圆田螺保护酶分子在大运河污染评价中的应用   总被引:2,自引:2,他引:0  
[目的]研究以中华圆田螺(Cipangopaludina cahayensis)保护酶作为京杭大运河不同污染程度环境监测的可行性。[方法]通过测定螺内脏团中超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)的活性,比较室内模拟法、样点笼内放养法和直接采样法测定保护酶分子标志物活性评价大运河污染方法的优劣。[结果]中华圆田螺组织抗氧化保护酶系统的SOD、POD、CAT活性是指示污染的敏感指标。CAT活性在污染环境中被抑制,且在临界范围内随着污染毒性作用的加强,抑制作用也加强。POD活性在污染环境中被激活,但各时间段的增幅变化却不尽相同。SOD的变化不稳定,它在30 d暴露后的降低表明其较适合作为短时间的检测指标。[结论]中华圆田螺保护酶作为京杭大运河不同污染程度环境监测的分子标志物是可行的,样点笼内放养测定保护酶分子标志物活性评价法最优,直接采样测定保护酶活性法不可取。  相似文献

Background

Symmetric dimethylarginine (SDMA) has been increasingly used as a marker of early chronic kidney disease (CKD) in cats, but little is known about the influence of comorbidities on SDMA in this species.

Hypothesis

Hypertrophic cardiomyopathy (HCM) and diabetes mellitus (DM), independently of CKD, are associated with changes in serum SDMA.

Animals

Ninety‐four cats (17 with CKD, 40 with HCM, 17 with DM, and 20 healthy controls).

Methods

Case‐control study. Clinical examination, echocardiography, ECG, blood pressure, CBC, biochemistry, thyroxine, and SDMA measurement were performed. Urinalysis was performed in controls and cats with CKD and DM. Analysis of variance was used to compare overall differences in the log‐transformed SDMA data among groups. A random forest algorithm was applied to explore which clinical and other factors influenced serum SDMA.

Results

Median (range) serum SDMA for the renal group (positive control) was 19 (10–93) μg/dL, whereas for the control group (negative control), it was 10 (5–15) μg/dL. For the cardiac and diabetic groups, serum SDMA was 9 (4–24) μg/dL and 7 (3–11) μg/dL, respectively. The renal group had significantly higher SDMA concentrations and the diabetic group significantly lower SDMA concentrations compared to all other groups.

Conclusions and Clinical Importance

Serum SDMA concentrations in cats with HCM were not significantly different from those of healthy control cats. Cats with DM, however, had significantly lower SDMA concentrations than controls, a finding that needs further investigation and should be kept in mind when evaluating renal function of cats with this endocrinopathy.  相似文献

Effects of structurally different insecticides, permethrin, chlorfluazuron, chlorfenapyr, prothiofos, methomyl, and thiocyclam on expression of hsp90, hsp70, hsp20.7, and hsp19.7 were examined using cultured cells of the cabbage armyworm, Mamestra brassicae. Significant induction of hsp90, hsp70, hsp20.7, and hsp19.7 expression was observed in response to chlorfenapyr. No induction was observed for the remaining five insecticides. The chlorfenapyr-induced expression was time-dependent and concentration-dependent. A significant reduction in expression levels of hsp70, hsp20.7, and hsp19.7 was observed in the time course when the cells exposed to chlorfenapyr were transferred to chlorfenapyr-free medium. These results suggest that hsp70, hsp20.7, and hsp19.7 might be useful to assess cellular distress or injury by chlorfenapyr.  相似文献

Background

Myocardial injury, detected by cardiac troponin I and T (cTnI and cTnT), has been associated with long‐term death in the noncardiac human intensive care unit (ICU).

Hypothesis

Presence of myocardial injury predicts 1‐year case fatality in critically ill dogs with systemic inflammation.

Animals

Thirty‐eight dogs with evidence of systemic inflammation and no primary cardiac disease.

Methods

Prospective cohort study. In dogs admitted to the ICU with evidence of systemic inflammation, blood samples were obtained at ICU admission for measurement of cTnI and cTnT, and cTnI was measured once daily during ICU hospitalization. Receiver operating characteristic (ROC) curves were used to examine prognostic capacity of admission cTnI, admission cTnT, and peak cTnI concentrations.

Results

One‐year case fatality rate was 47% (18/38 dogs). Admission cTnI concentrations were (median [range]) 0.48 [0.004–141.50] ng/mL, and peak cTnI concentrations were 1.21 [0.021–141.50] ng/mL. Admission cTnT concentrations were 15 [<13–3744] ng/L. For each marker, non‐survivors had significantly higher concentrations than survivors (= .0082–.038). ROC analyses revealed areas under curves [95% CI] of 0.707 [0.537–0.843] for peak cTnI and 0.739 [0.571–0.867] for admission cTnT, respectively. At the optimal cut‐off, concentrations were 1.17 ng/mL (peak cTnI) and 23 ng/L (admission cTnT), sensitivities were 72% and 72%, and specificities were 70% and 80%, respectively.

Conclusions and Clinical Importance

While peak cTnI and admission cTnT are significantly related to 1‐year case fatality in critically ill dogs with systemic inflammation, low sensitivities and specificities prevent their prediction of long‐term outcome in individual patients. Troponins might play a role in identification of dogs at long‐term risk of death.  相似文献

Background

Myocardial injury detected by cardiac troponin I and T (cTnI and cTnT) in cardiac disease is associated with increased risk of death in humans and dogs.

Hypothesis

Presence of myocardial injury predicts long‐term death in cats with hypertrophic cardiomyopathy (HCM), and ongoing myocardial injury reflects change in left ventricular wall thickness over time.

Animals

Thirty‐six cats with primary HCM.

Methods

Prospective cohort study. Cats with HCM were included consecutively and examined every 6 months. Echocardiography, ECG, blood pressure, and serum cTnI and cTnT were evaluated at each visit. Cox proportional hazards regression analysis was performed to evaluate prognostic potential of serum troponin concentrations at admission and subsequent examinations. Correlations were used to examine associations between troponin concentrations and cardiac hypertrophy.

Results

Troponin concentrations at admission were median [range] 0.14 [0.004–1.02] ng/mL for cTnI, and 13 [13–79.5] ng/L for cTnT. Both were prognostic for death (P = .032 and .026) as were the last available concentrations of each (P = .016 and .003). The final cTnT concentration was a significant predictor of death even when adjusting for the admission concentration (P = .043). In a model containing both markers, only cTnT remained significant (P = .043). Left ventricular free wall thickness at end‐diastole (LVFWd) at admission was correlated with cTnI at admission (r = 0.35, P = .035), however no significant correlations (r = 0.2–0.31, P = .074–.26) were found between changes in troponin concentrations and left ventricular thickness over time.

Conclusions and Clinical Importance

Myocardial injury is part of the pathophysiology leading to disease progression and death. Low sensitivities and specificities prevent outcome prediction in individual cats.  相似文献

Residues of arbuscular mycorrhizal fungi (AMF) may be important for agroecosystem functioning due to their ability to promote soil aggregation, especially in coarse textured soils with little biomass input and low capacity to conserve soil organic matter (SOM). Our aim was to assess the fate of AMF residues with prolonged arable cropping in coarse textured soils in a subtropical savannah assuming that glomalin-related soil protein (GRSP), especially the MAb32B11-immunoreactive fraction, mainly constitutes material of AMF origin. In three agroecosystems on the South African Highveld, surface soils were sampled. The former grassland soils had a history of up to 98 yr of cropping. We measured four GRSP fractions: Bradford-reactive soil protein (BRSP) and immunoreactive soil protein (IRSP), and easily extractable fractions of both. The primary grassland sites exhibited generally low contents of SOM and low GRSP contents. Prolonged arable land use of former grassland soils reduced the content of GRSP further. The decline could be described with a mono-exponential function with rate constants ranging from 0.04 to 0.41 yr−1. Depending on the GRSP fraction, steady-state conditions were reached after 11-92 yr on a level of 39-69% of the initial contents. We conclude that even though GRSP fractions had the same hypothesized origin, they comprised pools with different stability or replacement rate. Easily extractable IRSP was lost most rapidly. In contrast to carbon, nitrogen and microbial residue dynamics, GRSP contents were not reduced below a certain steady-state level, despite potentially negative management effects on AMF, such as tillage, inclusion of fallows into crop rotation and fertilization with inorganic phosphorus. The steady-state GRSP contents coincided with low, but steady agroecosystem yields under the given cropping management.  相似文献

To assess the potential of fatty acid (FA) compositions to act as biomarkers in the soil food web, two species of Collembola, Folsomia candida and Proisotoma minuta, were switched to four possible diets: Cladosporium cladosporioides (a common soil fungus), Panagrellus redivivus (a bacteria feeding nematode), Zea mays (maize) and Alnus glutinosa (alder). The change in FA content of the Collembola was observed over the following 39 days. The four diets produced significant shifts in the FA compositions of the Collembola, with P. redivivus causing the most extreme changes; Collembola fed P. redivivus gained complex FA compositions similar to those of the nematode diet. Changes in the relative abundances of some FAs were found to follow negative exponential curves, as the components either accumulated in, or were removed from, the FA pool in the Collembola; abundance half-lives varied between 0.5 and 22.4 days, indicating that Collembolan FA compositions changed readily with the input of new exogenous components. The results demonstrate that Collembolan FA compositions are influenced by diet, and that the abundances of FAs such as i15:0, i17:0 and 18:1(n-7) may be used as biomarkers of nematode consumption by Collembola. In contrast, the C20 polyunsaturated FAs cannot be used as biomarkers for nematode predation as Collembola possess the ability to biosynthesise high abundances of these compounds when not provided by the diet.  相似文献

The issue of animal welfare in aquaculture is of growing interest and there is an increasing consumer demand for documentation of safe and ethically defendable food production. In this context, we have looked for molecular markers among those genes whose expression is modified by the different farming conditions. We have compared gene expression of sea bass farmed at different population densities by differential display, and we have obtained six bands differentially expressed whose sequences have been deposited in the public databases; two of them were suppressed by high population density, while four were induced by the treatment. These genes can be used as biomarkers, and together with a panel of stress-related genes of sea bass (D. labrax) that we have already obtained, could allow the rapid diagnosis of the welfare status of a fish using RT-PCR. We are certain that the new molecular techniques will find their place in the everyday management of fish farming. On the other hand, we are also aware that the scarcity of genomic resources for some fish species, in spite of their economical interest, will retard the beneficial effects that modern biotechnology could bring to aquaculture industry. Therefore, an effort should be made to reduce, as far genomic resources are concerned, the gap that separates farmed species from model organisms such as Danio rerio and Fugu rubripes.  相似文献
