芦荟属植物含芦荟素结构研究   总被引:4,自引:0,他引:4  
应用植物解剖学、组织化学、荧光显微镜、透射电镜及植物化学技术,研究了芦荟属AloeL.植物茎、叶的结构与芦荟素类物质积累的关系。结果表明,叶维管柬内的大型薄壁细胞为芦荟素的主要贮存结构。其来源于原形成层,属特化的韧皮部大型薄壁细胞。芦荟素在叶的同化组织细胞的质体中合成,通过质外体途径转运到维管柬鞘细胞,由其胞间连丝运送到大型薄壁细胞内贮存。该属植物叶内维管柬的密度、大型薄壁细胞的大小和同化组织的厚度与芦荟素含量呈正相关。同一植物中芦荟素的含量为幼叶〉成熟叶〉衰老叶,叶片顶部〉中部〉基部,并与其维管柬的密度呈正相关。研究结果澄清了有关芦荟中含芦荟素结构的一些争议。  相似文献

为了解决铁皮石斛种苗稀缺的现状,提高种苗繁殖率,以铁皮石斛种子(成熟率80%~90%)、带芽茎段、叶片3种材料作为外植体,进行组织培养研究.结果表明:铁皮石斛最适宜外植体材料是种子,最适宜的类原球茎诱导培养基为1/2MS,最适宜的类原球茎增殖培养基为MS+KT1.5 mg/L+NAA0.5 mg/L+CM100 mL/L+活性炭0.3 g/L,最适宜类原球茎分化培养基为MS+KT1.5 mg/L+NAA0.5 mg/L+100 mL/L苹果汁,最适宜生根培养基为MS+IBA1.5 mg/L.  相似文献

为了探究PEDV(猪流行性腹泻病毒)流行毒株SHpd/2012侵染细胞的特性,研究采用IFA(间接免疫荧光)及绘制病毒一步生长曲线等方法,对感染PEDVSHpd/2012株后的Vero细胞(非洲绿猴肾细胞)病变过程进行研究。结果发现,CPE(细胞病变)主要表现为细胞肿胀、变圆、皱缩、聚集成合胞体甚至从培养瓶壁大量脱落。IFA结果显示,接毒后12h病毒开始增殖,在接毒后36h时感染细胞数量最多,其后病毒增殖速度逐步下降,60h时趋于平稳。病毒一步生长曲线结果显示,SHpd/2012株感染细胞后24~60h的毒价一直处于较高水平,60h后开始下降。此外,本研究成功克隆并真核表达了PEDV S蛋白,为研究PEDV与受体蛋白的互作及病毒的致病机制奠定基础。  相似文献

Advanced maternal age is a risk factor for female infertility, and placental dysfunction is considered one of the causes of pregnancy complications. We investigated the effects of advanced maternal aging on pregnancy outcomes and placental senescence. Female pregnant mice were separated into three groups: young (3 months old), middle (8–9 months old), and aged (11–13 months old). Although the body weights of young and middle dams gradually increased during pregnancy, the body weight of aged dams only increased slightly. The placental weight and resorption rate were significantly higher, and live fetal weights were reduced in a maternal age-dependent manner. Although mRNA expression of senescence regulatory factors (p16 and p21) increased in the spleen of aged dams, mRNA expression of p16 did not change and that of p21 was reduced in the placenta of aged dams. Using a cytokine array of proteins extracted from placental tissues, the expression of various types of senescence-associated secretory phenotype (SASP) factors was decreased in aged dams compared with young and middle dams. The aged maternal placenta showed reduced immune cell accumulation compared with the young placenta. Our present results suggest that models using pregnant mice older than 8 months are more suitable for verifying older human pregnancies. These findings suggest that general cellular senescence programs may not be included in the placenta and that placental functions, including SASP production and immune cell accumulation, gradually decrease in a maternal age-dependent manner, resulting in a higher rate of pregnancy complications.  相似文献

本研究旨在揭示山核桃水溶性成分的神经营养活性及活性因子。利用脱脂山核桃水溶性提取物处理SH-SY5Y细胞,用倒置相差显微镜观察细胞突触生长,用qPCR检测相关基因表达,使用HPLC 及LC-MS对山核桃水溶性成分神经营养活性因子进行分离鉴定。结果显示,山核桃的水溶性成分能够显著促进SH-SY5Y细胞的突触生长,并可以显著上调SH-SY5Y细胞NF160 基因的表达。HPLC及LCMS检测结果发现其水溶性成分主要组成为胞苷、尿苷、鸟苷、腺嘌呤、腺苷等。由此可知,山核桃水溶性成分通过上调NF160基因表达具有神经营养活性。  相似文献

以高粱无菌苗幼叶为试验材料,对颗粒状胚性愈伤组织的获得、细胞悬浮系的建立及影响悬浮细胞生长的主要因素进行了研究。结果表明:幼叶在MS(Murashige and Skoog)+2mg/L 2,4-D培养基上诱导出的愈伤组织,经2~3次继代筛选后获得了浅黄色、颗粒状胚性愈伤组织;胚性愈伤组织接种于液体培养基中,于25±1℃、黑暗条件下,经45~60d的悬浮震荡培养建立了高质量的细胞悬浮系,细胞生长曲线呈“S”型,细胞密度可达6.45×10 5~5.08×10 6个/mL以上,活细胞率可达72.76%,近圆细胞率可达87.50%;培养基的基本成分、激素种类和水平对悬浮细胞生长状态有很大的影响,相比1/2MS培养基,MS培养基为适宜的培养基,2,4-D对保持细胞系的稳定增殖至关重要,适宜的浓度为1mg/L;培养液中加入0.5mg/L KT或6-BA会造成悬浮液褐化,影响悬浮细胞的生长;最适宜的细胞悬浮培养基为L2培养基(MS+1mg/L 2,4-D,30g/L蔗糖,pH5.8),震荡培养转速为110~120r/min,继代周期为7d,新旧培养基的接种比例为2∶1。  相似文献

Pancreas disease (PD) caused by salmonid alphavirus (SAV) severely affects salmonid aquaculture during the seawater phase. To characterize immune cells in target tissues for SAV infection, heart, pancreas and pyloric caeca were analysed from two groups of fish adapted to seawater for 2 and 9 weeks. The sections were scored for the relative abundance of cells expressing MHC class II, IgM, CD3, CD8 or neutrophil/granulocyte markers using immuno‐histochemical techniques. In general, necrosis of tissue was more severe in fish infected at 2 weeks post‐seawater transfer (wpt) compared with those infected at 9 wpt. At 9 wpt, there were higher numbers of MHC II+ cells in heart, pancreas and pyloric caeca, IgM+ cells in heart and pancreas, and CD3+ cells in pancreas compared to those infected at 2 wpt. The majority of the immune cells infiltrating PD‐affected tissues were MHC II+ and CD3+ cells suggesting that antigen‐presenting cells and T lymphocytes are the main types of immune cells responding to SAV infection. All the investigated cell types were also observed in pyloric caeca of infected fish, suggesting that this tissue may play a role in the immune response to SAV.  相似文献

The histopathology and ultrastructure of the intestine of mullets, Liza ramada and Liza saliens, from Comacchio lagoons (northern Italy) naturally infected with myxozoans and helminths were investigated and described. Sixty‐two (80.5%) of 77 mullets harboured one or more of the following parasites species: Myxobolus mugchelo (Myxozoa), Neoechinorhynchus agilis (Acanthocephala), Haplosplanchnus pachysomus and Dicrogaster contractus (Digenea). Co‐occurrence of helminths with myxozoans was common. The main damage caused by digeneans was destruction of the mucosal epithelium of the villi, necrosis and degeneration of intestinal epithelial cells. More severe intestinal damage was caused by acanthocephalans which reach the submucosa layer with their proboscis. At the site of helminths infection, several mast cells (MCs), rodlet cells (RCs), mucous cells and few neutrophils and macrophages were observed in the epithelium. RCs and mucous cells exhibited discharge activity in close vicinity to the worm's tegument. M. mugchelo conspicuous plasmodia were encysted mainly in muscle and submucosa layers of the intestine. Indeed, spores of M. mugchelo were documented within the epithelial cells of host intestine and in proximity to MCs. Degranulation of the MCs near the myxozoans was very frequent.  相似文献

Lippia alba hydrolate (LAH) is produced as a secondary product in essential oil extraction. The objective of this study was to evaluate the use of LAH in a simulation of the transport of tambaqui (Colossoma macropomum) juveniles. A series of 17 hr simulations were performed using load densities of 30, 60 and 90 juveniles per L and four LAH concentrations of 0.4%, 1%, 2% and 5%, with a control of just water. Fish survival, temperature, dissolved oxygen, conductivity, pH, ammonia excretion, alkalinity water hardness and net Na+, K+, Cl fluxes, were all evaluated before and after the transport simulations. Mucous cell densities in the gills were also determined microscopically, post simulation. The results showed that addition of 5% LAH achieved better survival during the simulation than the water control. Dissolved oxygen was significantly lower with an increased load density. Also, conductivity, NH4 excretion and net ionic fluxes (K+ and Cl?) were lower with 5% LAH, although mainly where higher load densities were tested. Mucous cell density was lower in LAH concentrations of 2% and 5%, again with higher load densities. In view of these findings, it is suggested that 5% LAH is an effective sedative for use in the transport of tambaqui, mainly with high load densities. This work is the first study of LAH and highlights its potential applications in aquaculture management.  相似文献
