Four components of partial resistance toPhytophthora infestans were measured after inoculation in the greenhouse and in the field ofSolanum arnezii x hondelmannii, S. berthaultii, S. circaeifolium, S. leptophyes, S. microdontum, S. sparsipilum, S. sucrense andS. vernei, and four hybrid progenies ofS. microdontum withS. tuberosum. The four components were infection efficiency, lesion growth rate, generation time and sporulation capacity. The results were compared with resistance ratings derived from field experiments, and with observations made on the potato cultivars Bintje, Bildstar, Libertas and Pimpernel. Genetic variation for all components was found, while the relative importance of the components of partial resistance appeared to vary between the species. InS. microdontum, generation time, infection efficiency and lesion growth rate, and inS. tuberosum infection efficiency, lesion growth rate and sporulation capacity appeared positively associated, but in other species no such association was found. A strong hypersensitive reaction, the expression of which appeared to depend on environmental conditions, was found inS. microdontum. The effects of exposure period and phosphine concentration on mortality of susceptible and resistant Sitophilus oryzae (L) were investigated. Although S oryzae is one of the world's most serious pests of stored grain there are few data on the practical significance of phosphine resistance in this species. The strains investigated were an Australian susceptible strain, a homozygous resistant strain exhibiting a level of resistance common in Australia and an unselected field strain from China with a much stronger resistance. Fumigations were carried out at 25 degrees C on adults and mixed-age cultures. For adults of all three strains and mixed-age cultures of the susceptible and resistant Australian strains, the relationship between concentration and time could be described by equations of the form Cnt = k. In all cases n < 1, indicating that time was a more important variable than concentration. In all fumigations of adults the resistant strains were harder to kill than the susceptible strain. However, in fumigations of mixed-age cultures, which contained the tolerant pupal stage, the difference between susceptible and resistant strains was more pronounced at lower concentrations than higher concentrations. For example, at 0.02 mg litre-1 the estimated LT99.9 for mixed-age cultures of the Australian resistant strain (27 days) is 3.4 times that of the susceptible strain (8 days), but at 1 mg litre-1 there is no difference between the two strains (4 days). Limited data on the Chinese resistant strain supported this finding. Twenty-three days exposure at 0.02 mg litre-1 had no effect on mixed-age cultures of this strain, but there were no survivors after 5 days exposure to 1 mg litre-1. 相似文献

以苜蓿为原料,采用酸碱法沉淀叶蛋白凝聚物。冷冻干燥后,分析叶蛋白中常规营养成分的含量和氨基酸组成,以及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶的含量;得到的苜蓿叶蛋白分别在常温、4℃、-20℃、-40℃条件下储藏,在2周、4周、8周、12周时取样,分析超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶活性在储藏过程中的变化。结果表明:苜蓿叶蛋白中常规营养成分的含量远高于日常食品,各种氨基酸种类齐全;苜蓿叶蛋白中含有丰富的SOD和谷胱甘肽过氧化物酶;低温对SOD和谷胱甘肽过氧化物酶的酶活性保存率保存效果最好。 相似文献

Disease resistance mediated by the resistance gene Xa21 is developmentally controlled in rice. We examined the relationship between Pathogenesis Related (PR) defense gene expression and Xa21-mediated developmental disease resistance induced by Xanthomonas oryzae pv. oryzae (Xoo). OsPR1a, OsPR1b, and OsPR1c genes were cloned and their induction was analyzed, in addition to the OsPR10a gene, at the juvenile and adult stages in response to a wildtype Xoo strain that induces a resistance response (incompatible interaction) and an isogenic mutant Xoo strain that does not (compatible interaction). We found that the adult stage leaves are more competent to express these OsPR1 genes and that the Xa21 locus is required for the highest levels of induction. 相似文献

The aim of this study was to compare the viability of 7- and 8-day-old equine embryos cooled and stored for 6 or 24 hours in two different transport systems. Embryos (n = 97) were recovered on day 7 or 8 and assigned to 10 groups (n = 10/group). Embryos within the same age group (D7 or D8) were evaluated immediately after collection (Group-0h) or after storage in an Equitainer at 5°C for 24 hours in 5 ml Emcare Holding Solution (EHS) (Group-E-24h) or 5 ml Ham's F10 (Group-H-24h) or in a refrigerator at 5°C in 500 ml Emcare Flushing Solution (EFS) for 6 hours (Group-B-6h) or 24 hours (Group-B-24h). After collection or storage, embryos were incubated in 1 μg/ml DAPI to determine the percentage of dead cells per embryo (DAPI positive, fluorescent cells). Subsequently, embryos were fixed in 4% paraformaldehyde and re-stained with DAPI to determine the total number of cells. The percentage of dead cells in group-0h and B-6h was similar and significantly lower than for embryos stored for 24 hours in groups B-24h, E-24h, and H-24h. The percentage of dead cells was similar for embryos stored in an Equitainer (groups E-24h and H-24h) and was significantly higher for embryos stored 24 hours in EFS (Group B-24h). Within each storage system (0h, B-6h, B-24h, E-24h, and H-24h) no significant difference in the percentage of dead cells was observed between 7- and 8-day-old embryos. Storage in 500 ml EFS at 5°C for 6 hours resulted in embryos of better quality than after the traditional 24-hour storage in an Equitainer, suggesting that this simplified system offers a good alternative for short-term storage and transport. 相似文献
