试验旨在探究日粮中添加不同水平β-胡萝卜素(β-C)对牦牛皮下脂肪颜色及组织维生素A含量的影响。试验选取12头2~3岁牦牛为研究对象,随机分为4个组,每组3个重复。各处理组日粮分别在基础日粮中添加0(对照)、720(低剂量)、1 440(中剂量)、1 620 mg/d(高剂量)β-C,饲养90 d后,分别测定牦牛脂肪、肌肉色度、背膘厚度及不同组织中β-C和代谢产物维生素A的含量,结果表明:日粮中添加不同水平β-C,牦牛背部、胸部脂肪及半腱肌的黄度值(b*)显著高于对照组(P<0.05),亮度值(L*)和红度值(a*)没有显著变化(P>0.05);牦牛背膘厚度在中剂量组达到最大值;对照组牦牛血清中β-C及维生素A含量显著低于各试验组(P<0.05),且维生素A含量随β-C添加量的上升呈上升趋势(P<0.05);牦牛皮下脂肪、腹腔脂肪、心脏、肝脏、脾脏、肺脏、肾脏及空肠等组织器官中β-C含量在中剂量组均达到最大值,除肝脏中维生素A含量较高外,其他组织中维生素A含量在各处理组间差异不显著(P<0.05)。综上所述,日粮添加β-C影响牦牛皮下脂肪颜色,且在中剂量组牦牛皮下脂肪、心脏、肝脏、脾脏及空肠等组织器官中β-C含量达到最大值,故推荐其最佳摄入量为1 440 mg/d;在日粮中添加β-C影响牦牛肝脏和血清中维生素A含量,对其余组织器官中维生素A含量没有影响。  相似文献

In order to investigate the genetic diversity and the origin of evolutionary relationship of Zhongdian yak,we analyzed the complete sequence of 15 individuals Cytb gene,its sequence polymorphism was analyzed,and the phylogenetic tree was constructed.The results showed that the length of the nucleotide sequence were 1 140 bp,with nucleotide frequencies of 26.3%,31.8%,13.1% and 28.8% for T,A,G and C,respectively.Three haplotypes were identified of 15 individuals,with 3 polymorphic sites,including two conversions,one transversion,haplotype diversity was 0.2571 and nucleotide diversity was 0.00035.Phylogenetic analysis suggested that Zhongdian yak and Bos mutusc clustered firstly,then gathered with Bison bison,which indicated that there were high genetic similarity and closer genetic relationship,genetic similarity with other cattle genus was relatively low.Combining with the proof of molecular biology and paleontology,the result supported the point that Bos grunniens and Bos mutus were classified as an alone genus in Bovinae.  相似文献

为了研究性别对查吾拉地区牦牛的产肉性能及肉营养成分的影响,随机选取8岁健康的查吾拉牦牛10头,公母各半,对不同组别间牦牛进行屠宰试验比较肉营养品质的差异。结果表明,母牦牛屠宰率、净肉率、胴体率、骨肉比均高于公牦牛,但无统计学差异。肉营养成分分析结果显示,母牦牛肉中维生素A、E、B12、半胱氨酸、组氨酸、精氨酸、钙、硒、脂肪酸含量高于公牦牛(P<0.05)。研究结果表明,查吾拉牦牛富含较高的营养成分,可为人类提供优质健康的动物蛋白,具备作为优质牦牛肉源生产的潜力。相比较,母牦牛肉优于公牦牛。  相似文献

In this study,hypoxia inducible factor-1α(HIF-1α) gene was cloned from yak spleen by RT-PCR,and it was divided into two sections for amplification.The sequence and protein structure were analyzed using related bioinformatics tools.The length of the coding region was 2 472 bp,which encoded 823 amino acids.Its molecule mass was 92.13 ku,and PI was 5.09.It displayed as a hydrophilic protein,whose secondary structure was mainly composed of random coil and α-helix.It had 69 phosphorylation sites and 4 N-glycosylation sites.Phylogenetic tree displayed that Maiwa yak,Bos grunniens,Bos mutus and Bos taurus gathered into one cluster.Maiwa yak HIF-1α gene was successfully cloned in this study,it provided a viable reference for further study of genetic characteristics of HIF-1α.  相似文献

旨在克隆获得牦牛StAR基因编码序列(CDS)并进行生物信息学分析,探究其mRNA组织表达特性。本研究以屠宰场采集的成年母牦牛心、肝、脾、肺、肾、卵巢、输卵管、子宫组织(n=5),不同年龄(胎牛、1岁、2岁)牦牛的卵巢(n=3),不同发情周期(卵泡期、黄体期)的牦牛卵巢(n=3),黄体期黄牛的卵巢(n=3)及实验室冻存的牦牛颗粒细胞为研究材料。以牦牛黄体期卵巢cDNA为模板,用逆转录PCR克隆StAR基因,并使用MEGA7.0和ExPASy-ProtParam等软件分析其生物信息学特性;采用实时荧光定量PCR技术分析牦牛StAR基因组织表达特性。结果发现,StAR基因CDS区长858 bp,编码285个氨基酸,StAR蛋白总体带正电荷,属于碱性亲水稳定蛋白,无跨膜结构及信号肽,主要存在于细胞质和线粒体; StAR基因具有较高的保守性,符合物种进化规律。牦牛StAR基因在卵巢表达水平最高(P<0.01),且2岁时卵巢表达水平极显著高于胎牛和1岁龄(P<0.01),黄体期卵巢表达水平极显著高于卵泡期(P<0.01);黄体期黄牛卵巢中StAR基因的表达量极显著高于牦牛(P<0.01);在颗粒细胞的体外培养过程中StAR基因表达量逐渐上升,在培养24 h时达到高峰(P<0.01),随后显著降低。综上所述,StAR基因序列较为保守,在牦牛卵巢组织中表达最高,且表达水平随年龄与卵巢周期而变化,提示StAR基因可能参与牦牛卵巢及黄体功能相关的繁殖调控。  相似文献

对1株分离自西藏那曲地区养殖场有出血性症状牦牛的致病性菌进行分子鉴定及药敏分析,为西藏地区牦牛出血性疾病提供治疗依据。通过对病死牦牛肺脏、肝脏进行细菌分离纯化获得疑似菌株,对所得疑似菌株进行形态学观察与哥伦比亚血平板试验筛选出疑似致病菌株,再对疑似致病菌株进行生理生化鉴定试验、16S rDNA通用引物检测及测序、同源性比对,后经药敏试验得到该疑似致病菌株的敏感药物,最后通过动物回归试验验证其敏感药物的治疗效果。结果表明,通过对病死牦牛肺脏、肝脏分离纯化获得6株疑似菌株,经形态学观察试验、哥伦比亚血平板试验筛选出1株具有溶血性的革兰氏阳性球菌S-4。经生理生化鉴定试验、16S rDNA测序、同源性比对,鉴定S-4菌株为表皮葡萄球菌;药敏试验结果显示,S-4菌株对恩诺沙星、新霉素、多黏菌素B、卡那霉素、环丙沙星敏感,对氟苯尼考、多西环素中介敏感,对链霉素、红霉素、四环素、青霉素、头孢氨苄耐受;动物回归试验显示,该菌株具有致病性,且恩诺沙星、新霉素、卡那霉素3种药物治疗效果良好,多黏菌素B、环丙沙星治疗效果差。本试验获得1株具有致病性的牦牛源表皮葡萄球菌,该致病菌在养殖过程中可使用恩诺沙星、新霉素、卡那霉素3种药物进行治疗。  相似文献

To investigate the species of cellulase-producing Bacillus in yak rumen,10 samples of rumen content were aseptically collected from 10 adult Maiwa yaks to isolate the heat-resistant Bacillus by water bath at 80 ℃ for 20 min.The cellulase-producing strains were screened using the CMC-Na medium and Congo red staining.The 16S rRNA gene sequence of those cellulose-producing strains were amplified and sequenced.The results showed that 64 strains were isolated from the 10 samples.Total 23 strains were identified as cellulase-producing bacillus,including 16 strains of Bacillus cereus,7 strains of Bacillus thuringiensis.Furthermore,phylogenetic analysis showed that the 16 Bacillus cereus strains were clustered into two branches:One isolate was clustered into a branch alone,the other 15 isolates were clustered into a branch which clustered into 5 small branches,showing that there was certain genetic diversity in the isolates of Bacillus cereus.And all 7 Bacillus thuringiensis strains were clustered into a branch.Hence,the results layed the foundation of investigating the species of cellulase-producing Bacillus in yak rumen and developing probiotics special for yak.  相似文献

This study was aimed to understand the characteristics of length polymorphism with repeat sequence of keratin associated protein 1 (KAP1) family genes in yak. KAP1 family genes of yak and cattle were sequenced, and compared with sheep KAP1 family gene sequences. The results showed that cattle KAP1 family genes were located in chromosome 19, according to location of sheep KAP1 family genes in the chromosome and similarity with cattle KAP1 family genes, renaming the cattle KAP1 family (according to the gene location of chromosome) B2D, B2A, KAP1-1 and B2C genes into KAP1-4, KAP1-1, KAP1-2 and KAP1-3 gene, respectively. KAP1 family genes in the 3'and 5' flank were highly conserved, the difference between family genes mainly in the the repeat sequence region, which yak KAP1 to KAP4 genes were found 30 bp length polymorphism. There were B(CCQTS)A1(CCQPT) repeat sequence and a new repeat sequence C(SIQTS). The results indicated that the repeat sequence was the key of the polymorphism of KAP1 family genes, which might be relate to combination with keratin protein.  相似文献

The effects of prior climate change on yak breed distributions are uncertain. Here, we measured changes in the distributions of 12 yak breeds over the past 50 years in China and examined whether the changes could be attributed to climate change. Long‐term records of yak breed distribution, grey relational analysis, fuzzy sets classification techniques and attribution methods were used. Over the past 50 years, the distributions of several yak breeds have changed in multiple directions, mainly shifting northward or westward, and most of these changes are related to the thermal index. Driven by climate change over the past years, the suitable range and the distribution centers of certain yak breeds have changed with fluctuation and have mainly shifted northward, eastward or southward. The consistency of observed versus predicted changes in distribution boundaries or distribution centers is higher for certain yak breeds. Changes in the eastern distribution boundary of two yak breeds over the past 50 years can be attributed to climate change.  相似文献
