以来自湖北省的HB柚柑橘衰退病毒(Citrus tristeza virus,CTV)分离物(CTV-HB1)为材料,利用RT-PCR技术对其CP基因进行克隆,序列分析结果表明:CTV-HB1与典型茎陷点分离物SY568和NUagA的核苷酸和推定氨基酸的序列相似性均在97%以上,而与CTV速衰分离物T36和弱毒分离物T385和T30的核苷酸和氨基酸序列相似性较低,均在95%以下。将CP基因片段连接到表达载体,转化大肠杆菌后诱导重组蛋白的表达。SDS-PAGE和Westen-blot分析结果表明,经IPTG诱导后产生了预期大小的重组蛋白,该蛋白可与CTV-L5提纯病毒制备的多克隆抗体发生特异性免疫反应。 相似文献

对干旱胁迫下3a生复叶槭的挥发物进行了鉴定。当复叶槭受到干旱胁迫时,9种化合物释放量升高(包括:丁醇、戊醇、反—2—己烯—1—醛、顺—3—己烯—1—醇、戊醛、戊酸、己醛、己酸、苯乙酮),而长叶烯释放量降低:复叶槭复水后苯乙酮升高,但其他挥发物释放量降低。通过GC—EAD检测了光肩星天牛对其中某些化合物的触角电位反应,并使用诱捕器和人工合成单体进行了田间生测实验。丁醇、戊醇和顺—3—己烯—1—醇能引起光肩星天牛的触角反应,这一结果在田间实验中得到了证实。这表明干旱胁迫在光肩星天牛选择寄主植物方面起着重要作用。 相似文献

This experiment was conducted to study the effect of different levels of wheat roti (WR) on nutrient utilization and blood metabolites in Asian elephants fed roughages ad libitum. Nine (3 M, 6 F) Asian elephants (14–52 years of age, 1909–3968 kg BW) were used in an experiment based on replicated Latin square design. Animals in each group (n = 3) were assigned to one of the three dietary treatments in a manner that animals in all the three groups were exposed to all the three treatments in three different periods. Each feeding trial comprised 30 days (25 days of adaptation and 5 days collection period). The amount of WR fed to the elephants was 0.18, 0.12 and 0.06% of BW in groups I, II and III, respectively. They were allowed to forage in the nearby forests for 6 h/day and to bathe for 2 h/day. The animals had ad libitum access to cut Rohini (Mallotus philippensis) trees in their night shelter. Intake and apparent digestibility of dry matter (DM), crude protein (CP), gross energy (GE), Ca, P, Fe, Cu and Zn were measured. Feed consumption was not significantly different among the groups. Significant (p < 0.01) decrease in digestibility of DM and GE and blood glucose concentration was observed with decreased level of WR in the diet. Feeding of WR at 0.06% of BW supplied adequate amount of DE, CP, Ca, P, Fe, Cu and Zn to meet requirement for adult maintenance. Feeding of WR in excess of 0.06% of BW supplied DE in excess of requirement, increased blood glucose concentration which may cause obesity and other associated health problems. 【目的】 研究不同氮素含量人工饲料对亚洲玉米螟Ostrinia furnacalis(Guenée)生长发育和繁殖的影响,分析玉米螟种群激增的内在N素营养驱动。【方法】 测定5个氮素含量水平(3.94%、7.33%、10.69%、18.84%、30.31%)人工饲料对亚洲玉米螟个体生长发育及种群增殖的影响,构建实验种群生命表。【结果】 25℃,低氮水平N1(氮素含量3.94%)幼虫发育缓慢且未能正常羽化,当氮素含量增加到18.84%(N4)时,幼虫历期明显缩短,内禀增长率(0.13)、净增殖率(66.14)、周限增长率(1.14)、孵化率(90.06%)最高,平均世代历期(30.79)、种群加倍时间(5.17)最短,并与其他4个氮素水平有显著差异(P<0.05),且其化蛹率(98.66%)、羽化率(95.45%)、存活率(91.33%)、平均单雌产卵量(120.12±9.73)最高;但当氮素含量提高到30.31%后,内禀增长率(0.04)、净增殖率(44.71)、周限增长率(1.04)最低,平均世代历期(92.36)、种群加倍时间(16.85)最长,25和30℃结果与20℃接近。3个温度各处理蛹期、成蛾寿命、雌虫生殖力无显著差异(P>0.05)。【结论】 氮素含量对亚洲玉米螟个体发育与种群增殖关系密切,在一定区间内随氮含量增加生长加快,种群增殖能力增强,且高氮30.31%和低氮3.94%均对玉米螟生长有明显抑制作用。 相似文献

土地利用/土地覆被变化模拟是土地科学的重要内容。随着全球变化研究的不断深入,LUCC变化模拟已逐步深入到农作物空间。开展长时序农作物空间变化模拟有助于揭示农业生产过程中“人类-自然”综合体的复杂关系。然而目前农作物空间模拟研究较少,已有模型在空间映射关系,对象识别等尚存技术局限,模型应用也主要集中在粮食作物。本文构建了基于适宜概率的柑橘生产空间分配模型,对四川省柑橘生产空间分布进行了模拟研究。结果显示:该模型较好地反映了1980—2015年柑橘统计特征,各时期统计面积和模拟面积的相对误差均<25%。在县域尺度上,统计面积和模拟面积的相关系数在0.987 6~0.999 9,呈现明显的线性相关,达到了极显著相关。1980—2015年间,四川省柑橘空间快速扩张,柑橘模拟面积的时序变化与统计面积一致。柑橘空间集中在川中丘陵区和成都平原区。空间格局从分散零星向区域集聚转变,大致形成了成都平原、川南、川东北3个柑橘集中区。SPAM-Citrus模型误差受空间分辨率、空间分配规则、土地覆被、作物分布点的共同影响。在应用中应尽量选择种植年限较长的采样点,同时面向研究对象和区域,选择合适分辨率和空间分配规则,提高预测结果的准确性。 相似文献

In order to determine the immunomodulatory effect of dietary levamisole in Asian catfish (Clarias batrachus), fish were fed four different diets for 10 days: a formulated diet as control and the same diet supplemented with 50, 150 or 450 mg levamisole kg?1 feed. The serum bacterial agglutination titre against Aeromonas hydrophila as a measure of specific immunity, serum haemagglutination titre, natural haemolytic complement activity (ACH50), myeloperoxidase and lysozyme activities, total protein level and oxidative radical production by neutrophils as a measure of non‐specific immunity as well as disease resistance against A. hydrophila challenge to separate vaccinated and non‐vaccinated groups were evaluated at 0, 1, 2 and 3 weeks after last administration of levamisole. Levamisole supplement at the lowest level (50 mg kg?1) significantly enhanced oxidative radical production and serum myeloperoxidase (MPO) content immediately after 10 days of feeding, which reached peak values after 3 and 2 weeks of feeding respectively. Haemolytic complement and haemagglutination titre were significantly enhanced after 3 and 1 weeks respectively. Haemolytic complement activity and MPO activities were significantly raised to 150 mg kg?1 after 3 and 2 weeks, respectively. At the highest level of levamisole feeding (450 mg kg?1) significant decreases in superoxide production and complement activity were measured immediately after levamisole feeding, which returned to the normal level after 1 week post‐ feeding. Fish were challenged with a virulent strain of A. hydrophila at 0, 1, 2 and 3 weeks after levamisole feeding, and the cumulative per cent survival was recorded over 10 days. Feeding levamisole at 50, 150 or 450 mg kg?1 increased per cent survival in vaccinated fish immediately after levamisole feeding, and survival was significantly higher at 450 mg kg?1. There was no difference in mortality patterns in non‐vaccinated fish. The results support the use of levamisole at 50 mg kg?1 feed for 10 days as an immunostimulant in Asian catfish farming. 相似文献

Four African wild strains (Egypt, Ghana, Senegal and Kenya) and four established Asian farmed strains of Nile tilapia, Oreochromis niloticus (popularly known in the Philippines as Taiwan, Thailand, Singapore and Israel) were analysed electrophoretically at 30 protein loci to estimate genetic differences among the strains. Genetic distance values among the strains revealed a clustering of the farmed strains with Egypt and Ghana O. niloticus, a slight separation of the Senegal strain and a larger separation of the Kenya strain. This profile may reflect the origins of the few founder populations of this species previously introduced to Asia. It also confirms the wider genetic divergence of the Kenya strain (O. niloticus vulcani) from the others studied here, which are all O. n. niloticus. Observed heterozygosities of the strains ranged from 0.026 to 0.071, with the African wild strains the lower values (mean Ho = 0.036) and the farmed strains the higher ones (mean Ho = 0.056). The implications of these results to the ongoing tilapia genetic improvement programme in the Philippines are discussed. 相似文献

Asian catfish, Clarias batrachus, were fed semi-purified basaldiets containing 0, 0.1, 0.5, 1, 3 and 5 mg biotin kg–1diet for 60 days. Fish fed the control diet (no biotin) showed(P < 0.05) higher mortality, lower weight gain, specificgrowth rate (SGR), feed efficiency ratio (FER) and protein efficiencyratio (PER) than in fish fed diets supplemented with biotin. The highestweight gain, SGR, FER and PER were noticed in fish fed 1 mg biotinkg–1, followed by 0.5, 5, 3 and 0.1 mg biotinkg–1, except for PER (followed by 0.5, 5, 0.1 and 3 mgbiotin kg–1). Quadratic analysis showed that the optimumdietary biotin requirements for maximal weight gain, PER and PER were2.49, 2.54 and 2.52 mg kg–1, respectively. Liver biotinconcentrations were influenced by levels of biotin in the diet.Concentration of liver biotin increased as level of dietarysupplementation increased and no biotin was detected in the liver of thecontrol fish. Liver pyruvate carboxylase and acetyl CoA carboxylaseactivities were higher in fish fed biotin-supplemented diets than incontrols. Biotin concentrations, pyruvate carboxylase and acetyl CoAcarboxylase activities in liver associated with normal growth rangedfrom 10.59 to 10.66 g g–1, 147.97 to 148.18 units mgprotein–1 and 12.76 to 12.78 units mg protein–1, respectively. Biotin deficiency symptoms such as anorexia, darkskin colour and convulsions were observed in fish fed the control diet.The optimum dietary biotin requirement for maximal growth of C.batrachus is about 2.49 mg kg–1 diet. 相似文献
