Three new aspochracin-type cyclic tripeptides, sclerotiotides M–O (1–3), together with three known analogues, sclerotiotide L (4), sclerotiotide F (5), and sclerotiotide B (6), were obtained from the ethyl acetate extract of the fungus Aspergillus insulicola HDN151418, which was isolated from an unidentified Antarctica sponge. Spectroscopic and chemical approaches were used to elucidate their structures. The absolute configuration of the side chain in compound 4 was elucidated for the first time. Compounds 1 and 2 showed broad antimicrobial activity against a panel of pathogenic strains, including Bacillus cereus, Proteus species, Mycobacterium phlei, Bacillus subtilis, Vibrio parahemolyticus, Edwardsiella tarda, MRCNS, and MRSA, with MIC values ranging from 1.56 to 25.0 µM. 相似文献

AIM: To investigate the role of cAMP and cGMP in the regulation of bFGF gene expression of rat cardiomyocytes. METHODS: 8-Bromo-cAMP and sodium nitroprusside were added into the media to raise the concentrations of cAMP and cGMP of cultured cardiomyocytes, in situ hybridization and immunohistochemistry methods were used to identify the mRNA and protein expression of bFGF. The amount of bFGF mRNA and protein expression were detected by computor imaging analysis system MIAS exe. RESULTS: At 24 h after adding 8-Bromo-cAMP(0 1 mmol/L), bFGF mRNA and protein expression elevated significantly , at 48 h and 72 h it still expressed higher than that of control group( P< 0.05). But after adding sodium nitroprusside (1 mmol/L),bFGF expression decreased significantly,and especially at 48 h. CONCLUSION: The regulation of bFGF expression of cardiomyocytes is related to cAMP signal pathway. 相似文献

In the Mediterranean, blue whiting, Micromesistius poutassou, constitutes a traditional fisheries resource. Over several decades, blue whiting landings in the Catalan coast (northwestern Mediterranean) have displayed cyclical variations, of c. 6 yrs, slightly decreased to five in the last two decades, as shown through wavelet analysis. These fluctuations have persisted under very different levels of fishing effort. This study evaluates the hypothesis that deep‐water formation in the adjacent Gulf of Lions, and the enhanced primary productivity related to it, determines recruitment strength in blue whiting that results ultimately in the observed periodicity of the blue whiting landings. The link between landings and environmental drivers was explored using lagged cross‐correlations, with 0‐ and 1‐yr lag. The variables considered included large‐scale indices [North Atlantic Oscillation (NAO) and the Atlantic Multi‐decadal Oscillation (AMO)], Mediterranean climate indices [MO and Western Mediterranean Oscillation (WeMO)], and variables defining the local environmental conditions in the northwestern Mediterranean (sea‐air heat flux, winter air temperature anomaly and Rhône river runoff). Significant correlations were only found between landings (1961–2011) and sea‐air heat flux, which is generally taken as an indicator of processes of deep water convection, at 0 and 1‐yr lag. These results suggest that the observed fluctuations in blue whiting landings respond to oceanographic processes taking place in the Gulf of Lions. 相似文献

参考Saborio和Soto创建的名为蛋白质错误折叠循环扩增(protein misfolding cyclic amplification,PMCA)的方法建立一种22L毒株系朊蛋白错误折叠循环扩增方法,以便研究22L毒株系异常朊蛋白(scrapie PrP,PrPSc)的合成机制。在阴离子去污剂十二烷基硫酸钠(SDS)存在的条件下,这种方法可以通过超声降解从而重复扩增产物,并在仓鼠脑匀浆中快速有效的模仿体内朊病毒PrPSc的复制,因此可以在几个小时内高浓度扩增PrPSc。已有试验结果发现,PrPSc体外转化率在使用粗提的脑组织匀浆时比使用纯化重组PrP更高,这个发现暗示着高效率的PrPSc转化可能还需要探索其他未知的宿主因素。研究结果表明,建立的这种改良的PMCA技术可用于研究PrPSc合成的生物化学机制,以此来进行诊断与治疗方法的研究。 相似文献
