本研究旨在探索延边黄牛肌球蛋白重链3(MYH3)和肌球蛋白重链8(MYH8)基因多态性及其与生长性状的关联性,应用DNA直接测序法和高分辨率熔解曲线分型技术对120头24月龄延边黄牛MYH3、MYH8基因的遗传变异进行分析,并结合延边黄牛生长性状数据进行了关联分析。结果显示,在MYH3基因的29602748位点检测到一处T>G突变,包含2个等位基因:T和G,存在3种基因型:TG、TT和GG;在MYH8基因的29406042位点检测到一处C>T突变,包含2个等位基因:C和T,存在3种基因型:CT、CC和TT。关联分析显示,MYH3基因29602748位点T>G突变显著影响24月龄延边黄牛十字部高和腰角宽,表现为GG基因型十字部高显著高于TT基因型(P<0.05),腰角宽显著高于TT和TG基因型(P<0.05)。MYH8基因29406042位点C>T突变显著影响24月龄延边黄牛体重和胸围,表现为CT和TT基因型体重显著高于CC基因型(P<0.05);TT基因型胸围显著高于CC基因型(P<0.05)。本研究结果表明,MYH3、MYH8基因多态性与延边黄牛的部分生长性状相关,可作为现代肉牛育种中分子标记辅助选择的候选基因。  相似文献

拷贝数变异(copy number variation,CNV)具有多种形式的变异结构,在品种多样性、生物进化和疾病相关性等研究中起着重要作用,并具有片段长度大、覆盖范围广等特点。随着分子生物学的发展及DNA测序技术的日渐成熟,人们对遗传变异的研究不断向DNA分子水平深入,多态性标记在畜禽育种中已逐渐成为动物育种研究的趋势和主流。由于CNV对基因的调控和表达所造成的影响更为显著,因此,CNV在重要畜禽中的研究越来越多。目前,已检测出大量有关畜禽重要经济性状的基因序列变异,并有许多研究均表明CNV与动物的重要经济特征及疾病的发生有关。笔者主要通过参考国内外相关的研究报道,简述了CNV的相关研究背景、概念、突变机制,归纳总结了CNV对牛、羊、猪、鸡的经济性状、繁殖性状和疾病调控的影响,以期通过对这些重要畜禽的基因组学研究揭示其适应性遗传机理和表型性状差异的遗传基础,开发相应的分子遗传标记,为畜禽的标记辅助选育提供理论基础。  相似文献

为探究寡腺苷酸合成酶1(oligoadenylate synthase 1,OAS1)基因多态性与松辽黑猪繁殖性状的关联性,试验选取130头松辽黑猪母猪为研究对象,利用Sanger直接测序法测序查找OAS1基因外显子1~8的SNP位点,使用SPSS 19.0软件分析OAS1基因SNP位点与松辽黑猪繁殖性状的关联性。结果显示,在松辽黑猪OAS1基因外显子2、3和6上共检测到33个突变位点;其中在外显子2的110 bp处存在1个SNP位点(G110C),存在3种基因型:GG、GC和CC;在外显子3的176 bp处存在1个SNP位点(C176T),存在3种基因型:CC、CT和TT;在外显子6的145 bp处存在1个SNP位点(C145T),存在3种基因型:CC、CA和AA;在166 bp处存在1个SNP位点(G166A),存在3种基因型:GG、GA和AA;在206 bp处存在1个SNP位点(A206G),存在3种基因型:AA、AG和GG。卡方适合性检验结果显示,松辽黑猪OAS1基因G110C突变位点符合Hardy-Weinberg平衡状态,C176T、C145A、G166A和G206A位点均偏离Hardy-Weinberg平衡状态。群体遗传参数分析结果显示,各SNPs位点遗传杂合度均位于中等水平,为中度多态(0.25<PIC<0.5)。关联分析结果发现,G110C位点GC基因型个体总产仔数、产活仔数和断奶仔猪数均显著高于GG基因型个体(P<0.05);C176T位点CT基因型个体断奶仔猪数显著高于CC基因型个体(P<0.05);C145T位点CC基因型个体总产仔数和产活仔数均显著高于AA基因型个体(P<0.05);G166A位点GA基因型个体断奶仔猪数显著高于GG基因型个体(P<0.05);A206G位点GG基因型个体总产仔数和产活仔数显著高于AA基因型个体(P<0.05)。结果表明,OAS1基因外显子区存在突变位点,对松辽黑猪部分繁殖性状有显著性影响。  相似文献

为探索瘦肉型猪组合配套杂交方法和效果而进行试验。试验采用HN121(温氏杜洛克猪)、HN212(法系皮特兰♂×温氏杜洛克♀)为父本,以HN204(温氏大白♂×美系长白♀)为母本,对HN323(HN121×HN204)和HN424(HN212×HN204)两个组合配套进行研究。结果表明,HN424组合与HN323组合相比母猪的窝产仔数、窝产活仔数、仔猪初生窝重、21日龄仔猪成活率、21日龄窝重等繁殖性状差异不显著(P>0.05);HN424组合仔猪的体重、背膘厚降低5.63%(P<0.05),瘦肉率提高4.95%(P<0.05),眼肌面积增大33.08%(P<0.01),肉色评分降低7.19%(P<0.05),大理石纹评分降低11.30%(P<0.01),滴水损失率提高10.63%(P<0.01),其他遗传性状差异不显著;HN424组合与HN204二元母系相比,日增重提高9.26%(P<0.05),背膘厚减少16.41%(P<0.01),料肉比减少8.02%(P<0.05);HN323组合与HN204二元杂交母系相比,日增重提高7.20%(P<0.05),背膘厚减少11.43%(P<0.01),料肉比降低5.35%(P<0.05)。说明采取四元杂组合和三元杂交组合均对“大×长”(HN204)二元杂交有显著的遗传改良作用,“皮×杜×长×大”四元杂组合可显著提高猪的瘦肉率、眼肌面积,但肉色、大理石纹、滴水损失率等肉质指标变差,建议在“皮×杜×长×大”四系组合配套杂交中加入能改良肉质基因的猪种。  相似文献

通过田间试验,就制种玉米密度对土壤水分、农艺性状、产量构成及产量的影响进行了研究分析。结果表明:1在拔节期以前,低密组的土壤含水量比高密组的高;在拔节期到灌浆期,高密组的比低密组的高。在整个生育期,拔节期前土壤含水量比较高,而后逐渐降低,吐丝期达到最低,然后又逐步上升。2在一定密度范围内,制种玉米株高、亩产量随着密度的增大而增加单株产量产量相反;叶长、穗粗、穗粒数先随着密度的增大而减小,后随着密度的增大而增大;叶宽、穗长、百粒重、出籽率先随着密度的增大而增加,后随着密度的增大而减小;叶面积、茎粗、穗位先随着密度的增大而减小,后随着密度的增大而增加,然后又随着密度的增大而减小,秃顶变化规律相反。  相似文献

采用田间试验,以豫麦69为试验材料,研究了不同水分处理下常规灌溉和一体化垄作沟灌的冬小麦产量与穗部性状的相互关系。结果表明,同常规灌溉方式相比,一体化垄作沟灌方式下,冬小麦的穗粒数、籽粒质量及产量分别增加了5.5356%、7.5489%、7.7454%,但穗数减少了0.4302%。常规灌溉和一体化垄作沟灌方式下,产量与穗数、穗粒数以及籽粒质量均正相关,但一体化垄作沟灌方式下的相关系数较常规灌溉方式大。一体化垄作灌溉有利于改善农田小气候、发挥作物的边行优势和提高小麦产量。  相似文献

This study investigated the effects of amino acids (AA) supplementation in low crude protein (CP) diets on growth performance and carcass characteristics in finishing gilts. One hundred and eighty gilts (59.1 ± 5.1 kg) were randomly allotted to one of five diets which consisted of a high CP (15.6%) diet or four low CP (11.6%) diets for 50 days. The low CP diets were supplemented with lysine + threonine + methionine (LCM), LCM + tryptophan (LCT), LCT + valine (LCV) or LCV + isoleucine (LCI), respectively. Gilts were housed at six pigs per pen with six pens per treatment. At the end of the 50‐day experiment, 30 gilts (one pig per pen) with average body weight (BW) of 98 kg were killed to evaluate carcass traits. The pigs fed the diet supplemented with LCV obtained the highest average daily gain (ADG), which was higher than those of pigs fed the diet supplemented with LCM (P < 0.05). Dietary supplementation with tryptophan, valine and isoleucine in low CP diets increased ADG (linear and quadratic effect, P < 0.05), serum levels of valine (quadratic effect, P < 0.05) and isoleucine (linear and quadratic effect, P < 0.05) and immunoglobulin G (IgG) and IgA (linear and quadratic effect, P < 0.05) in finishing gilts. © 2016 Japanese Society of Animal Science  相似文献

Our study investigated the effects of condensed tannins (CT) on rumen in vitro methane (CH4) production and fermentation characteristics by incubating lucerne in buffered rumen fluid in combination with different CT extracts at 0 (control), 40, 80 and 120 g CT/kg of substrate DM. Condensed tannins were extracted from four sainfoin accessions: Rees ‘A’, CPI63763, Cotswold Common and CPI63767. Gas production (GP) was measured using a fully automated GP apparatus with CH4 measured at distinct time points. Condensed tannins differed substantially in terms of polymer size and varied from 13 (Rees ‘A’) to 73 (CPI63767) mean degree of polymerization, but had relatively similar characteristics in terms of CT content, procyanidin: prodelphinidin (PC: PD) and cis:trans ratios. Compared to control, addition of CT from CPI63767 and CPI63763 at 80 and 120 g CT/kg of substrate DM reduced CH4 by 43% and 65%, and by 23% and 57%, respectively, after 24‐h incubation. Similarly, CT from Rees ‘A’ and Cotswold Common reduced CH4 by 26% and 46%, and by 28% and 46% respectively. Addition of increasing level of CT linearly reduced the maximum rates of GP and CH4 production, and the estimated in vitro organic matter digestibility. There was a negative linear and quadratic (p < 0.01) relation between CT concentration and total volatile fatty acid (VFA) production. Inclusion of 80 and 120 g CT/kg of substrate DM reduced (p < 0.001) branched‐chain VFA production and acetate: propionate ratio and was lowest for CPI63767. A decrease in proteolytic activity as indirectly shown by a change in VFA composition favouring a shift towards propionate and reduction in branched‐chain VFA production varied with type of CT and was highest for CPI63767. In conclusion, these results suggest that tannin polymer size is an important factor affecting in vitro CH4 production which may be linked to the CT interaction with dietary substrate or microbial cells.  相似文献

A study was conducted to assess the combined effect of heat stress and nutritional restriction on growth and reproductive performances in Malpura rams. Twenty‐eight adult Malpura rams (average body weight (BW) 66.0 kg) were used in this study. The rams were divided into four groups: CON (n = 7; control), HES (n = 7; heat stress), NUS (n = 7; nutritional stress) and COS (n = 7; combined stress). The study was conducted for a period of 2 months. CON and HES rams had ad libitum access to their feed while NUS and COS rams were under restricted feed (30% intake of CON rams) to induce nutritional stress. The HES and COS rams were kept in climatic chamber at 42 °C and 55% relative humidity for 6 h a day between 10 : 00 h and 16 : 00 h to induce heat stress. Body weight increased significantly (p < 0.05) in CON as compared to NUS and COS. When compared within groups, scrotal width morning, scrotal width afternoon, scrotal circumference morning and scrotal circumference afternoon were significantly (p < 0.05) larger in CON while smaller in COS rams. The higher testicular length was recorded both during morning (p < 0.05) and afternoon (p < 0.01) in COS rams while the lowest in NUS rams. The highest plasma testosterone concentration was recorded in CON and lowest in COS rams. Semen volume and mass motility also differed significantly (p < 0.05) between the groups. The highest semen volume and mass motility was recorded in CON and NUS while lowest in both HES and COS rams. It can be concluded from this study that when two stressors occur simultaneously, they may have severe impact on reproductive performance of rams.  相似文献

Myostatin is a negative regulator of muscle growth and development in mammals, and variation in ovine myostatin gene (MSTN) has been demonstrated to be associated with variation in the muscularity of sheep. Polymerase chain reaction–single‐stranded conformational polymorphism (PCR–SSCP) was used to look for single nucleotide polymorphisms (SNPs) in a 304‐bp amplicon from the promoter region of ovine MSTN. Sequence analyses revealed two previously identified SNPs (c.?2449G/C and c. ?2379T/C) that resulted in three haplotypes (H1 (c.[?2449G; ?2379C]), H2 (c.[?2449C; ?2379C]) and H3 (c.[?2449G; ?2379T]). The effect of these SNPs on growth and carcass traits was investigated in 357 NZ Romney lambs. General linear mixed‐effect models revealed that sheep with the genotype c.?2449GC had a higher loin meat yield (p = 0.032) and proportion loin yield (p = 0.028), than those with the genotype c.?2449GG. The genotype c.?2379CC was associated with an increase in three weight traits: birthweight (p = 0.003), tailing weight (p = 0.009) and weaning weight (p = 0.028), when compared with the genotype c.?2379TC, but it was not found to have an association with growth rate. This suggests that c.?2379T/C has an effect that originates at, or before birth. Haplotype H3 was associated with a decrease in birthweight (p = 0.002), tailing weight (p = 0.003) and weaning weight (p = 0.011). Haplotype H2 was associated with increased loin yield (p = 0.012) and proportion loin yield (p = 0.002). The SNPs may have value as genetic markers for improved Romney breeding.  相似文献
