Thirty-two group A isolates of rotavirus detected in faecal samples from diarrhoeic piglets, were selected for P and G genotyping using a Multiplex RT-PCR. Ten isolates, from animals less than 8 days old, characterized an outbreak of diarrhoea caused by group A rotavirus in animals. P[7],G3 (CRW8-like) and P[7],G5 (OSU-like) genotypes were detected in 5 animals each. Isolates of a group A rotavirus of genotypes compatible with the OSU prototype were those most frequently identified in single infections in older animals (20/32 strains). In addition to these, 20 isolates from piglets with diarrhoea caused by group A rotavirus, collected between May 1998 and June 1999, but not from the outbreak month, were analysed. These isolates were used to compare the types observed on the farm outside the outbreak in May 1999 and the CRW8-like genotype was found in none of these faecal samples. P[7],G5 was the most frequent genotype (10/20 strains). No outbreak of diarrhoea caused by rotavirus in 1-week-old piglets was found in any other period during the 13 months of this study.  相似文献

通过RT-PCR扩增新城疫病毒F基因,利用禽痘病毒复合启动子、SV40 PolyA终止信号序列构建F基因表达盒。将F基因表达盒与loxp-GFP-loxP序列串联插入禽痘病毒重组臂,构建了禽痘病毒转移质粒载体。在脂质体介导下转染CEF,获得了带有绿色荧光信号的重组病毒rFPVFGFP。通过二次转染,利用Cre-loxP位点特异性重组将重组病毒基因组的GFP自动去除,结果获得了只含新城疫病毒F基因表达盒的重组禽痘病毒rFPVF。试验结果表明,rFPVF复制稳定,表达的F蛋白具有免疫原性,能够刺激鸡只产生抗新城疫病毒的中和抗体。  相似文献

Pyriproxyfen, an analog of the juvenile hormone (JH) in insects, has been evaluated in vivo and in vitro on an important stored product pest, the mealworm Tenebrio molitor. On newly ecdysed pupae, this insect growth regulator (IGR) was applied topically (at 0.10 and 0.20 μg/insect). Enzyme immunoassay measurements showed a decrease of the ecdysteroid titer in the hemolymph of treated specimens. In addition, the treatment induced an increase of the protein concentration in the hemolymph, however, there was no significant effect on the protein band pattern. Using in vitro pupal integument explant cultures, pyriproxyfen (at 1 and 10 μM in the medium) caused a significant inhibition of ecdysteroid amounts and of the thickness of new cuticle secreted. These new results are discussed in relation to the physiology of this IGR with a JH action.  相似文献

Diaporthe helianthi the causal agent of sunflower (Helianthus annuus) stem canker, causes significant reductions in yield and oil content in most sunflower-growing areas. With the aim of enhancing host resistance, we selected in vitro sunflower calli against culture filtrates of two pathogen isolates (7/96 and 101/96). This technique may be an effective and rapid tool to discriminate the most virulent D. helianthi isolate and to screen for host resistance in the early stage of a breeding programme. Further investigation on the mechanisms involved in defence pathways showed no induction of salicylic acid and pathogenesis-related proteins in calli, indicating that the host resistance is not associated with Systemic Acquired Resistance but probably other biochemical mechanisms.  相似文献

Relative gene expression pattern of fatty acid transport proteins (FATP and cd36), intracellular fatty acid-binding proteins (FABP3, FABP10 and FABP11), β-oxidation-related genes [carnitine palmitoyl transferase II (CPTII), peroxisome proliferator-activated receptor β (PPARβ), acyl-CoA oxidase (AOX), long-chain fatty acyl-CoA synthetase (FACS), acyl-CoA dehydrogenase (dehydrogenase)] and uncoupling protein 2 (UCP2) was assessed by RT-qPCR in Atlantic salmon muscle (red and white), liver, heart, myosepta and visceral fat. FABP11, a FABP isoform not previously described in Atlantic salmon, was highly expressed in visceral fat and myosepta and at the lower level in red muscle, white muscle, myosepta and heart. Furthermore, Atlantic salmon were fed either a diet containing fish oil (FO) or a complete replacement of FO with a vegetable oil blend (55% rapeseed oil, 30% palm oil and 15% linseed oil; VO) for the production cycle (27 months from start of feeding and until ∼4.5 kg mean weight). The expression of genes related to β-oxidation, fatty acid uptake and transport in the white muscle indicate ( n  = 3) significant down-regulation in VO fed Atlantic salmon and correlated with previously reported white muscle triacylglycerol stores and β-oxidation. FABP11 in visceral fat and myosepta was also down-regulated in VO fed fish.  相似文献

对月季花瓣衰老过程中可溶性蛋白的SDS-PAGE进行了分析,结果表明,蛋白条带变化表现出3种情况:(1)从瓶插开始到结束蛋白条带基本保持稳定;(2)随花瓣衰老,蛋白条带逐渐减弱、消失;(3)随着花的衰老,有新的蛋白条带出现。对花进行ABA或乙烯促衰处理,也导致相应的某些蛋白条带的变化,说明这些蛋白的变化可能与衰老有关。  相似文献

The humoral immune response to crude outer membrane proteins (comp) of S. typhimurium in mice has been characterized. Maximal and quicker antibody response was observed when 50 micrograms of comp was injected intraperitoneally. The comp of smooth C5 strain of S. typhimurium evoked antibody response to both lipopolysaccharide (LPS) and proteins. Absorption of these sera with LPS coated erythrocytes eliminated the antibodies to LPS completely, while the antibody level to protein was left unaltered. The comp from rough mutant (lacking O-specific chain of LPS) of S. typhimurium elicited antibodies to proteins but not to LPS. These results indicate the concomitant production of antibodies to Salmonella outer membrane proteins also. The significance of such antibodies in protection and diagnosis has been discussed.  相似文献

Low temperature is one of the major limiting environmental factors which constitutes the growth, development,productivity and distribution of plants. Over the past several years, the proteins and genes associated with freezing resistance of plants have been widely studied. The recent progress of domestic and foreign research on plant antifreeze proteins and the identification and characterization of plant antifreeze protein genes, especially on expression regulatory mechanism of plant antifreeze proteins are reviewed in this paper. Finally, some unsolved problems and the trend of research in physiological functions and gene expression regulatory mechanism of plant antifreeze proteins are discussed.  相似文献
