Apoptosis is proposed to be a major cause of death in shrimp viral infections. From our previous study, an apoptosis-related gene, Pm-Alix, was identified from the black tiger shrimp. Its expression was high in defence-related tissues including haemocytes and the lymphoid organ. To clarify its possible role in shrimp, we used Pm-Alix as bait in a yeast two-hybrid analysis to search for Alix interacting proteins in shrimp. Two cDNA sequences discovered had homology to a predicted ubiquitin C of the purple sea urchin, Strongylocentrotus purpuratus, and to a guanylyl cyclase of the red swamp crayfish, Procambarus clarkii. In vitro pull-down assays confirmed positive interaction between Pm-Alix and both proteins. Tissue distribution analysis revealed that Pm-Alix and the two binding partners were widely expressed in various tissues but more highly expressed in haemocytes. However, no significant positive or negative correlation was found in the expression of these genes as shrimp approached morbidity and death after challenge with white spot syndrome virus. Thus, the results suggested that Alix and its interacting partners did not play a direct role related to shrimp death.  相似文献

The defence ability of Fenneropenaeus chinensis and Marsupenaeus japonicus against the intrusion of the white spot syndrome virus (WSSV) was compared after injecting WSSV intramuscularly by recording cumulative mortality, diagnosing the virus and examining variations in immunological parameters including the total haemocyte counts (THCs), phagocytic percentage (PP), plasma protein concentration (PPC), phenoloxidase (PO) and nitric oxide synthase (NOS) activity. The results showed that the variations in the immunological parameters of F. chinensis and M. japonicus showed similar trends. The THCs of the two species decreased significantly postchallenge of WSSV. The virus was detected at 78 h in M. japonicus and at 42 h in F. chinensis after infection, which was in correlation with the accumulative mortality, and the variations in PO, PP, NOS and PPC in the two species. All shrimps of F. chinensis in the mortality experiment died within 66 h, much more quickly than M. japonicus, whose cumulative mortality reached 100% after 198 h. In conclusion, the immune system of M. japonicus has a stronger resistant ability to antagonize and endure the invasion of WSSV than that of F. chinensis.  相似文献

A disease of Macrobrachium rosenbergii, the giant freshwater prawn, farmed in China was recently recorded in Zhejiang, Jiangsu, Shanghai, Guangxi and Guangdong provinces. The clinical sign of the disease, which develops in post-larvae (PL), is a whitish appearance of the muscles, particularly noticeable in the abdomen. Mortalities may reach 100% in some hatcheries. Investigations by transmission electron microscopy after negative staining of diseased PL homogenates showed the presence of two types of viral particles: one, unenveloped, icosahedral in shape, 26-27 nm in diameter, the second, much smaller, about 14-16 nm in diameter, designated extra small virus particle (XSV). The large virus has a genome with two pieces of ssRNA (RNA-1 and RNA-2), of 3 and 1.2 kb, respectively. Hybridization tests confirmed that this large virus is closely related to M. rosenbergii nodavirus (MrNV) which was isolated from diseased prawns in a hatchery in the French West Indies. Its very small size and hypothesized biochemical and biological characteristics suggest XSV is a new type of crustacean virus. As XSV has always been found associated with the larger virus (nodavirus) and is located in muscle and connective cells of diseased animals, it could be an autonomous virus, a helper-type virus or a satellite-like virus.  相似文献

2004年12月中下旬~2005年4月对辽宁沿海地区刺参越冬苗发病严重地区,进行了跟踪调查分析。调查结果显示,2004年刺参越冬苗发病比往年早,来势凶猛,持续时间长,流行面广,具较明显的流行病特征,病症异于往年,发病厂家达50%以上,部分地区超过80%,严重地区80%~90%;从患病参体内分离出2株优势细菌和一种新的球状病毒,经负染和超薄切片在电镜下观察,证实有大量球状病毒存于参体内。  相似文献

鲤春病毒血症病毒糖蛋白的截短表达及其免疫原性分析   总被引:1,自引:1,他引:1  
为了揭示鲤春病毒血症病毒(spring viremia of carp virus,SVCV)糖蛋白编码基因的主要免疫原性决定区域,本研究对SVCV的糖蛋白基因进行截短表达,并用纯化的重组蛋白作为免疫原制备了兔抗血清以分析其免疫原性。通过SOSUI以及DNAStar 6.0软件对SVCV糖蛋白基因的跨膜区、亲水性以及抗原表位进行分析后,采用RT-PCR对该基因主要抗原决定区域编码片段进行扩增,并构建重组表达质粒p GEX-Gtr,对其进行诱导表达后获得截短的SVCV糖蛋白的重组蛋白。将表达的重组蛋白进行纯化复性后,作为免疫原制备兔抗血清,采用ELISA法检测其效价,采用免疫印迹以及间接免疫荧光技术检测该重组蛋白的免疫原性。研究结果显示,截短表达的糖蛋白编码基因长1317 bp,编码439个氨基酸(29~467),推测分子量为49.6 k D。利用该重组蛋白制备的兔抗血清,其与重组蛋白的反应效价为1∶64000,。免疫印迹及间接免疫荧光结果显示,该兔抗血清与SVCV-HN株能发生特异性反应,表明该重组蛋白与天然的病毒表面糖蛋白的免疫原性无差异。上述研究结果表明,截短表达的重组蛋白具有很好的免疫原性,可应用于免疫诊断技术研发与基因工程疫苗的研制。  相似文献

Surveillance data on the distribution of viral haemorrhagic septicaemia virus (VHSV) in the North Sea (UK), targeting Atlantic herring in areas with previous virus detection, were obtained from research cruises conducted during 2005. The sensitive molecular approach of real‐time RT‐PCR (qRT‐PCR) was applied alongside a newly developed endogenous positive control assay specific for herring (elongation factor 1α) to ensure integrity of template. Three hundred and five pools from 1937 individual herring were tested, and no evidence of VHSV in association with wild Atlantic herring was detected. Samples were obtained from Scottish waters where marine aquaculture is conducted. The results confirm that previous tissue culture studies have most likely not significantly underestimated the prevalence of carrier herring in this area. The significance of migratory species such as herring as a reservoir species for VHSV, with the potential to translocate virus genotypes between geographical areas, is discussed.  相似文献

根据禽网状内皮组织增生症病毒(REV)长末端重复序列LTR基因保守区域设计并合成一对引物,建立基于SYBR GreenI模式的实时荧光PCR方法(Real-timePCR)。以常规PCR产物为标准品建立标准曲线,并对方法的特异性、敏感性、可重复性以及再现性、检出率进行评价。结果表明,该方法只从REV阳性样本检出扩增信号,不与其它病原出现交叉反应;熔解温度为86.8±0.5℃,无引物二聚体;扩增产物片段大小为270bp,与预期大小相符,测序结果证实为REV靶序列;在1.01×102~1.01×109拷贝之间呈现良好的线性关系,相关系数为0.999,扩增效率为99.7%,最低可检测101拷贝/反应阳性标准品,比常规PCR敏感103倍;组内变异系数、组间变异系数分别为4.80%~5.72%、0.75%~3.87%;对16例临床疑似病例进行检测,阳性率为81.25%(13/16),随机抽取7份阳性扩增产物进行测序,证实为REV序列。本研究建立的SYBR GreenI实时荧光PCR特异性强、通量高、灵敏度高、检测速度快,为REV的快速检测、分子流行病学调查以及监测REV污染疫苗情况提供新方法。  相似文献

【目的】应用酵母双杂交技术构建新城疫病毒的血凝素-神经氨酸酶(HN)蛋白线性中和表位区的诱饵载体pGBKT7-HN-neu,为筛选靶向新城疫病毒HN蛋白的中和性纳米抗体奠定基础。【方法】合成HN蛋白的线性中和表位区HN-neu,将其克隆至酵母双杂交系统诱饵载体pGBKT7中,构建诱饵载体pGBKT7-HN-neu,经酶切鉴定和测序验证正确后,将pGBKT7-HN-neu转化酵母菌Y2H Gold,检测其在酵母细胞中的毒性和自激活作用。【结果】成功合成了HN-neu,构建的pGBKT7-HN-neu在酵母细胞Y2H Gold中无毒性和自激活能力。【结论】成功构建了诱饵载体pGBKT7-HN-neu。  相似文献

为评价不同类型病毒抑制剂在不同发病条件下对烟草花叶病的防控效果,选用2种生物制剂(AHO、宁南霉素)和2种化学农药(吗胍·乙酸铜、烯·羟·硫酸铜)进行田间药效试验.大田接种烟草花叶病毒(TMV)的试验表明,AHO和吗胍·乙酸铜对TMV的平均防效较高,分别为41.8%和39.8%,宁南霉素和烯·羟·硫酸铜对TMV的平均防效较低,分别为25.4%和14.5%;在自然发病状态下,AHO对TMV的平均防效为45.5%,宁南霉素的平均防效次之,为36.6%,而烯·羟·硫酸铜和吗胍·乙酸铜的平均防效则分别为28.7%和16.2%.AHO在不同带毒环境下防控烟草花叶病的效果都优于其他3种市售药物,表明其具有开发成为绿色抗烟草花叶病新型农药的潜力.  相似文献
