The inoculum sources of ascospores of Pleospora allii and of conidia of its anamorph Stemphylium vesicarium were investigated in relation to the brown spot disease epidemiology on pear. Dead and living leaves of three pear varieties (Abate Fétel, Conference and William), seven grasses (Poa pratensis, Festuca rubra, Festuca ovina, Lolium perenne, Digitaria sanguinalis and Setaria glauca) and Trifolium repens, which are used in pear orchard lawns, were inoculated with conidia of Stemphylium vesicarium virulent on pear and incubated under controlled-environment. Stemphylium vesicarium was always re-isolated from dead leaves of the considered plants, but not from symptomless green or yellowish living leaves. The fungus was occasionally re-isolated from leaf segments showing unspecific necrosis. Inoculation of pear leaves with isolates from grasses demonstrated that the fungus did not lose pathogenicity. Pseudothecia, ascospores and conidia were produced on all the dead inoculated leaves; differences between specimens were found for phenology of pseudothecia, their density and size, and for the number of conidia produced. Pseudothecia were produced faster in the lawn species than in pear leaves, and their density was higher, especially for S. glauca, L. perenne and P. pratensis. Ascospore maturation and ejection was more concentrated for the pseudothecia developed on pear leaves than for those on F. ovina and S. glauca. All the lawn species produced more conidia than pear leaves.  相似文献

We performed a 3-year field survey in France to characterize the dynamics of sexual reproduction in Mycosphaerella linicola, the causal agent of pasmo, during the interepidemic period. Cohorts of fruiting bodies were sampled from linseed straw during the autumn and winter and carefully observed, focusing on pseudothecia, asci, and ascospores. A sequence of experimental steps corresponding to Koch's postulates confirmed in July 2014, for the first time in France and continental Europe, the widespread presence of the sexual stage of M. linicola in plant host tissues. The developmental dynamics of pseudothecia on straw, expressed as the change over time in the percentage of mature pseudothecia, was similar in all three years. Pseudothecia appeared in late summer, with peak maturity reached in October. A temporal shift, thought to be due to early autumn rainfall, was highlighted in one of the three years. These observations suggest that sexual reproduction plays a significant role in the epidemiology of pasmo in France. A resurgence of M. linicola infections in spring flax is thought to have occurred in recent years, due to the increase in the area under flax. The presence of the sexual stage of this pathogen probably increased the quantitative impact of residues of winter linseed (used for oil) and flax straw (left on the soil for retting and used for fibres) as an interepidemic “brown bridge”. This case study highlights how certain parts of a disease cycle, in this case the sexual phase, can become crucial due to changes in production conditions.  相似文献

Because epidemics of successive cropping seasons are not independent, epidemiological studies need to encompass the processes occurring during the transmission of epidemics from one season to the next. With Leptosphaeria maculans, infected stubble allows carry‐over of the fungus. Generation experiments using recurrent selection on field plots are a useful means of comparing the effects of selection pressures. However, the full life cycle of the fungus, from plant infection to the next generation of ascospores, has not yet been achieved under controlled conditions. Studies were undertaken to achieve an experimental set‐up with sexual reproduction under controlled conditions. Cankered oilseed rape stems were produced under controlled conditions, after inoculation with a mixture of 12 isolates across both mating types. Stems were cut longitudinally and attached to styropore plates. Stem halves were incubated outside or in climate chambers regularly soaked in tap water to ensure maturation. Incubation was stopped when mature pseudothecia were observed. In all three independent experiments, more stem halves had pseudothecia when incubated under controlled conditions (30–100%) than incubated outside (0–80%). To the authors’ knowledge, this is the first study achieving the full life cycle of the fungus under controlled conditions, from infection of the plant to mature pseudothecia. This opens up the prospect of running experiments year‐round to better understand inoculum production, to compare fungal fitness, or to run generation experiments with exotic pathogen populations.  相似文献
