水稻颖花自然开放过程中茉莉酸(JA)生物合成的变化 总被引:1,自引:0,他引:1
外源茉莉酸(JA)及其甲酯(MeJA)对水稻、小麦、黑麦和高粱等禾本科植物颖花开放具有强烈的诱导效应,然而内源JA是否参与颖花开放的调控目前还缺乏充分的证据。本研究应用高效液相色谱-串联质谱(HPLC-MS/MS)系统检测了粳稻武运粳7号和籼稻明恢63颖花自然开放过程中JA水平变化,并以实时定量反转录PCR (real-time RT-PCR)技术分析了武运粳7号颖花开放过程中JA生物合成途径关键基因表达变化。结果发现,水稻颖花JA水平的变化与开颖进程相一致。JA水平在上午颖花开颖前较平稳,开颖时急剧上升至峰值,闭颖后又下降。开颖时JA水平的峰值比开颖前1 h提高4~5倍。与JA水平变化相对应,催化JA生物合成途径关键步骤同工酶的编码基因OsDAD1-3、OsLOX-RCI1、OsAOS1、OsAOC和OsOPR7的表达在开颖时会有不同程度的上调,闭颖后又下调。同一稻穗上正在开放的颖花JA含量及上述JA生物合成途径基因的表达水平也比已开放和未开放的颖花高。颖花开放时JA水平及其生物合成基因表达的上升和闭颖后的下降有力地表明内源JA参与水稻颖花开放的调控。 相似文献

近些年来,水稻免耕抛秧栽培在中国广西等部分地区发展迅速。为了探讨免耕抛栽水稻产量形成特点,以金优253(OryzasativaL.)为试验材料,以常耕抛栽水稻为对照,从叶面积大小、剑叶光合速率与衰老、库的大小及源库比率等方面研究了免耕抛栽水稻的源库特性。结果表明,免耕抛栽水稻早、晚季产量分别为5.97t/hm2和7.53t/hm2,常耕抛栽水稻分别为6.19t/hm2和7.63t/hm2,其产量差异在早季和晚季均不显著。与常耕抛栽水稻相比,免耕抛栽水稻的每穗总粒数和单位面积上的总粒数较多,各生育期的叶面积指数较大,灌浆期剑叶光合速率、蒸腾速率、气孔导度、叶绿素含量较高,而过氧化物酶和过氧化氢酶活性及丙二醛的含量较低,在生理上存在一定的优势。但是,免耕抛栽水稻的源库协调能力较差,粒叶比和结实率下降,致使免耕抛栽水稻在产量上表现出一定的劣势。免耕抛栽水稻粒叶比和结实率的下降是由于其生产的干物质更多地分配在叶片中、贮藏在茎鞘中的干物质在抽穗后向穗部运转的能力较差造成的。 相似文献

Enhanced anther culture efficiency of indica rice (Oryza sativa L.) through modification of the culture media 总被引:1,自引:0,他引:1
The production of microspore-derived green plants from anther culture of indica rice is generally very low compared with japonica cultivars. A modified anther culture medium, consisting of a higher KNO3 content (31 mm ) and casein hydrolysate (CH, 500 mg/1) but without ammonium salts, was tested in comparison with a medium consisting of the widely-used N6 medium nitrogen background, using four indica × indica F1 hybrids as test materials. Green plant regeneration frequency was at least three-fold higher in the microspore-calli derived from the former medium than in those derived from the modified N6 medium. More than 700 microspore-derived plants were raised in the field. Another study was carried out using indica × japonica and indica × javanica F1 hybrids. The results indicated that a medium with higher (3.5 mm ) ammonium sulphate may induce a higher frequency of anthers with microspore-calli but not necessarily lead to a larger number of green-plant regenerating calli. Subsequently, using the indica cv. ‘IR-43’ as the test material, use of a lower level (1.75 mm ) of (NH4)2SO4, in addition to KNO3 (31 mm ), was found to be better than CH (500 mg/l) for anther-response as well as green plant regenerability of the derived microspore-calli. Nitrate-nitrogen or ammonium-nitrogen alone elicited poor response. Twenty-five media involving combinations of KNO3 (20–34 mm ) and (NH4)2SO4 (1–3 mm ) were tested for their effects on anther response. Combinations involving KNO3 (31–34 mM) and (NH4)2SO4 (2.0–2.5 mm ) were found superior not only for achieving greater anther response but also, for subsequent green-plant regeneration. This contrasts with the 28 mm of KNO3 and 3.5 mm of (NH4)2SO4 in the widely-used N6 medium developed for japonica rice. Other than potassium nitrate and ammonium sulphate, and potassium phosphate to some extent, the levels of other inorganic salts tested did not make any significant difference to the process of anther response. Based on these results, modified media with three levels of ammonium sulphate were tested for anther culture efficiency of indica × japonica and indica × javanica derivatives (F3s). Microspore-calli derived from a medium of a lower (1.75 mm ) level of (NH4)2SO4 showed a higher regeneration potential overall than those derived from a higher (3.5 mm ) level. A revised medium has been suggested, on the basis of these results, for the realization of improved anther culture efficiency and, consequently, improved feasibility of using doubled haploids in genetic and breeding research with indica rice. 相似文献

Summary A polymorphic microsatellite locus has been located closely linked to the Waxy gene of rice. Primers were designed to allow detection of the microsatellite by utilising the polymerase chain reaction. 以高抗稻瘟病、直链淀粉含量(AC)较高的三黄占2号和高感稻瘟病、AC较低的丽江新团黑谷衍生的重组自交系群体为研究材料,从性状的相关性和控制两性状的基因在染色体上的位置关系剖析稻瘟病抗性和稻米AC的内在关系。结果表明,两性状间没有显著的相关性。3个与AC相关的QTL分别被定位在第5、6和7染色体上,其加性效应均来自丽江新团黑谷,起降低AC的作用。比较这些QTL与先前对同一群体鉴定的稻瘟病抗性基因(主效基因和QTL)在染色体上的位置,表明控制这两性状的基因上没有紧密连锁关系, 亦没有显著的基因间互作。据此认为,通过亲本的合理选择和分子标记辅助选择可以把三黄占2号稻瘟病持久抗性与理想AC整合到同一品系中,育成优质、抗病的优良品种。 相似文献

Summary Developmental stages during gametogenesis of rice were histologically examined in the period from differentiation of reproductive organs to anthesis. Plants were exposed to acute X-rays of 20 Gy. Radiosensitivity and mutation frequency were investigated in relation to the developmental stages of reproductive organs. The most radiosensitive stage, as measured by reduction of the M1 pollen-and seed-fertilities, was the last premeiotic interphase. Mutations induced at different developmental stages were scored in M3 strains. Sterility mutants and short-culm mutants were most frequently observed. Grain shape, panicle morphology, heading-date and endosperm character mutants were induced at a relatively low frequency. The overall mutation frequency varied with the developmental stage at the time of irradiation. The highest overall mutation frequency was observed when radiation was applied 10 days before anthesis, the late tetrad stage of microspores. Radiation exposure of florets at the late tetrad stage was found to be a more efficient method of inducing a large number of mutations than radiations applied to seeds and fertilized egg cells. 相似文献

Summary There is much interest in the inter-varietal fertility of Asian cultivated rice. In this study, we analyzed the fertility of hybrids in a diallel set of 210 crosses involving 21 parents representing a broad range of the cultivated rice germplasm including landraces, primitive cultivars, modern elite cultivars and parents of hybrid rice. The materials were also carefully selected with respect to indica-japonica classification including typical indica/japonica, more or less intermediate and wide compatibility varieties. The level of hybrid fertility varied widely among the crosses from almost completely sterile to fully fertile. In general, hybrid fertility of intra-subspecific crosses, i.e., indica by indica (I×I) and japonica by japonica (J×J), is much higher than inter-subspecific crosses (I×J or J×I). The fertility varied widely in hybrids involving wide compatibility varieties and also in inter-subspecific crosses not involving wide compatibility varieties. An analysis of variance showed that both the main effects of indica and japonica parents and the interaction between the parents are highly significant in determining hybrid fertility. We speculate that, in addition to wide compatibility, the overall genetic difference between the indica and japonica parents, that may involve a series of minor mutations, play an important role in determining the hybrid fertility. There are also genes of sizable effect that influence fertility in hybrids resulting from specific combinations of the parents.Abbreviations I×I

The Pi-ta gene from indica introgressed into japonica rice has been used to control the blast disease caused by the fungal pathogen
Magnaporthe grisea (Herbert) Barr. (anamorph Pyricularia oryzae Cav.) worldwide. A single nucleotide length polymorphism (SNLP) was identified at the intron region of the Pi-ta gene to develop a codominant Pi-ta gene marker suitable for genotyping with an automated machine. The DNA primer specific to the resistant Pi-taallele was labeled with blue dye (FAM, 6-carboxyfluorescein) as a forward primer, the DNA primer specific to the susceptible
pi-ta allele was labeled with green dye (HEX, 4,7,2′,4′,5′,7′-hexachloro-6-carboxyfluorescein) as another forward primer and the
DNA primer identical to both Pi-ta/pi-ta alleles was unlabeled as the reverse primer for polymerase chain reaction (PCR). Using these three primers, a 181-bp blue
peak in homozygous resistant and a green peak of 182–183 bp in homozygous susceptible, and both peaks in heterozygous plants
were produced by PCR. The utility of marker was verified using a segregating F2 population, inbred cultivated lines, dominant markers and pathogenicity testing. A codominant Pi-ta marker was thus developed for effective Pi-ta assisted selection for crop improvement. Using highly homologous competitive primers for allele detection by PCR can benefit
the study of genome organization of the complex locus.
