Cyanobacteria are common members of the freshwater microbiota in lakes and drinking water reservoirs, and are responsible for several cases of human intoxications in Brazil. Pseudanabaena galeata and Geitlerinema splendidum are examples of the toxic species that are very frequently found in reservoirs in Sao Paulo, which is the most densely populated area in Brazil. In the search for toxic strains collected from water reservoirs and maintained in the Cyanobacterial Culture Collection (CCIBt) of the Institute of Botany of Brazil, the acetic acid extracts (AE) of P. galeata CCIBt 3082 and G. splendidum CCIBt 3223 were analyzed by planar chromatography, which indicated the absence of cyanotoxins. Animal tests were then carried out, and both extracts were found to induce toxic effects in mice when administered intraperitoneally. The present study aimed to investigate whether the oral ingestion of the above mentioned cyanobacteria extracts would also induce toxic effects in mice. Necropsy and histopathological studies were conducted using tissue samples from the animals, which were euthanized one week after the administration of the extracts. The AE of P. galeata did not cause death but did induce transient symptoms, including eyebrow ptosis, straub tail, and pain. The euthanized animals presented hemorrhage in the liver, whereas the histological analysis showed disorganization of the hepatic parenchyma, necrosis, hyperemia, and proximity of the centrilobular vein in the liver. In addition, alterations in the convoluted tubules of the kidneys were observed, and the lungs were unaffected. The AE of G. splendidum caused only one death, and induced transient symptoms, such as dyspnea, paralysis, and pain, in the other mice. The necropsy of the euthanized mice showed hemorrhage in the lungs and liver. The lungs presented hemorrhagic focuses, alveolar collapse, and granulomatous foci. The liver presented hemorrhagic and enlarged sinusoids, hyperemia, proximity of the centrilobular vein, and disorganization of the hepatic parenchyma. Some areas also exhibited an inflammatory infiltrate and calcified tissue inside blood vessels. Necrosis and rupture of the convoluted tubule cells were observed in the kidneys. Further analysis of the both extracts indicated the lack of hemolytic activity, and the presence of two unknown anti-AChE substances in the AE of G. splendidum. Thus, P. galeata and G. splendidum are producers of novel toxins that affect mammals when administered orally.  相似文献

The oceans remain a major source of natural compounds with potential in pharmacology. In particular, during the last few decades, marine cyanobacteria have been in focus as producers of interesting bioactive compounds, especially for the treatment of cancer. In this study, the anticancer potential of extracts from twenty eight marine cyanobacteria strains, belonging to the underexplored picoplanktonic genera, Cyanobium, Synechocystis and Synechococcus, and the filamentous genera, Nodosilinea, Leptolyngbya, Pseudanabaena and Romeria, were assessed in eight human tumor cell lines. First, a crude extract was obtained by dichloromethane:methanol extraction, and from it, three fractions were separated in a Si column chromatography. The crude extract and fractions were tested in eight human cancer cell lines for cell viability/toxicity, accessed with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) and lactic dehydrogenase release (LDH) assays. Eight point nine percent of the strains revealed strong cytotoxicity; 17.8% showed moderate cytotoxicity, and 14.3% assays showed low toxicity. The results obtained revealed that the studied genera of marine cyanobacteria are a promising source of novel compounds with potential anticancer activity and highlight the interest in also exploring the smaller filamentous and picoplanktonic genera of cyanobacteria.  相似文献

Lyngbyatoxin A from the marine cyanobacterium Moorea producens (formerly Lyngbya majuscula) is known as the causative agent of “swimmer’s itch” with its highly inflammatory effect. A new toxic compound was isolated along with lyngbyatoxin A from an ethyl acetate extract of M. producens collected from Hawaii. Analyses of HR-ESI-MS and NMR spectroscopies revealed the isolated compound had the same planar structure with that of lyngbyatoxin A. The results of optical rotation and CD spectra indicated that the compound was a new lyngbyatoxin A derivative, 12-epi-lyngbyatoxin A (1). While 12-epi-lyngbyatoxin A showed comparable toxicities with lyngbyatoxin A in cytotoxicity and crustacean lethality tests, it showed more than 100 times lower affinity for protein kinase Cδ (PKCδ) using the PKCδ-C1B peptide when compared to lyngbyatoxin A.  相似文献

In this study, we investigated forty cyanobacterial isolates from biofilms, gastropods, brackish water and symbiotic lichen habitats. Their aqueous and organic extracts were used to screen for apoptosis-inducing activity against acute myeloid leukemia cells. A total of 28 extracts showed cytotoxicity against rat acute myeloid leukemia (IPC-81) cells. The design of the screen made it possible to eliminate known toxins, such as microcystins and nodularin, or known metabolites with anti-leukemic activity, such as adenosine and its analogs. A cytotoxicity test on human embryonic kidney (HEK293T) fibroblasts indicated that 21 of the 28 extracts containing anti-acute myeloid leukemia (AML) activity showed selectivity in favor of leukemia cells. Extracts L26-O and L30-O were able to partly overcome the chemotherapy resistance induced by the oncogenic protein Bcl-2, whereas extract L1-O overcame protection from the deletion of the tumor suppressor protein p53. In conclusion, cyanobacteria are a prolific resource for anti-leukemia compounds that have potential for pharmaceutical applications. Based on the variety of cellular responses, we also conclude that the different anti-leukemic compounds in the cyanobacterial extracts target different elements of the death machinery of mammalian cells.  相似文献

Marine pelagic diazotrophic cyanobacteria of the genus Trichodesmium (Oscillatoriales) are widespread throughout the tropics and subtropics, and are particularly common in the waters of New Caledonia. Blooms of Trichodesmium are suspected to be a potential source of toxins in the ciguatera food chain and were previously reported to contain several types of paralyzing toxins. The toxicity of water-soluble extracts of Trichodesmium spp. were analyzed by mouse bioassay and Neuroblastoma assay and their toxic compounds characterized using liquid chromatography coupled with tandem mass spectrometry techniques. Here, we report the first identification of palytoxin and one of its derivatives, 42-hydroxy-palytoxin, in field samples of Trichodesmium collected in the New Caledonian lagoon. The possible role played by Trichodesmium blooms in the development of clupeotoxism, this human intoxication following the ingestion of plankton-eating fish and classically associated with Ostreopsis blooms, is also discussed.  相似文献

Dust emission to the atmosphere from wind‐eroded soils has many environmental impacts, including soil degradation and air pollution. Various agricultural land uses alter the topsoil properties and thus affect dust particle characteristics as well as loading of biological components into the air. In the present work, the richness and abundance of bacterial communities in topsoils of semiarid loess that are associated with dust emissions were studied by high throughput sequencing methods and were found to be affected by land uses: conventional agriculture, organic agriculture alternating with grazing, uncontrolled grazing activities and natural nondisturbed soil. Moreover, bacterial diversity was shown to be influenced by the contents of sand, CaCO3 and particulate matter in the topsoil. Of all bacteria taxa detected, cyanobacteria were found to be most strongly influenced by land use: Natural and grazing lands were highly abundant with cyanobacterial reads (about 33%), whereas conventional agriculture lands and organic agriculture lands alternating with grazing contained only 7% cyanobacteria. When examining macro‐aggregates in two soils (natural and grazing), approximately 44% of reads were found to be affiliated to cyanobacteria, whereas in micro‐aggregates, their concentration decreased to about 11%. Intensive agricultural use leads to a reduction in soil aggregation and significantly decreases cyanobacteria abundance, in turn increasing dust emission potential and loss of topsoil materials to the atmosphere. Atmospheric nitrogen that is fixed by associative cyanobacteria can be released into the surrounding soil environment providing a key source of N for arctic ecosystems. Yet, little is known about nitrogen fixation by Biological Soil Crusts (BSCs) within hummock-hollow complexes that are typical of many arctic environments. In this study, we examined spatial and temporal patterns in N2-fixation, dinitrogenase reductase (nifH) gene abundance and release of N in a low arctic hummock-hollow ecosystem. The impacts of cyanobacteria on N status in soil were evaluated by assessing soil nitrogen in relation to the cyanobacterial associations found on Hummock and Hollow BSCs. In addition, potential P limitation of N2-fixation by cyanobacteria was assessed for Hummock and Hollow BSCs. The tops of hummocks and the bottoms of hollows were areas of high N2-fixation, whereas minimal N2-fixation occurred on the sides of hummock-hollow complexes. Compared with Hummock BSCs, Hollow BSCs had a higher mean growing season N2-fixation rate, a higher mean growing season nifH abundance, a higher mean total %N and δ15N values closer to that of atmospheric N2. Soil N status was linked to rates of N2-fixation by BSCs indicating that these N2-fixing associations act as important point sources of soil N in this low arctic ecosystem. Over the course of a growing season temporal variation in N2-fixation and nifH abundance were weakly linked suggesting that N2-fixation was carried out by complex communities of diazotrophic microorganisms and that factors such as nutrient availability may limit N2-fixation to a greater extent than nifH abundance.  相似文献

  • 1. In Australia, the carp Cyprinus carpio L. is regarded as a threat to the native fish and the aquatic environment. In recent years, Botany Wetlands, a significant coastal wetland in the Sydney region, has been invaded by the undesirable cyprinids, carp and goldfish (Carrasius auratus L.).
  • 2. In 1996 a cyprinid removal programme commenced at Botany Wetlands with the objective of managing the invasive species, increasing fish diversity, reducing cyanobacterial blooms and hence enhancing the aquatic habitat. Using electrofishing and gill netting, 4073 carp and 261 goldfish, amounting to 10 117 kg of cyprinid biomass were removed between 1996 and 2004. The captured carp ranged from 60 to 835 mm. Males matured before females. Carp between 350 and 680 mm in length grew at the rate of 1.66±0.38 g day?1.
  • 3. The success of the programme was monitored by assessing four indicators related to carp populations and two related to habitat. The former included the pattern of length–frequency distribution, mean weight per size class, condition factor (CF) and the catch per unit effort (CPUE), and the latter the Secchi disc transparency and cyanobacterial counts. After 8 yr of removal, the plots of carp length–frequency distribution flattened, CF decreased from 2.86 to 1.82 and CPUE decreased from 97 kg day?1 to 50 kg day?1. A 10‐fold decrease occurred in cyanobacterial counts and the Secchi disc transparency increased by 20%. More than 20 000 fingerlings of native Australian bass Macquaria novemaculeata (S.) have been introduced to the wetlands, increasing the potential for juvenile carp predation and biodiversity.
