本文筛选出了六株纤维素降解菌,其中菌株f8在稻草固态发酵培养基中产CMC酶活和FPA酶活最大分别为260.32 U和18.90 U,经初步鉴定为康宁木霉。菌株f3既能降解纤维素,又能产油脂,且在PDA液体培养基上油脂产率达到25%。经初步鉴定为绿色木霉。 相似文献

木材细胞通道畅通与否,直接关系到难干材的干燥、难浸渍防腐剂、阻燃剂木材的处理。开启木材细胞液体流动通道主要从纹孔膜入手,作者在前人研究基础上探讨了木材细胞液体移动形式及细胞通道开启的途径,归纳了3种开启木材细胞通道方式,一是采用化学处理,二是生物处理,三是借助外力(机械力、蒸汽力、冰冻力)等方式迫使木材细胞壁纹孔膜破裂,从而疏通或扩大细胞液体流动通道。阐述了每一细胞通道开启方式的适用范围以及实际操作中面临的问题,以便于能更好的运用到木材干燥、防腐、阻燃、染色等木材加工领域。 相似文献

目的:研究大肠癌p53的表达与病理、增殖细胞核抗原(PCNA)、癌胚抗原(CEA)及p53、PC-NA、CEA与淋巴结转移的关系。方法:应用链霉菌素-生物素(SP)免疫组化法,观察44例经病理确诊的大肠癌石腊标本中的p53表达、PCNA的阳性率和CEA的表达型式。结果:大肠癌p53阳性表达率为52.3%;大肠癌p53阳性表达与性别、年龄及肿瘤的部位、分化程度和湿润深度无关(P>0.05),p53阳性表达者其淋巴结转移率较阴性者高(P<0.05);p53阳性表达及有淋巴结转移者其细胞增殖活性分别较p53阴性表达及无淋巴结转移者高(P<0.05);p53阳性表达及有淋巴结转移者其CEA表型均以胞浆型和间质型为主(P<0.05)。结论:检测p53和PCNA表达及CEA表型对判断大肠癌的恶性程度、预测其林巴结转移趋势和预后及指导临床治疗有重要价值。 相似文献

Based on their composition, marine algae, and namely red seaweeds, are good potential functional foods. Intestinal mucosal barrier function refers to the capacity of the intestine to provide adequate containment of luminal microorganisms and molecules. Here, we will first outline the component of seaweeds and will summarize the effects of these on the regulation of mucosal barrier function. Special attention will be paid to unique components of red seaweeds: proteins and derived peptides (e.g., phycobiliproteins, glycoproteins that contain “cellulose binding domains”, phycolectins and the related mycosporine-like amino acids) together with polysaccharides (e.g., floridean starch and sulfated galactans, such as carrageenans, agarans and “dl-hybrid”) and minerals. These compounds have been shown to exert prebiotic effects, to regulate intestinal epithelial cell, macrophage and lymphocyte proliferation and differentiation and to modulate the immune response. Molecular mechanisms of action of peptides and polysaccharides are starting to be elucidated, and evidence indicating the involvement of epidermal growth factor receptor (EGFR), insulin-like growth factor receptor (IGFR), Toll-like receptors (TLR) and signal transduction pathways mediated by protein kinase B (PKB or AKT), nuclear factor-κB (NF-κB) and mitogen activated protein kinases (MAPK) will also be summarized. The need for further research is clear, but in vivo experiments point to an overall antiinflammatory effect of these algae, indicating that they can reinforce membrane barrier function. 相似文献

Dicathais orbita is a large Australian marine gastropod known to produce bioactive compounds with anticancer properties. In this research, we used bioassay guided fractionation from the egg mass extract of D. orbita using flash column chromatography and identified fractions containing tyrindoleninone and 6-bromoisatin as the most active against colon cancer cells HT29 and Caco-2. Liquid chromatography coupled with mass spectrometry (LCMS) and 1H NMR were used to characterize the purity and chemical composition of the isolated compounds. An MTT assay was used to determine effects on cell viability. Necrosis and apoptosis induction using caspase/LDH assay and flow cytometry (PI/Annexin-V) and cell cycle analysis were also investigated. Our results show that semi-purified 6-bromoisatin had the highest anti-cancer activity by inhibiting cell viability (IC50 = ~100 µM) and increasing caspase 3/7 activity in both of the cell lines at low concentration. The fraction containing 6-bromoisatin induced 77.6% apoptosis and arrested 25.7% of the cells in G2/M phase of cell cycle in HT29 cells. Tyrindoleninone was less potent but significantly decreased the viability of HT29 cells at IC50 = 390 µM and induced apoptosis at 195 µM by increasing caspase 3/7 activity in these cells. This research will facilitate the development of these molluscan natural products as novel complementary medicines for colorectal cancer. 相似文献

The pattern of cytokeratin proteins in the epidermal cells of loach was studied by immunotechniques and partial separation of the epidermal cells. Two monoclonal antibodies, namely 8F7 and 1C45, against the cytokeratin proteins of the loach epidermis were prepared. these two monoclonal antibodies exhibit distinctive results in immunohistochemical staining. The 8F7 monoclonal antibody stains mainly with the epithelial cells, while the 1C45 monoclonal antibody stains specifically with the club cells. The pattern of cytokeratin proteins in the club cells and the epithelial cells of various epidermal layers was further determined by partial separation of these cells. Immunoblotting analysis of the cell fractions confirms the cytokeratin proteins to be differentially expressed in the club cells and the epithelial cells. However, the cytokeratin proteins expressed in the epithelial cells of the basal, middle and outer layers are same. The results indicate that differentiation of the epithelial cells seems limited during their translocation from basal to upper layers, but in those cells that do differentiate into club cells, the cytokeratin pattern changes. 相似文献

A new cell line named CCF‐K104 predominantly consisting of fibroblastic cells showed optimal growth at temperatures from 25 °C to 30 °C. Serial morphological changes in the cells induced by Cyprinid herpesvirus 3 (CyHV‐3) included cytoplasmic vacuolar formation, cell rounding and detachment. Mature virions were purified from CyHV‐3‐infected CCF‐K104 cells by sucrose gradient ultracentrifugation and had a typical herpesvirus structure on electron microscopy. Infectious CyHV‐3 was produced stably in CCF‐K104 cells over 30 viral passages. Our findings showed that CCF‐K104 is a useful cell line for isolation and productive replication of CyHV‐3. A temperature shift from 25 °C to 15 °C or 35 °C did not allow serial morphological changes as observed at 25 °C for 14 days. Under the same conditions, real‐time PCR showed that CyHV‐3 was present with low viral DNA loads, suggesting that CyHV‐3 may establish latent infection in CCF‐K104 cells. Amplification of the left and right terminal repeat sequences of the CyHV‐3 genome arranged in a head‐to‐tail manner was detected by nested PCR following an upshift in temperature from 25 °C to 35 °C. Bacteria inhabiting the xylem of Norway spruce (Picea abies (L.) H. Karsten) were investigated. The trees had been wounded and artificially inoculated with fungi and bacteria obtained from wounds of naturally infected spruce. One and five growing seasons after inoculation the Gram‐negative bacterial population present in the stem of inoculated trees were analysed. The Gram‐negative bacteria isolated from the trees were identified on the basis of morphological, biochemical and physiological tests and whole‐cell fatty acid composition. The predominant strains were Enterobacteriaceae fermenter strains (E. agglomerans or E. sakazakii), fluorescent and yellow pigmented Pseudomonas, Acinetobacter and Moraxella spp. All Gram‐positive bacteria were Bacillus species. The Gram‐negative bacteria of Norway spruce differed from the Gram‐positive species in possessing stronger lipolytic activity and in their ability to utilize pine resins for growth. Gram‐positive bacteria were generally able to utilise cellulose and hemicellulose, whereas among the Gram‐negative bacteria only one xylanolytic (yellow Pseudomonas) strain was found. 相似文献
