To investigate genes involved in intramuscular adipogenesis in ruminants, 16 genes with dramatic variable expression were selected. These were selected from the differentiation‐ and proliferation‐phase libraries of our previous serial analysis of gene expression (SAGE) studies of a clonal bovine intramuscular preadipocyte (BIP) cell line. We harvested the BIP cells over 12 days after adipogenic stimulation with all‐trans retinoic acid (ATRA). Quantitative real‐time PCR confirmed the earlier SAGE study results of the expression patterns of 15 of the genes. On day 6, TG accumulation increased significantly in the BIP cells but was completely inhibited in the 3T3‐L1 cells (the monogastric reference). ATRA enhanced expression levels of six genes whereas it suppressed expression of eight genes on day 3 of adipogenesis in the BIP cells. Forty‐eight hours after transfection, the messenger RNA expression level of the adipose differentiation‐related protein (ADFP), encoded by one of the upregulated genes, in the ADFP small interference RNA (siRNA)‐transfected cells was 3.5% of that in negative control‐transfected cells. Also, 6 days after induction the TG level in the ADFP siRNA‐transfected cells was 21.8% lower than that in negative control‐transfected cells. This analysis of gene expression profiles after ATRA treatment will contribute to our understanding of the molecular mechanisms involved in bovine intramuscular adipogenesis.  相似文献

This study was designed to compare the fatty acid profiles of four types of fat depots from bulls, steers, and heifers. All animals were Polish Holstein-Friesian and Limousin crossbreds fattened semi-intensively—silage at libitum and concentrate in the amount corresponding to 30% of their net energy requirements. The fatty acid profile in intramuscular, intermuscular, and external and internal fat was determined. The intramuscular fat of bulls was most abundant in total PUFAs and n-6 PUFAs, and functional fatty acids C 18:2, C 20:4, and C 22:5 in comparison with steers and heifers. Regardless of sex category, intramuscular and external fat were characterized by higher levels of UFAs and a more desirable MUFA/SFA ratio than the remaining fat types. It should also be noted that external fat was more abundant in CLA than other fat types, and that the highest CLA content was found in bull fat, compared with the remaining sex categories of cattle.  相似文献

microRNA(miRNA)是一类广泛存在于真核生物中的内源性单链非编码小RNA分子,长度为21~22 nt,以特异的序列互补结合方式调控基因的表达。在经典的miRNA作用机制中,miRNA通常与靶基因3'UTR种子区序列完全或不完全互补配对,从而在转录后水平调控靶基因的表达。越来越多的报道证实,miRNA还可与靶基因5'UTR区、编码区及启动子区结合,进而参与复杂的基因调控过程。随着高通量测序技术和分子生物学实验技术的发展,大量miRNA被鉴定,其参与调控的生物学机制也被逐渐揭示。大量研究表明,miRNA能够参与动物生殖、生长发育、新陈代谢和疾病调控等生物学过程,对维持正常的生命活动具有重要意义。在猪的遗传改良工作中,对主要经济性状(繁殖、生长发育、胴体肉品质、抗病等)进行改良,选育优良品种,是未来养殖业发展的必然趋势。作者就miRNA生成和作用机制及近年来在猪生殖调控、肌肉发育、脂肪沉积和抵抗疾病感染等方面的研究进行综述,为系统了解miRNA在猪重要经济性状调控中的应用提供参考,并为深入开展相关遗传育种研究工作提供依据。  相似文献

Kruppel样因子15(KLF15)是一种新发现的Kruppel样因子家族(KLFs)成员,为一种真核锌指蛋白转录因子。KLF15在动物体内呈现多组织表达特性,该基因过表达能促进动物脂肪沉积,且能影响肉质性状候选基因和Ⅰ型肌纤维基因的表达。因此,深入研究KLF15对脂肪沉积和肌纤维类型的影响及其可能机制,将为改善动物肉品质提供新的思路。本文综述了KLF15的发现、结构特征、表达规律及其与脂肪沉积和Ⅰ型肌纤维之间的关系。  相似文献

Two groups of cows infected with the bovine leukosis virus (BLV) were kept on two different diets and the fat content of their milk was assayed. The results were compared with those obtained from two comparable groups of BLV-free cows. The cows in each group were of similar ages, those in the groups on the poorer diet being 1–4 months post partum, while those on the richer diet were 5–7 months post partum. The mean percentage of fat in the milk from uninfected cows on the poorer diet was 2.94 while that from the similar infected cattle was 3.06. Uninfected cows on the richer diet produced milk containing 3.39% fat, while those that were infected produced milk containing 3.30% fat. No statistical differences in milk fat production were observed between the BLV seropositive and seronegative cows.  相似文献

The poultry industry is considered one of the most efficient industries in producing animal protein. Poultry production consists of several main segments: broilers, commercial eggs, turkeys, ducks, and other poultry, including geese, guinea-fowl, pheasants, quail, and ratites. Currently, ostrich meat is marketed as a healthy red meat because it is characterized by high polyunsaturated fatty acid contents, low saturated fatty acid content, and low cholesterol level compared with red meats, such as beef, while tasting similar to lean beef. Ostrich meat contains lower sodium and higher iron contents than other kinds of meat, making it preferable to hypertensive people and those with anemia. Taken together, ostrich meat is considered as an ideal red meat for individuals seeking healthier lifestyle. Moreover, many different ostrich products, such as leather and feathers, are used by people. The current review paper considers the quality characteristics of ostrich meat, including physical traits, and compares it to other types of meats, such as bovine and other poultry meats.  相似文献

试验通过研究不同生长速度阿勒泰羊主要血液生化指标含量和尾脂中瘦素(Leptin)基因的表达水平,探讨阿勒泰羊生长速度与脂肪沉积之间的关系。选取300只出生时间相近的阿勒泰羊公羔,正常饲喂至6月龄,记录日增重,取平均日增重排前10%和后10%的公羔各30只,分为生长快速组(HBW)和生长慢速组(LBW),从生长快速组随机选12只分为抑制生长组(饲喂低能量饲粮以抑制其生长,HBW-75%,n=6)和生长快速对照组(饲喂标准饲粮,HBW-100%,n=6),生长慢速组随机选12只分为促进生长组(饲喂高能量饲粮以促进生长,LBW-125%,n=6)和生长慢速对照组(饲喂标准饲粮,LBW-100%,n=6);预饲期7 d,正饲期30 d,试验结束后称重并采集血液,屠宰后采集尾部脂肪。检测血液TG、CK、TSH、T3、T4、S.S、GH、INS、PG、Leptin含量或活性及尾部脂肪Leptin mRNA和蛋白表达量,并观察尾部脂肪细胞形态学变化。结果显示,HBW-100%和LBW-100%组初体重与末体重均差异显著(P<0.05),HBW-75%和HBW-100%组初体重与末体重均无显著差异(P>0.05);LBW-100%和LBW-125%组初体重差异显著(P<0.05),末体重无显著差异(P>0.05)。LBW-100%组的T4、S.S、Leptin、TSH、T3含量显著低于HBW-100%组(P<0.05);LBW-125%组T3含量显著高于LBW-100%组(P<0.05);HBW-75%组GH、PG和Leptin含量显著低于HBW-100%组(P<0.05)。LBW-125%组尾部脂肪细胞面积大于LBW-100%组,HBW-75%组小于HBW-100%组(P<0.05)。Leptin mRNA和蛋白表达量检测结果表明,HBW-100%组的Leptin mRNA表达量与LBW-100%组无显著差异(P>0.05),Leptin蛋白表达量极显著高于LBW-100%组(P<0.01)。LBW-125%组Leptin mRNA和蛋白的表达量均显著高于LBW-100%组(P<0.05)。HBW-75%组Leptin mRNA的表达量极显著低于HBW-100%组(P<0.01),Leptin蛋白表达量显著低于HBW-100%组(P<0.05)。综上所述,生长快速组与生长慢速组中,Leptin的表达量与体重和体脂有一定关联;在生长慢速组促进生长后体重和体脂显著增加,Leptin的表达量呈上升趋势;在生长快速组抑制生长后体重和体脂减少,Leptin的表达呈下降趋势。改变生长速度对Leptin的表达量会产生影响,从而改变机体的脂肪沉积状况。  相似文献

Clenbuterol(CLB,盐酸克伦特罗),俗称瘦肉精。由于其在动物体内广泛残留,可能导致食用此类产品的人中毒而被禁止使用,但已有研究表明该制剂可显著减少猪脂肪沉积,因此,对其分子机制进行深入研究将具有重要的理论与实践价值。本研究通过对家猪饲喂高剂量clenbuterol(≥1.5mg/kg体重),提高瘦肉率10%左右。通过实时荧光定量PCR技术对脂肪组织中与脂类代谢密切相关的4个基因研究发现,饲喂clenbuterol的猪脂肪组织内载酯蛋白D(apolipoprotein D,apoD)mRNA丰度增加超过2倍,而脂蛋白酯酶(lipoprotein lipase,lpl)、硬酯酰辅酶A去饱和酶(stearoyl-CoA desaturase,scd)和激素敏感脂酶(hormone sensitive lipase,hsl)mRNA丰度变化不明显。因此,推断clenbuterol通过改变部分脂代谢关键基因的表达来实现减少脂肪积累。载酯蛋白D可能是CLB应答基因之一。  相似文献

Alternative sources of mesenchymal stem cells (MSCs) for replacing bone marrow (BM) have been extensively investigated in the field of bone tissue engineering. The purpose of this study was to compare the osteogenic potential of canine MSCs derived from adipose tissue (AT), BM, umbilical cord blood (UCB), and Wharton''s jelly (WJ) using in vitro culture techniques and in vivo orthotopic implantation assays. After canine MSCs were isolated from various tissues, the proliferation and osteogenic potential along with vascular endothelial growth factor (VEGF) production were measured and compared in vitro. For the in vivo assay, MSCs derived from each type of tissue were mixed with β-tricalcium phosphate and implanted into segmental bone defects in dogs. Among the different types of MSCs, AT-MSCs had a higher proliferation potential and BM-MSCs produced the most VEGF. AT-MSCs and UCB-MSCs showed greater in vitro osteogenic potential compared to the other cells. Radiographic and histological analyses showed that all tested MSCs had similar osteogenic capacities, and the level of new bone formation was much higher with implants containing MSCs than cell-free implants. These results indicate that AT-MSCs, UCB-MSCs, and WJ-MSCs can potentially be used in place of BM-MSCs for clinical bone engineering procedures.  相似文献
