Using hydroponics and novel non‐destructive pot culture systems which enable inoculation at specific tuber development stages, the dynamics of common scab infection patterns in potato were studied in order to provide more precise identification of tuber physiological factors associated with susceptibility. At the whole‐tuber level, infection percentages were greatest when Streptomyces scabiei inoculation occurred early; at 2 weeks after tuberization (WAT) 68% of tubers became infected, contrasting with late inoculation (8 WAT), when only 4% infection occurred. The first‐formed internodes were most susceptible to infection, whilst later‐forming and slower‐expanding internodes were less susceptible. Detailed tuber physiological examination of internode 2 showed that pathogen‐induced changes, including increased phellem (periderm) thickness, cell layers and phellem suberization (key physiological features believed critical to S. scabiei infection) were promoted through S. scabiei inoculation. Sequential harvesting showed enhanced phellem suberization (28% greater than the control) within 7 days of pathogen exposure, while phellem thickness and layer responses were also initiated early in the infection process (10–14 days after pathogen exposure) and these responses were independent of symptom expression. Differences in cultivar response were observed, with greater phellem suberization observed 10 days after tuberization (DAT) in the common‐scab‐tolerant cv. Russet Burbank than in the susceptible cv. Desiree. Likewise, Russet Burbank had thicker and more numerous cell layers in the phellem (up to eight cell layers) during early tuber growth (20–30 DAT) than Desiree (up to six cell layers). 相似文献

Biological control of soil-borne pathogens by arbuscular mycorrhizal (AM) fungi has been repeatedly demonstrated. However, their role in the control of above-ground hemibiotrophic pathogens is less conclusive. Here, we investigated in vitro the impact of an AM fungus on Phytophthora infestans in potato plants. The leaf infection index was decreased in mycorrhizal potato plants. Real-Time Quantitative PCR revealed the induction of two pathogenesis related genes (PR1 and PR2) in the leaves of mycorrhizal plants shortly after infection with P. infestans. These results suggested a systemic resistance in mycorrhizal plants, related to the priming of the two PR genes in potato. 相似文献

A dicarboxylic acid polymer (AVAIL) modifies the soil immediately around fertilizer—potentially improving crop phosphorus (P) uptake efficiency and yield. Study objectives were to evaluate potato (Solanum tuberosum L.) response to seasonal applications of liquid and dry AVAIL blended P fertilizer on calcareous soils with low to moderate soil test P. Field experiments conducted 2005–2008 included comparisons of monoammonium phosphate (MAP; 11-52-0) broadcast and/or ammonium polyphosphate (APP; 10-34-0) liquid band applications with or without AVAIL in various fall or spring applications. AVAIL increased US No. 1 yields for selected P rate/source/timing combinations in each of the four years; increased United States No. 1 yields overall in 2006 and 2008; and increased total yield and yield of large (>284g) tubers in 2006. The greatest responses to AVAIL occurred on soils with high lime concentrations. No seasonal advantage was observed in any case. 相似文献

The management of genebank collections of cultivated potato is costly due to the need for in vitro maintenance and virus eradication. Therefore, it is important to set up conservation strategies, which prevent duplicates
entering the collections. In this study, 32 Nordic potato landraces were studied for 57 morphological traits and analysed
for amplified fragment length polymorphism (AFLP). Most landraces could be distinguished based on the morphological characters,
except five accessions. Using five primer combinations to generate 114 reproducible AFLPs, of which 63 (55%) were polymorphic,
the five morphologically indistinguishable accessions were placed into two groups with identical AFLP patterns, suggesting
that some of the accessions were redundant for long-term preservation. The AFLP data showed that the Nordic collection of
potato landraces is composed of genetically different clones, and morphological analysis revealed a wide range of variability.
This variability seems to be distributed randomly over the Nordic region since the cluster analysis based on AFLPs and morphological
traits revealed no grouping based on the country of origin. Principal component analysis suggests that fewer morphological
traits than used in this study will be sufficient to discriminate between different genotypes of cultivated potato (Solanum tuberosum L.). Future possibilities for rationalising potato collections are discussed. 相似文献

A potential for reduced soil macroporosity (below 12% soil volume) under direct drilling, with a concomitant increase in soil relative saturation, is associated with an increase in crown and root rots in Prince Edward Island field crops. Four long-term tillage systems (moldboard plowing, paraplowing-direct drilling, rotary cultivation and direct drilling) were compared in relation to the pathogenic fungal complexes formed in a two crop rotation in spring barley (Hordeum vulgare L.) and soybean (Glycine max L. Merrill) over a 3 year period in a cool humid region of eastern Canada. The principal phytopathogenic fungal complex of Rhizoctonia solani Kühn, Fusarium avenaceum (Fr.) Sacc. and F. oxysporum Schl. remained constant over the treatments. Tillage practice did not affect the number of colony forming units of R. solani in the rhizosphere. The recovery of R. solani from root tissues tended to be lower following conservation tillage and was attributed to antagonism associated with elevated numbers of saprophytic trash microflora concentrated at the soil surface. Disease levels in potato (Solanum tuberosum L.) plantlet bioassays were not influenced significantly by soil source or tillage regime. However, plantlet growth tended to be depressed following transplantation into soil from soybean plots in 1993. Under optimum soil physical conditions conservation tillage did not appear to influence disease levels in barley and soybean rotations. 相似文献

Amplified Fragment Length Polymorphisms (AFLPs) were employed within the taxonomically difficult Solanum nigrum L. complex in order to characterize the genetic diversity present in a collection of the Gatersleben Genebank, to classify taxonomically unknown material, and to correlate the clustering of the examined accessions with their geographic origin. The results from AFLP analyses using two primer combinations on 44 entries from five species led to the detection of four major clusters, simultaneously uncovering significant differences in the levels of genetic diversity within or between species. S. americanumexhibited the highest infraspecific variation despite close geographic origins, simultaneously being placed in a clearly separated cluster in comparison to the other examined species of the complex. In addition, these other species showed even less interspecific variation than was found at the infraspecific level in S. americanum. In terms of taxonomy, the application of AFLPs helped in the classification of 13 black nightshade accessions formerly only listed as Solanum sp. This also was confirmed by morphological determination. Furthermore, one accession formerly classified as S. physalifolium var. nitidibaccatum i) clustered with AFLPs and ii) was identified morphologically as S. villosum. Contradictory classifications remain for two further entries from the same species, found within the S. nigrum cluster after AFLP analyses, while belonging to S. physalifolium var. nitidibaccatum according to herbarium specimen. Finally, as indicated by the information on provenance in geographically separated subclusters in S. americanum and partially in S. villosum, clues on the currently unknown origin of accessions from the genebank seem feasible by AFLP data. 相似文献

The effects of supraoptimal root-zone temperature (30 °C) on growth and development of roots were investigated in water and soil culture with cuttings of two potato clones DTO-2 (heat tolerant) and LT-1 (heat sensitive) at different plant ages, with different compositions of the nutrient solution, and different light intensities. It is suggested that the negative relation between cutting age and heat susceptibility observed in our experiments, is a reflection of the decreasing shoot/root dry weight ratio rather than of age. Heat damage of root systems increased with the concentration of the nutrient solution, nitrate being the main factor responsible. The influence of light intensity on temperature mediated restriction of root growth was relatively small. However the roots of plants grown under low light intensity (60 W m−2 PAR) appeared to be less stressed by supraoptimal temperature, i.e. roots appeared brighter, thinner and more ramified than plants growing at high light intensity (190 W m−2 ). 相似文献

The common potato, Solanum tuberosum spp. tuberosum (tbr), is a tetraploid species with a narrow genetic base, but with a large number of related species that harbor great genetic diversity for numerous characters of agronomic interest. S. kurtzianum (ktz) and S. ruiz-lealii (rzl) are wild diploid relatives with potential resistance to adverse biotic and abiotic factors. To evaluate if this wild germplasm can be incorporated by conventional crosses into the tbr gene pool, pollen-pistil compatibility relations and seed set in interspecific crosses with tbr were investigated. In 4x tbr × 2x ktz crosses and the reciprocals, 89% and 52.2% of genotypic combinations (respectively) were compatible at the pollen-pistil level. Seeds were obtained from some genotypic combinations in both directions of the cross, suggesting that functional 2n gametes might be produced by particular ktz genotypes. In 2x rzl × 4x tbr crosses and their reciprocals, 35% and 11.7% (respectively) of the genotypic combinations were compatible at the pollen-pistil level, but no seeds were obtained. These results indicate that the reproductive isolation between 4x tbr and 2x ktz is incomplete, and that gene exchange between them is feasible. Summary About 13,000 somaclones of 17 cultivars and clones of potato were obtained from in vitro callus cultures and individually
planted in a greenhouse, followed by tuber generations grown in the field. These plants were subjected to the multistage selection
procedure commonly used in potato breeding. Over a period of five years and three field generations the tuber number, size,
shape, eye depth, starch content, starch yield and tuber appearance of these somaclones were assessed and compared with that
of the controls. These characters varied depending on donor genotype and trait. The frequency of variants was assessed and
there were acceptable proportions of desirable abberrants and invariants among the somaclones. Depending on trait the average
gain rate for all donor genotypes ranged between 0.2 and 2.3% for-deviants, between 12.2 and 15.5% for invariants and between
0.1–1.4% for +deviants.
It is concluded that this technique should be used into potato breeding programmes to improve commercially important characteristics
of specific cultivars and breeding clones. 相似文献
