR2 in a sorghum resistant genotype in comparison to a susceptible genotype following inoculation with the anthracnose pathogen (Colletotrichum sublineolum). In addition, SbLRR2 expression was found to be induced strongly by methyl-jasmonate treatment. Functional analysis was performed in SbLRR2 over-expression (OE) Arabidopsis plants, which showed enhanced resistance against the necrotrophic pathogens Botrytis cinerea and Alternaria brassicicola. In addition, the OE lines were found to have elevated expression of several jasmonate acid (JA)-associated genes and higher endogenous JA contents. Hence, the SbLRR2-mediated defense responses in transgenic Arabidopsis are likely to be dependent on JA-signaling through increased JA production. On the other hand, the OE lines remained susceptible to Pseudomonas syringae pv. tomato like the wild type plants. Consistently, there was no up-regulation of salicylic acid (SA) defense marker gene expression or SA levels in the OE lines. Our results suggested that SbLRR2 is potentially useful for enhancing resistance against necrotrophic pathogens in transgenic dicot crops.     

被动式电子标签在监测高原鼢鼠种群生态变化中的应用

地下啮齿动物种群生态学研究经常需要对动物个体进行标记,而传统的剪耳、剪趾标记法会造成动物伤害、影响动物行为等,与动物福利相悖。利用PIT标签于2014年秋季在祁连山东段标记高原鼢鼠30只,研究高原鼢鼠种群密度及越冬后体重、体长和尾长变化。结果表明,样地内高原鼢鼠估算种群密度为44只/hm2;越冬后高原鼢鼠平均体重为222.07g,显著高于越冬前平均体重193.74g,平均体长为19.4cm,显著高于越冬前平均体长18cm;雌性与雄性的体重增长率无显著性差异(P0.05)。被动式电子标签同时可适用于地下啮齿动物的种群生态学研究,是地下啮齿动物种群生态学研究中的一项新型技术手段。     

储粮害虫磷化氢抗性分子遗传学研究进展

由于长期单一使用磷化氢,储粮害虫对其产生的抗药性日益增强,已成为威胁粮食安全储藏的重大问题。自从发现两个磷化氢抗性基因rph1和rph2,以及完成赤拟谷盗全基因测序以来,储粮害虫磷化氢抗性基因的相关研究迅速发展。近年来,在抗性基因的鉴定、表达、进化、突变等研究方面取得了一定成果,为储粮害虫磷化氢抗性治理提供了新的科学指导。本文就储粮害虫磷化氢抗性分子遗传学的研究进展进行综述。
