辣椒新品种湘椒6号的选育

湘椒６号是由湖南地方良种伏地尖中选出的自交系Ｆ８１－１－２从北方引进的甜椒品种经６代单株选择而成Ｌ２４－１自交系配制而成的一代杂各。该品种植株生长势强,耐寒、耐湿,果实粗牛角形,果色深绿,微辣,商品性好。从定值致收获３５￣４５天,６６７ｍ＾２产鲜椒３０００ｋｇ。田间表现抗病毒病、疮痂病。     

Effects of oxidized high-density lipoprotein on membrane fluidity and the expression of lymphocyte function associated antigen-1 of monocyte

AIM: To investigate the effects of oxidized high-density lipoprotein (oxHDL) on membrane fluidity and the expression of lymphocyte function associated antigen(LFA-1) of monocyte. METHODS: The membrane fluidity of THP-1 cells was assayed by fluorescence anisotropy with DPH (1,6-dipheny-1,3,5-hexatriene), a fluorescent probe; The LFA-1 expression on THP-1 cells were assayed by flow cytometry with indirect immunofluorescence.RESULTS: The membrane fluidity of THP-1 cells was reduced by 45% and 52% respectively (P＜0.01) after incubation with 100 μg protein/mL oxHDL and oxLDL for 20 h; oxHDL could stimulated the expression of LFA-1 on THP-1 cells, the positive cell rate and mean fluoresence intensity (MFI) increased by 39% and 45% respectively versus control group (P＜0.01) after incubation with cells for 24 h. CONCLUSIONS: By increasing the expression of LFA-1 on monocyte, oxHDL can promote the monocyte-endothelium adhesion; oxHDL also can reduce the membrane fluidity of monocyte, that will limit the returning of monocytes from subendothelium back to the circulation. This suggested that oxHDL may play an important role in atherogenesis.     

Effects of EBV infection on growth and apoptosis of nasopharyngeal carcinoma cell line

AIM: To explore the effect of EBV infection on growth and apoptosis of nasopharyngeal carcinoma(NPC) cell line.METHODS: NPC cell line CNE1 was directly infected by Epstein Barr virus (EBV). The expression of EBV-latent membrane protein 1 (EBV-LMP1) and bcl-2 were detected by immunohistochemistry method (LSAB). The growth of NPC cells was identified by MTT method. Apoptotic carcinoma cells were detected by flow cytometry analysis and the terminal deoxynucletidyl transferase-medicated dUTP-biotin nick end labeling (TUNEL) methods. RESULTS: EBV-LMP1 was positive in CNE1 infected by EBV(E-CNE1). Compared with CEN1, the growth of E-CNE1 apparently increased (P＜0.01). No apoptotic carcinoma cells were detected and bcl-2 postive cells were 2%~3% respectively in 2 kinds of NPC cells.CONCLUSION: Growth of NPC cells is enhanced by EBV infection and EBV-LMP1 expression, but no influence on expression of bcl-2 and apoptosis of NPC cells.     

CYP450 epoxygenase/EET pathway attenuates adipose inflammation of obese mice through HIF-1α

AIM To investigate the effects of cytochrome P450 （CYP450） epoxygenase/epoxyeicosatrienoic acid （EET） pathway on insulin resistance in obese mice, and to explore the possible mechanisms. METHODS High-fat diet-induced obesity model was established in C57BL/6Cnc mice, and the obese mice were randomly divided into 3 groups, including obesity group （treated with saline; n=10）, EET group （treated with 11,12-EET; n=10） and EET inhibitor 14,15-epoxyeicosa-5（Z）-enoic acid （EEZE） group （n=10）. Normal C57BL/6Cnc mice （n=10） treated with saline served as control. Protein expression of CYP2J2 （one of CYP450 epoxygenases） and hypoxia-inducible factor-1α （HIF-1α） was measured by Western blot. Vessel-like structure was detected by immunofluorescence staining. The serum levels of insulin, tumor necrosis factor-α （TNF-α）, interleukin （IL）-1β, IL-6 and monocyte chemoattractant protein-1 （MCP-1） were measured by ELISA. RESULTS In obese mice, homeostasis model assessment of insulin resistance （HOMA-IR） values were increased, the protein level of CYP2J2 was reduced, and the protein level of HIF-1α was increased in adipose tissues as compared with the controls （P<0.05）. The serum levels of MCP-1, IL-1β, IL-6 and TNF-α were also significantly increased in obese mice （P<0.05）. After treatment with 11, 12-EET, the HOMA-IR values were decreased compared with vehicle-treated obese mice, HIF-1α expression levels were decreased in the adipose tissue, and the serum levels of MCP-1, IL-1β, IL-6 and TNF-α were reduced （P<0.05）. Immunohistochemical results of adipose tissue from vehicle-treated obese mice showed a marked decrease in vessel-like structures （CD31-positive） compared with normal control mice （P<0.05）. EET treatment significantly increased the newly formed vessel-like structures in the visceral adipose tissues of obese mice as compared with vehicle-treated obese mice （P<0.05）. CONCLUSION High-fat diet-induced obesity and insulin resistance are closely related to the CYP450 pathway. Exogenous EETs effectively decrease obesity-induced insulin resistance possibly through pro-angiogenesis and attenuation of hypoxia and inflammation.     

Progress in roles of MNX1 antisense RNA 1 in malignant tumor

MNX1 antisense RNA 1 （MNX1-AS1） is a newly discovered long non-coding RNA （lncRNA）, which is highly dysregulated in various carcinomas and its expression level is closely related to the overall survival and prognosis of patients. MNX1-AS1 regulates the occurrence and development of carcinomas by endogenous competitive adsorption of miRNA, regulating cell cycle, inducing epithelial mesenchymal transformation and activating multiple signaling pathways. The in-depth study of the carcinogenesis of MNX1-AS1 is useful for the early diagnosis, targeted therapy and prognostic assessment of relevant carcinomas. This article reviews the roles of MNX1-AS1 in malignant tumor.     

辣椒新品种平椒3号的选育

平椒3号是以自交系99101为母本,自交系9949为父本配制的早熟一代杂种。果实羊角形,青熟果深绿色,果纵径17.7cm,果肩径3.0cm,果肉厚0.25cm,平均单果质量29.5g,肉质脆嫩,辣味浓且有香味,商品性好。田间对疫病、炭疽病、病毒病的抗性明显优于猪大肠。露地栽培每667m2青椒产量为3700kg左右,适宜西北地区及四川、山西、湖南等嗜辣地区露地栽培。     

植物源杀菌剂大黄素甲醚对烟草病毒病的防治效果

为明确大黄素甲醚对烟草病毒病的防治效果,最佳施药时期及施用量。以0.1%大黄素甲醚水剂为试验药剂,8%宁南霉素水剂和20%盐酸吗啉胍可湿性粉剂为对照,开展田间小区试验。结果表明,0.1%大黄素甲醚水剂于移栽定植期施药防效最好,高浓度达62.81%,中、高浓度即制剂量80克/亩、100克/亩的防效与对照药剂均无显著性差异。田间观察发现大黄素甲醚对烟草有一定促生长作用。     

不同区域及栽培密度下耐密植芝麻新品种豫黑芝1号产量及产量相关性状变化分析

芝麻是我国重要的特色优质油料作物。提高芝麻单产水平是实现我国粮油安全生产的重要举措。为建立芝麻高产高效栽培模式,本研究以耐密植黑芝麻新品种豫黑芝1号为试验材料,在3个产区(漯河、信阳和三门峡),开展了不同栽培密度(12万株/hm2、15万株/hm2、18万株/hm2、21万株/hm2)下豫黑芝1号产量及产量相关性状变化分析。结果显示,豫黑芝1号在不同产区的生育期差异明显;种植密度对豫黑芝1号的株高、有效果节数、主茎果轴长、单株蒴果数、千粒重和单株产量等性状变化程度不同;对小区产量及单株产量影响显著。在21万株/hm2密度下,豫黑芝1号单产水平最高达到1463.33 kg/hm2(三门峡)。研究为芝麻高产高效生产提供了技术支持。     

山西苹果茎痘病毒遗传多样性分析

苹果茎痘病毒是苹果安全生产的巨大威胁,严重影响苹果产量和质量。探讨苹果茎痘病毒(Apple stem pitting virus, ASPV)在山西省的分布、变异和多样性,可为制定可持续的病毒管理策略提供理论依据。从山西省11个地区采集苹果叶片样品进行RT-PCR检测。结合在线数据库,利用相关软件对文中研究获得的序列进行系统发育、遗传多样性、重组以及种群遗传分化等分析。RT-PCR检测结果显示从山西省11个地区采集的409份苹果叶片样品中,ASPV的检出率为36.33%(临漪)至55.37%(平遥)。选择15个ASPV检测为阳性的样品,分别克隆其CP基因并与22个不同的ASPV分离物进行核苷酸和氨基酸一致性分析,发现37个ASPV分离物的核酸一致率为70.27～96.31%,氨基酸一致率为37.04～96.64%,其中,Shanxi-2与JX673826.1的核酸一致率最高,为90.86%, Shanxi-2与MN617853.1的核酸一致率最低,为70.40%。基于CP的进化树分析结果显示37个ASPV分离物被划分为2个进化群体。遗传多样性分析结果表明ASPV种群发生了收缩,且在两个...     

烟草丛顶病毒RNA依赖RNA聚合酶(RdRp)介导的体外复制体系

通过重叠PCR扩增得到烟草丛顶病毒(Tobacco bushy top virus,TBTV)中国分离物RdRp的编码序列,构建以pMALC2X为基础载体的原核表达载体pMAL-TB-RdRp。0.5 mM IPTG诱导可特异性表达分子量约为120 kDa的MBP-RdRp融合蛋白。温度梯度实验显示,18℃下诱导表达的MBP-RdRp融合蛋白的可溶性比例较高,约17%;经亲和层析纯化的MBPRdRp可特异性识别TBTV正链和负链的3'末端序列,催化体外复制;对正负链的3'末端的体外复制效率存在差别,识别负链3'末端的体外复制效率明显高于正链3'末端。本研究创建的TBTV RdRp介导的体外复制体系为进一步研究TBTV基因组复制调控奠定了基础。
