A potyvirus, for which the name Japanese hornwort mosaic virus (JHMV) is proposed, was isolated from Japanese hornwort plants
(Cryptotaenia japonica) with mosaic disease symptoms. The virus was used to inoculate mechanically 34 plants belonging to 33 species of 10 families.
Of these species seven from two families were infected. Faint chlorotic spots appeared on the inoculated leaves of Chenopodium quinoa and C. amaranticolor, but no systemic infection occurred in these plants. JHMV systemically infected only Umbelliferae plants; they did not infect
26 other species in eight families. JHMV was transmitted in a nonpersistent manner by aphids (Myzus persicae). The virus was a flexuous rod-shaped particle about 750 nm in length. Sequencing the nucleotides in the 3′ terminal region
of JHMV revealed that the coat protein contains 280 amino acids with a molecular mass of 32.2 kDa. The nucleotide sequence
of the coat protein of JHMV had the highest similarity with that of Zantedeschia mosaic virus (83.3%) compared to those of
other potyviruses (57.0%–64.9%). An antiserum against JHMV reacted strongly with JHMV and weakly with Potato virus Y. These results indicate that JHMV is a new potyvirus.

To identify possible sites of viral attenuation, the complete nucleotide sequences of two isolates of Zucchini yellow mosaic virus (ZYMV) were determined; a severe isolate Z5-1 and an attenuated isolate from Z5-1 (designated ZYMV-2002). The viral genome
of both isolates consisted of 9593 nucleotides in size and contained an open reading frame encoding a single polyprotein of
3080 amino acids. Comparison of the nucleotide sequences for Z5-1 and ZYMV-2002 revealed 14 nucleotide mutations, resulting
in seven amino acid substitutions with four in the HC-Pro region, two in the CI region, and one in the NIb region. These results
provide a genetic basis for future manipulation of the ZYMV reverse genetics system.

Wheat streak mosaic virus, vectored by the wheat curl mite Aceria tosichella Keifer, is a major limiting factor in wheat production in the Texas Panhandle. It is the most frequently encountered virus in the region, affecting both shoot and root biomass, and consequently it can drastically reduce both forage and grain yield, and water-use efficiency of the plant. In light of the continuing decline of water levels in the Ogallala Aquifer, its effect on water-use efficiency has significant implications for irrigated fields, especially with regard to energy costs for pumping water. This article estimates the potential economic losses from the virus infection. To achieve this objective, partial budgeting techniques, based primarily on data from field experiments, were used. Results indicate that Wheat streak mosaic virus causes grain and forage yield losses that significantly affect profits from wheat production. In addition, irrigated wheat producers are more negatively affected by the disease due to the economic losses from reduced water-use efficiency of infected plants. The economic insights from this article provide important implications for wheat management and research priority-setting. 相似文献

Improved tobacco cultivars introgressed with alleles of the recessive resistance va gene have been widely deployed in France to limit agronomical consequences associated with Potato virus Y (PVY) infections. Unfortunately, necrotic symptoms associated with PVY have been reported on these cultivars suggesting that PVY is able to overcome the resistance. A field survey was performed in France in 2007 to (i) estimate the prevalence of PVY in tobacco plants showing symptoms and (ii) characterize PVY isolates present in susceptible and va‐derived tobacco cultivars. A serological typing procedure, applied to 556 leaves collected from different French tobacco growing areas, was performed using polyclonal antisera raised against different viral species including PVY. Viral species were detected in 80·8% of leaves and PVY was present in 83·5% of infected samples. However, statistical analysis confirmed that the probability of a tobacco plant being infected with PVY is reduced in va hosts. Eighty‐six PVY isolates were mechanically inoculated on one susceptible and three va‐derived tobacco cultivars used as indicator hosts to define virulence of these isolates against alleles 0, 1 and 2 of the va gene. Both qualitative and quantitative analyses showed that 55 PVY isolates were able to overcome the three va alleles. Moreover, the monitored biological diversity of PVY isolates was higher in the susceptible tobacco hosts than in the va‐derived ones. This study helps to understand consequences of the deployment of the va gene in tobacco on diversity and virulence of PVY isolates. 相似文献

Potato virus Y (PVY) is the potato virus with the highest economic impact on seed potato production. Insecticides are efficient in controlling aphids, which are the vectors of this virus, but rarely limit virus spread in the field. Straw mulching and mineral oil spraying are known as alternatives to insecticides to reduce PVY incidence, but important year‐to‐year variation in efficacy has been observed with both of these techniques. Preliminary studies revealed the efficacy of intercropping in controlling PVY spread, but more data are needed to validate this observation. A four‐year field trial was conducted in Switzerland to assess the potential synergistic effect of combining mineral oil spraying with straw mulching to increase the protection of seed potato crops against PVY spread. Furthermore, the efficacy of intercropping with oat and hairy vetch was examined as a novel way to control in‐field PVY spread. The present work demonstrates that the modes of action of mineral oil and straw mulching are complementary and reduce the year‐to‐year variation observed with oil and straw when used alone as PVY control agents. The results also demonstrate the efficacy of intercropping for the control of PVY, and the mode of action of this novel control method is discussed. Overall, this work shows that it is possible to increase the protection of potato fields against PVY spread by combining control strategies with different modes of action that complement each other, such as mulching, oil spraying and intercropping. 相似文献

To investigate the role of environmental conditions on the selection of virulent Potato virus Y (PVY) isolates subject to pressure from the recessive resistance gene va in tobacco, a field survey was performed in Brazil where va‐derived genotypes have been recently introduced and now represent less than one‐third of cultivated tobacco genotypes. A serological analysis of 397 leaves collected from different Brazilian tobacco‐growing areas and mainly from plants with symptoms indicated that 52·4% of samples were infected by at least one of the viral species tested. PVY was present in 72·1% of infected samples. The probability of a plant being infected with PVY was reduced in va hosts. However, the biological characterization of PVY isolates on indicator hosts showed that 20 of the 29 tested isolates were able to overcome the alleles of the va gene. Moreover, the observed biological diversity of isolates was higher in susceptible tobacco genotypes than in va‐resistant ones. Comparison of these data with the PVY diversity in French tobacco fields shows that the use of va‐derived genotypes in two environments with contrasting climatic conditions, local hosts and cultural contexts, leads to a similar outcome: the prevalence of virulent isolates. These results strongly suggest an important role of the va gene in the modification of PVY populations. 相似文献
