Objective

To evaluate three volumes of lidocaine for spermatic cord block to perform castration in cattle.

Study design

Randomized blinded clinical study.

Animals

Thirty mixed-breed Nellore cattle, aged 28–40 months and weighing 395 ± 21 (352–452) kg [mean ± standard deviation (range)].

Methods

Cattle were restrained in a chute and allowed to stand without sedation. Three milliliters of 2% lidocaine without epinephrine were infiltrated subcutaneously at each site of scrotal incision in all animals. The animals were allocated to three groups of 10 animals each. Lidocaine 2% was injected into each spermatic cord using a volume of 2, 3 or 4 mL in groups A, B, or C, respectively. The total volumes of lidocaine used were 10, 12, and 14 mL in groups A, B, and C, respectively. The duration of surgery and the retraction of the testicle (scored as positive or negative according to retraction of the testicle) during the procedure were recorded. The data were statistically analyzed by one-way anova followed by Tukey’s and chi-square tests. Differences were considered significant when p < 0.05.

Results

The mean surgical time was shorter in group C than in groups A and B (p < 0.001). In groups A, B and C, 90%, 60% and 10% of the animals showed retraction of the testicle, respectively. Fewer animals retracted the spermatic cord in group C than in group A (p = 0.002) and B (p = 0.02).

Conclusions and clinical relevance

Optimal spermatic cord block was achieved by injection of 4 mL of 2% lidocaine 5 minutes before castration and following incisional infiltration of lidocaine, in adult cattle weighing about 400 kg.  相似文献

Many fillies and mares are accused of behaving badly by their owners or trainers, and their reproductive hormones and ovaries are commonly blamed for this. Overt oestrus behaviour, however natural, is undesirable when a horse is being ridden, trained and competed. More subtle behaviours may be attributed to the reproductive cycle and presented as the cause of poor performance in the elite competition mare. Sometimes behaviours are complex, perhaps at odds with what one expects for normal reproductive behaviour and in many cases may not be associated with the reproductive cycle at all, rather associated with pain of musculoskeletal or soft tissue origin. The situation represents a diagnostic dilemma to positively associate undesirable behaviours with the ovarian hormones or ovarian pain, before an attempt can be made to modify the behaviour through the suppression of oestrus. This article will review the reasons for presentation, the behavioural patterns of the reproductive cycle and the diagnosis of reproductive behavioural problems. The range of techniques for oestrus suppression will be reviewed along with their clinical application and any known current regulatory issues associated with their use.  相似文献

Leucettamols, bifunctionalized sphingoid-like compounds obtained from a marine sponge Leucetta sp., act as non-electrophilic activators of the TRPA1 channel and potent inhibitors of the icilin-mediated activation of the TRPM8 channel, while they are inactive on CB1, CB2 and TRPV1 receptors. Leucettamols represent the first compounds of marine origin to target TRPA1 and the first class of natural products to inhibit TRPM8 channels. The preparation of a small series of semi-synthetic derivatives revealed interesting details on the structure-activity relationships within this new chemotype of simple acyclic TRP modulators.  相似文献

Objective   To determine the effectiveness of a topical anaesthetic formulation (Tri-Solfen) with or without the administration of a non-steroidal anti-inflammatory drug (carprofen) on the pain and distress response associated with ring or surgical castration of ram lambs.
Procedures   Merino ram lambs (n = 78) were allocated to 10 treatment groups: 4 groups of knife-castrated lambs and 4 groups of ring-castrated lambs received carprofen (4 mg/kg SC) and Tri-Solfen; 2 control groups (sham) received carprofen at 0 or 4 mg/kg SC. Measurements included plasma cortisol and haptoglobin concentrations, haematology, and behaviour, including posture.
Results   Knife-castrated lambs had higher peak cortisol and integrated cortisol responses for the first 6 h after treatment and greater concentration s of circulating acute phase proteins than ring-castrated lambs, both of which were significantly different from the sham controls. Tri-Solfen applied to the knife castration wound significantly reduced both the peak plasma cortisol concentration and the integrated cortisol response for the first 6 h and improved lying behaviour in the first 12 h. Carprofen reduced the cortisol response to knife castration at 30 min, but elevated the cortisol responses at 24 and 48 h. Carprofen nearly halved the number of acute pain behaviours associated with ring castration. There were no significant additive or synergistic effects from combining the analgesic treatments. Tri-Solfen applied to the tail wound provided no detectible benefits during ring castration + tail docking.
Conclusions   The physiological and behavioural responses suggest that ring castration has less impact on the lamb than knife castration. The specific analgesic treatments can provide modest amelioration of the pain and discomfort associated with castration. Alternative doses or application methods may enhance their efficacy.  相似文献

Conus regius is a marine venomous mollusk of the Conus genus that captures its prey by injecting a rich cocktail of bioactive disulfide bond rich peptides called conotoxins. These peptides selectively target a broad range of ion channels, membrane receptors, transporters, and enzymes, making them valuable pharmacological tools and potential drug leads. C. regius-derived conotoxins are particularly attractive due to their marked potency and selectivity against specific nicotinic acetylcholine receptor subtypes, whose signalling is involved in pain, cognitive disorders, drug addiction, and cancer. However, the species-specific differences in sensitivity and the low stability and bioavailability of these conotoxins limit their clinical development as novel therapeutic agents for these disorders. Here, we give an overview of the main pharmacological features of the C. regius-derived conotoxins described so far, focusing on the molecular mechanisms underlying their potential therapeutic effects. Additionally, we describe adoptable chemical engineering solutions to improve their pharmacological properties for future potential clinical translation.  相似文献

Background

Chiari‐like malformation (CM) and syringomyelia (SM) cause a pain syndrome in Cavalier King Charles spaniels (CKCS). Clinical signs are not consistently apparent on neurologic examination, and owner reporting of signs provides vital clinical history. However, owner questionnaires for this disease are not well developed.

Objectives

To develop a tool to capture owner‐reported clinical signs for use in clinical trials and to compare owner‐reported signs with the presence of pain on neurologic examination and SM on magnetic resonance imaging (MRI).

Animals

Fifty client‐owned CKCS.

Methods

Owners completed a questionnaire and pain/scratch map. Each dog underwent a neurologic examination and craniocervical magnetic resonance imaging (MRI). Questionnaire responses were developed into scores, area of shading for pain/scratch maps was measured, and consistency of responses between these tools was assessed. Owner‐reported findings were compared with neurologic examination findings and presence and severity of SM on MRI.

Results

Thirty‐three dogs were symptomatic and 17 asymptomatic; 30 had SM. The most common sign of pain was crying out when lifted (n = 11). Extent of shaded areas on maps positively correlated with questionnaire scores for pain (r2 = 0.213, = 0.006) and scratch (r2 = 0.104, = 0.089). Owner‐reported findings were not significantly associated with presence or severity of SM or neurologic examination findings. Owner‐reported lateralization of signs was significantly associated with SM lateralization (< 0.0001).

Conclusions

The questionnaire and maps may be useful for clinical trials. Lack of association of owner‐reported signs with SM highlights our lack of understanding of the pathophysiology of pain in this disease.  相似文献

ObjectiveTo investigate a topical local anesthesia technique as a means to prevent and/or diminish pain in mice in a laboratory setting associated with tail vein injections performed by personnel in training.Study designProspective, randomized experimental trial.AnimalsThirty six adult female, 23–28 g CD-1 mice from an in-house training colony. They were acclimated to routine training and handling classes.MethodsEutectic mixture of local anesthetics (EMLA) cream (2.5% lidocaine/2.5% prilocaine) or a bland ointment control (n = 18) was applied on the tail prior to intravenous injection. The injections were performed by novices, who had never attempted the procedure, and experienced personnel. All participants were blinded to treatment groups. Three injection attempts were allowed per animal. The mice were observed and scored by blinded evaluators for behavioral and physiological changes, including respiratory rate, vocalization, tail flick, and escape behaviors, during and after the injection.ResultsThis study demonstrates that aversive behaviors induced by lateral tail vein injection were not changed by the preemptive application of EMLA cream. The aversive behaviors associated with lateral tail vein injection were significantly affected by the number of injection attempts and the individual's experience level.Conclusions and clinical relevanceTopical EMLA cream did not reduce signs of aversive reaction to tail vein injection and thus we did not find support for its use in mouse training programs for tail vein injections.  相似文献

ObjectiveTo evaluate the analgesic and physiological effects of epidural morphine administered at the sixth and seventh lumbar or the fifth and sixth thoracic vertebrae in dogs undergoing thoracotomy.Study designProspective, randomized, blinded trial.AnimalsFourteen mixed-breed dogs, weighing 8.6 ± 1.4 kg.MethodsThe animals received acepromazine (0.1 mg kg?1) IM and anesthesia was induced with propofol (4 mg kg?1) IV. The lumbosacral space was punctured and an epidural catheter was inserted up to the region between the sixth and seventh lumbar vertebrae (L, n = 6) or up to the fifth or sixth intercostal space (T, n = 8). The dogs were allowed to recover and after radiographic confirmation of correct catheter position, anesthesia was reinduced with propofol IV and maintained with 1.7% isoflurane. Following stabilization of monitored parameters, animals received morphine (0.1 mg kg?1) diluted in 0.9% NaCl to a final volume of 0.25 mL kg?1 via the epidural catheter, and after 40 minutes, thoracotomy was initiated. Heart rate and rhythm, systolic, mean and diastolic arterial pressures, respiratory rate, arterial hemoglobin oxygen saturation, partial pressure of expired CO2 and body temperature were measured immediately before the epidural administration of morphine (0 minute) and every 10 minutes during the anesthetic period. The Melbourne pain scale and the visual analog scale were used to assess post-operative pain. The evaluation began 3 hours after the epidural administration of morphine and occurred each hour until rescue analgesia.ResultsThere were no important variations in the physiological parameters during the anesthetic period. The post-operative analgesic period differed between the groups, being longer in T (9.9 ± 1.6 hours) compared with L (5.8 ± 0.8 hours).ConclusionsThe use of morphine, at a volume of 0.25 mL kg?1, administered epidurally over the thoracic vertebrae provided longer lasting analgesia than when deposited over the lumbar vertebrae.Clinical relevanceThe deposition of epidural morphine provided longer lasting analgesia when administered near to the innervation of the injured tissue without increasing side effects.  相似文献

ObjectiveTo investigate the relationship between anxiety and pain scores using the Glasgow Composite Measure Pain Scale–Short Form (CMPS-SF) in dogs.StudyProspective observational study.AnimalsA group of 18 dogs undergoing surgical management of stifle disease.MethodsPreoperatively dogs were scored using the CMPS-SF, the anxiety behaviour-based Reactivity Evaluation Form (REF), a Visual Analogue Scale (VAS) for anxiety and a sedation score. Assessments of pain, anxiety and sedation were repeated approximately 2–6 hours postoperatively. Dogs were divided into groups based on preoperative REF (‘Low REF’ and ‘High REF’), and VAS scores (‘Low VAS’ and ‘High VAS’). Scores (CMPS-SF, REF, VAS and sedation) were compared between groups using Mann–Whitney U tests. Preoperative and postoperative CMPS-SF, REF and VAS scores were compared using Wilcoxon signed-rank tests. Relationships between anxiety and CMPS-SF scores were assessed using a Spearman rank correlation coefficient. Scores are presented as median (range). A p value of < 0.05 was considered significant.ResultsWhen divided based on REF, CMPS-SF scores did not differ between groups preoperatively [Low REF: 2 (0–3), High REF: 2 (1–3); p = 0.509] or postoperatively [Low REF: 3 (2–5), High REF: 3 (2–5); p = 0.624]. When divided based on VAS, CMPS-SF scores did not differ between groups preoperatively [Low VAS: 2 (0–2), High VAS: 2 (1–3); p = 0.215] or postoperatively [Low VAS: 3 (2–5), High VAS: 3 (2–5); p = 1]. Postoperative REF [pre: 4.5 (2–8), post: 5 (4–10); p = 0.0105] and CMPS-SF scores [pre: 2 (0–3), post: 3 (2–5); p = 0.0318] increased significantly compared with preoperative scores.Conclusions and clinical relevanceNo apparent relationship exists between baseline anxiety levels and CMPS-SF scores. Understanding the influence of anxiety when using the CMPS-SF is important when assessing pain in dogs. Anxiety and pain may increase postoperatively in dogs undergoing orthopaedic surgery.  相似文献
