Expression and mechanisms of hsa_circ_087631 in patients with primary biliary cholangitis

AIM To explore the expression and mechanisms of circular RNA hsa_circ_087631 in the patients with primary biliary cholangitis （PBC）. METHODS RT-qPCR was used to detect the expression of hsa_circ_087631 in PBC patients and healthy controls. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was constructed and transfected into human acute T cell leukemia Jurkat cells， and then the expression levels of hsa_circ_087631， Bcl-6 mRNA and interleukin-21 （IL-21） mRNA were detected by RT-qPCR. Dual-luciferase reporter assay was performed to identify the interactions between hsa_circ_087631 and hsa-miR-346. RESULTS The expression of hsa_circ_087631 in the PBC patients was significantly increased compared with the healthy subjects. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was successfully constructed. The expression of hsa-miR-346 was significantly increased after the hsa-miR-346-overexpressing lentiviral vector was transfected into the Jurkat cells， while the expression levels of hsa_circ_0087631， Bcl-6 mRNA and IL-21 mRNA were significantly decreased. After wild-type or mutant hsa_circ_087631 vector and hsa-miR-346 mimics were transfected into 293T cells， the results of dual-luciferase reporter assay showed that hsa-miR-346 significantly decreased the luciferase activity of wild-type hsa_circ_087631 （P<0.01）， but the regulation did not change significantly after mutation of the predicted binding site. CONCLUSION Peripheral blood hsa_circ_087631 level is elevated in the PBC patients. The hsa_circ_087631/hsa-miR-346/Bcl-6 signaling may take effect in human T cells. Hsa-miR-346 significantly reduces the expression of hsa_circ_087631， but it may not be regulated by predicted binding sites.     

Minimally modified low-density lipoprotein up-regulates endothelin receptors in mouse mesenteric artery through p38 MAPK pathway

AIMTo investigate whether minimally modified low-density lipoprotein (mmLDL) affects the quantity and activity of endothelin （ET） type A (ET_A) and type B (ET_B) receptors in mouse mesenteric artery by activating p38 mitogen-activated protein kinase (MAPK) inflammatory pathway. METHODSThe KM mice were divided into normal saline (NS) group (injection of NS via caudal vein), mmLDL group (injection of mmLDL via caudal vein), LDL group (injection of LDL via caudal vein), mmLDL+SB 203580 group (injection of mmLDL via caudal vein and intraperitoneal injection of p38 MAPK pathway specific inhibitor SB 203580) and mmLDL+DMSO group (injection of mmLDL via caudal vein and intraperitoneal injection of DMSO). Mesenteric artery ring segment vasoconstriction dose-response curves affected by sarafotoxin 6c (S6c) and ET-1 were recorded by the myography system. The mRNA levels of ET_B receptor, ET_A receptor and interleukin-6 (IL-6) were detected by RT-qPCR. The protein levels of ET_B receptor, ET_A receptor, IL-6, p38 MAPK, p-p38 MAPK, NF-κB and p-NF-κB were determined by Western blot. The serum concentration of IL-6 was measured by ELISA. RESULTSThe contractile responses of the blood vessel segments to S6c and ET-1 were significantly increased by mmLDL (P<0.01). The mRNA and protein expression levels of ET_A receptor, ET_B receptor, and IL-6 significantly increased (P<0.01). The protein levels of p-p38 MAPK and p-NF-κB were significantly increased (P<0.01). The serum level of IL-6 was significantly increased (P<0.01). These effects of mmLDL were inhibited by p38 MAPK inhibitor SB 203580. CONCLUSION mmLDL increses the serum concentration of IL-6, up-regulates the expression of IL-6, ET_A receptor and ET_B receptor in mouse mesenteric artery, and enhances the vasoconstriction function medi?ated by ET_A and ET_B receptors, which is related to the activation of p38 MAPK inflammatory pathway and downstream NF-κB pathway.     

17β—雌二醇对雄性金鱼卵黄原蛋白的诱导作用

采用腹腔注射17β-雌二醇的方法诱导雄性金鱼卵黄原蛋白产生,注射浓度为0.05mg·g-1BW,诱导2周后取尾静脉血,离心分离血浆,进行血浆常规聚丙烯酰胺凝胶电泳,通过对卵黄原蛋白特性基团磷、脂和糖蛋白的染色,确定了卵黄原蛋白在电泳图谱上的位置,开发了一种简便、高效的定性卵黄原蛋白的聚丙烯酰胺凝胶电泳法。电泳结果表明,在0.05mg·g-1BW的注射浓度下,2周后17β-雌二醇诱导了雄性金鱼卵黄原蛋白产生,并通过ELISA检测卵黄原蛋白的平均含量为690.2ng·mL-1,与对照组雄性金鱼平均含量为10.7ng·mL-1的差异极显著(P<0.01),比雌性对照组检出量285.5ng·mL-1高1倍多;17β-雌二醇诱导组雄鱼血浆钙离子和血总蛋白含量明显增加。     

油菜素内酯对低温胁迫下冬小麦 wrab17基因表达的影响

为了探索油菜素内酯（BR）对低温胁迫下 wrab17基因表达模式的影响,以寒地冬小麦品种东农冬麦1号（Dn1）为试验材料,利用qRT-PCR技术克隆得到 wrab17基因的cDNA序列,利用生物信息学方法分析该序列的特征,采用qRT-PCR技术检测低温（5 ℃、0 ℃、-10 ℃、-25 ℃）胁迫下该基因在冬小麦叶片和分蘖节中的表达情况以及BR对 wrab17基因表达量的影响。结果表明, wrab17基因的编码区序列长度为735 bp,包含500 bp的完整开放阅读框,可编码167个氨基酸,属于稳定蛋白质,该蛋白质无跨膜区和信号肽。qRT-PCR分析表明,低温胁迫下对照组和BR处理组的Dn1叶片和分蘖节中, wrab17基因的相对表达量分别在-10 ℃和-25 ℃时最高,且BR处理组 wrab17基因的相对表达量均高于对照组,推测BR可通过提高 wrab17基因的表达量来增强冬小麦的抗寒能力。     

虹鳟IL-17多克隆抗体的制备及初步鉴定

本研究根据NCBI已发表序列设计引物,提取经植物血凝素刺激后的虹鳟Oncorhynchus mykiss头肾细胞总RNA,采用RT-PCR方法扩增IL-17成熟肽基因,测序结果表明所获得的序列与发表序列相一致。将该基因重组至原核表达载体pET32a中,并转化大肠杆菌Escherichia coli Rosetta,进行诱导表达。SDS-PAGE电泳结果表明:目的蛋白以包涵体形式表达,大小约为32k Da,重组蛋白经Ni-NTA系统纯化、复性后纯度达90%以上,以其免疫小鼠制备虹鳟IL-17多克隆抗体。ELISA结果显示其效价为1∶25 600,而间接免疫荧光结果显示所制备的多克隆抗体能够特异性地识别真核细胞中瞬时表达的虹鳟IL-17。本研究成功制备虹鳟IL-17多克隆抗体,为下一步IL-17在虹鳟黏膜免疫中的作用及其佐剂效应研究奠定基础。     

优质甘蓝型油菜黔油17号亲本原种的保优繁殖技术研究

通过优质甘蓝型油菜隐性核不育系98-116 AB与父本ZW 99004单株成对杂交,并在F1代杂交组合进行配合力测定,获得高配合力的成对株系材料。根据优质杂交油菜新品种黔油17号母本98-116 AB及父本ZW 004的自身特点,分别在人工隔离和自然隔离条件下进行其原原种及原种的保优繁殖技术研究,保证亲本原始的优良种性,确保大面积生产中杂交种的质量以及品种的生产潜力。     

IL-33/ST2 axis in systemic lupus erythematosus in relation to chronic kidney injury and disease activity

AIM: To elucidate the association between chronic kidney injury and interleukin-33 (IL-33; an alarmin)/suppression of tumorigencity 2 (ST2) in patients with systemic lupus erythematosus (SLE).METHODS: Serum levels of IL-33 and soluble ST2 (sST2) were assessed by ELISA in 50 SLE patients and 30 healthy controls (HC).RESULTS: The levels of IL-33 and sST2, and IL-33/sST2 ratio were significantly higher in SLE patients than those in the HC. The IL-33 and sST2 levels were positively associated with SLE disease activity index (SLEDAI), erythrocyte sedimentation rate (ESR), proteinuria and triglyceride, but negatively associated with complement C3. IL-33/sST2 ratio was positively associated with SLEDAI and estimated glomerular filtration rate (eGFR). Independent explanatory variables associated with high IL-33/sST2 included chronic kidney disease (CKD) staging and albumin (R²=0.442), especially CKD staging.CONCLUSION: Elevated serum sST2 and IL-33 levels in SLE patients are correlated with disease activity and risk factors of kidney injury. IL-33/sST2 ratio may serve as a potential biomarker for chronic kidney injury in SLE patients.     

纤维亚麻新品种黑亚17的选育

为实现优质、高产、高抗育种目标,黑亚17是采用外源总DNA导入技术,从法国品种Ariane为供体、品系81-8-6-3为受体的导入后代选育而成的纤维亚麻新品种。原茎、长麻、全麻、种子产量分别达到5595．9、873．1、1338．7和594．7kg·hm^-1,分别比对照增产9．0%、16．5％、11．7％和16．2％。长麻率19．5％,比对照高1．7个百分点;全麻率29．9％,比对照高1．1个百分点。于2007年3月通过黑龙江省农作物品种审定委员会审定推广。     

鲤鱼白细胞介素-1β全长cDNA的克隆·鉴定及其差异表达分析

[目的]进行鲤鱼白细胞介素-1β（IL-1β）全长cDNA的克隆、鉴定及其差异表达分析。[方法]利用DD-RTPCR方法获得差异表达cDNA片段,对有丝分裂原刺激的鲤鱼外周血白细胞cDNA文库进行筛选,克隆了鲤鱼IL-1β的全长cDNA,并进行了序列分析和差异表达分析。[结果]获得的阳性克隆含有1个大小为831 bp编码276个氨基酸的完整开放阅读框。聚类分析表明,鲤鱼IL-1β氨基酸序列与日本鲤鱼紧密聚为一支,氨基酸序列的同源性达95%,之后聚类顺序依次为鲫鱼、斑马鱼、猪、牛、马、人和小鼠。差异表达分析表明,经有丝分裂原刺激后前期（4 h）白细胞中IL-1β的表达量显著增大,但随着时间推移（12,24 h）并非一直较同期大,表达量总体趋势成峰形。[结论]为进一步研究IL-1β在体内的表达方式、功能特点和调控机理以及在炎症反应、应急反应和免疫应答的作用机制奠定了基础。     

杏鲍菇17号杂交菌株选育研究

[目的]选育高产优质的杏鲍菇杂交新菌株。[方法]利用不同菌株间产生的不同交配型的单核菌丝配对杂交形成双核菌丝的原理,用杏鲍菇11号菌株和13号菌株作亲本,进行单孢杂交育种。[结果]选育出杏鲍菇17号杂交新菌株,其子实体呈保龄球形,产量较高,生物效率达121.02%。[结论]杏鲍菇17号各项指标表现稳定,适合大面积推广。
