Summary Components of partial resistance, infection frequency and latency period, were determined in 71 winter and spring wheat genotypes in the seedling stage, after infection with three races of leaf rust (Felix 3B, Clement B and Betuwe 85C) at three different day/night temperature regimes (24/18°C, 18/12°C and 12/6°C). The genotypes were split into two groups and two separate experiments were carried out. Five genotypes, SVP 84039, Akabozu, Banco, BH 1146 and Orso, conferred a low infection frequency and a long latency period and Westphal 12A a long latency period, indicating a relatively high level of partial resistance. The correlation coefficient between infection frequency and latency period was low. Race-specificity was not found. There was a significant temperature effect on the latency period. In the second experiment the temperature x genotype interaction was significant. Temperature-response functions of transformed data demonstrated that the latency periods of four relatively resistant genotypes, Westphal 12A, Banco, BH 1146 and Orso and of Sarno and Mirela were most sensitive to temperature. The range between the genotypes with the longest and the shortest latency period was highest at 12°C. Summary Seedling emergence was closely correlated with coleoptile length and plant height among parents, F2 and F3, populations of crosses involving dwarf wheats Olesen Dwarf (CI 14497), Norin 10 derivative D6301, Tom Thumb derivative D6899, and the standard-height varieties Ramona 50 and Nainari 60. Genetic mechanisms that governed plant height also influenced coleoptile length, but the relative effects of genes showing dominant or epistatic effects appeared to be different. With respect to the two parents involved in each of 15 crosses, mean F2 coleoptile lengths were consistently closer to the low parent value than were corresponding mean F2 plant heights. A slight curvilinear relationship was also found between coleoptile length and plant height of F3 lines. The results suggest that selection of semidwarf wheats with long coleoptiles and improved emergence properties from crosses involving the dwarf wheats of this study would be unlikely. 相似文献

以草地早熟禾品种——新格莱德成熟种子为供试材料,在含3.0mg/L2,4-二氯苯氧乙酸(2,4-D)、0.5mg/L6-苄氨基嘌呤(6-BA)的MB5培养基中进行胚性愈伤组织诱导的培养。并从生长了7~9个月的胚性愈伤组织中分离出原生质体,将该原生质体置于KM8P培养基(含3.0mg/L2,4-D、0.5mg/L6-BA、100mg/L水解酪蛋白、100mg/L水解乳蛋白、1%蔗糖、0.4mol/L甘露醇)中进行了液体浅层培养。结果表明,新格莱德原生质体在上述KM8P培养基中培养3d后出现第1次细胞分裂,2~3周后形成小细胞团,此时添加低渗培养液2~3次,小细胞团持续分裂并形成愈伤组织。当愈伤组织块长至3~5mm时,转入固体培养基MS+3.0mg/L2,4-D+0.5mg/L6-BA和MS+0.5mg/L萘乙酸(NAA)+5.0mg/L6-BA上进行培养,使其细胞增殖和分化,且逐步形成完整的植株。 相似文献

Background: The Reflovet system is designed for chemical analysis of whole blood. However, plasma or serum is recommended for potassium analysis because of possible interference from RBC potassium. Because RBC potassium concentration is low in most canine erythrocytes, however, there should be little or no interference.
Objective: The objective of this study was to compare potassium results obtained in whole blood and in plasma from dogs using the Reflovet system.
Methods: Blood samples were collected from 104 dogs into lithium-heparin tubes. The potassium concentration was measured in whole blood, and subsequently the PCV was measured. Samples were centrifuged and the potassium concentration was measured in plasma. Comparisons were made using Deming's regression and Bland-Altman difference plots.
Results: There was very good correlation between results of potassium measurements in whole blood and plasma ( r = 0.93). Potassium values were moderately lower in whole blood: Potassiumblood = 0.912 × Potassium plasma + 0.119. Hemolysis had a negligible effect on the results, but the difference increased with the PCV value. In more than 90% of samples, the difference between the 2 measurements was ≤ 0.3 mmol/L.
Conclusion: There is only a negligible difference in most cases between potassium values in canine plasma and whole blood using the Reflovet system. 相似文献

主要将aFGF基因按照植物偏爱的密码子进行基因优化,同时加上信号肽、6-组氨酸标签及凝血酶切割因子,合成全基因,再将基因插入到改造的TΩ4AB质粒中,将aFGF基因和质粒上的增强子、启动子、Ω序列及ployA加尾序列双酶切,命名为TΩ4AB-aF,将TΩ4AB-aF插入到pBI121载体中命名为pBI121-TΩ4AB-aF。利用三亲融合法将pBI121-TΩ4AB-aF转入到农杆菌菌株LBA4404中,转化苜蓿。转基因苗用卡那霉素进行筛选;并对抗性苗进行PCR、RT-PCR、Western Blot检测。结果表明,aFGF在苜蓿中得到表达。为苜蓿作为植物生物反应器奠定理论基础。 相似文献

选用1344尾平均体重为105 g的健康湘云金鲫鱼种。随机分为4个处理,每个处理2个重复。Ⅰ组投喂基础饲料,Ⅱ、Ⅲ、Ⅳ组在基础饲料中分别添加4 m g/kg的黄酶素及10、20 g/kg葡萄籽等植物提取物。30 d后测定湘云金鲫的生长性能和鱼体成分。结果显示:Ⅲ组与Ⅰ组、Ⅱ组、Ⅳ组比较,增重率分别提高53.77%、45.43%、20.16%(P<0.05),饵料系数分别降低33.06%、31.02%、16.44%(P<0.05);鱼体蛋白质含量显著提高(P>0.05),体脂肪含量无显著变化(P>0.05)。因此,葡萄籽等植物提取物作为饲料添加剂能显著提高湘云金鲫的生长性能、降低饵料系数、改善鱼体品质,本试验条件下,其适宜添加量为10 g/kg。 相似文献
