大豆胞囊线虫(soybean cyst nematode,SCN)严重危害世界大豆生产,Rhg4(resistance to Heterodera glycines 4)是控制大豆SCN抗性的2个主效位点之一。本研究针对Rhg4(Gm SHMT)上的2个单核苷酸多态性(single nucleotide polymorphisms,SNP)位点开发了快速、经济、简便易行的CAPS(Rhg4-389)和d CAPS标记(Rhg4-1165),并用开发的2个标记鉴定了以大豆胞囊线虫应用核心种质为主的193份代表性抗感种质。结果表明,Rhg4-389和Rhg4-1165位点间存在显著连锁不平衡(P=0.0001,r2=0.87),可形成4种单倍型。Rhg4-389-G/Rhg4-1165-T和Rhg4-389-C/Rhg4-1165-A为优势单倍型,稀有单倍型Rhg4-389-G/Rhg4-1165-A和Rhg4-389-C/Rhg4-1165-T是在中国抗源中新发现的单倍型。结合193份种质对SCN 3号小种抗性鉴定分析发现,Rhg4-389-G和Rhg4-1165-T主要存在于抗病种质,它们形成的单倍型对抗病种质鉴定效率可达94.1%。本研究开发了可用于辅助大豆SCN抗性鉴定且方便育种家利用的CAPS/d CAPS标记,且用其摸清了应用核心种质等重要抗源在Rhg4位点的本底,为育种家有效利用这些优异抗源提供了重要信息。 相似文献

西瓜抗枯萎病育种的分子辅助选择 总被引:1,自引:1,他引:1
利用中国国内公开发表的西瓜抗枯萎病生理小种1基因紧密连锁标记4451_fon,首次对抗枯萎病育种创新的二倍体和四倍体种质进行分子鉴定。结果表明,全部二倍体单系和除编号D16、D17外的四倍体单系均表现为抗病基因型,判定为抗病材料,与人工接种鉴定结果完全一致;分子鉴定还表明,这些抗病单系为抗病纯合体,在以后的自交纯化过程中抗性基因不再分离和丢失,因此不必继续对其自交后代进行抗性选择。研究结果认为dCAPS标记4451_fon可以作为西瓜抗枯萎病分子辅助选择(molecular marker assisted selection,MAS)的重要工具,能够实现提高选择效率、加速育种进程的目的。 相似文献

白粉病是豌豆的主要病害之一,在全球范围内引起严重经济损失。防治豌豆白粉病最有效、经济和环境友好的方法是利用抗病品种。迄今,2个隐性抗白粉病基因er1、er2和一个显性抗白粉病基因Er3已在豌豆中被鉴定,其中er1基因在世界上被广泛应用于抗病品种培育。er1基因隶属MLO基因家族,其抗性由豌豆Ps MLO1基因座位功能丧失产生。X9002是甘肃省农业科学院培育的一个半无叶(afila)抗白粉病豌豆品系。本研究对X9002抗白粉病基因进行鉴定,开发用于抗白粉病基因选择的分子标记。遗传分析表明X9002对白粉病抗性由1个隐性单基因控制,SSR标记将该基因定位到豌豆第VI连锁群er1座位区域,标记AD60和c5DNAmet与其连锁。Ps MLO1基因序列分析发现,X9002存在一个未知大小和身份的片段插入,该类型突变也发生在含有er1-2等位基因的豌豆品种Stratagem和Franklin,表明X9002抗白粉病基因为er1-2。一个鉴定er1-2等位基因的功能标记Ps MLO1-650被开发,该标记为互引相标记,仅在感病植株中扩增,可以有效用于分子辅助选择。 相似文献

Pea bacterial blight (Pseudomonas syringae pv. pisi) has long been known to be present in pea growing areas of Spain and to cause serious crop losses, although there is no published
record of its occurrence. A collection of 16 isolates from a winter pea trial in Valladolid in 1991 which were shown in this
study to be P.s. pv. pisi races 4 and 6 would appear to be the first published record of the disease in Spain. This occurrence of races 4 and 6 is
the same as reported for winter-sown peas in the South of France. Ten Pisum sativum landraces from different geographical areas of Spain and considered to be representative of the traditional pea crop, were
tested for resistance to seven races of P.s. pv. pisi. Seedlings of each landrace were stem inoculated with the type strain of each race in a glasshouse. Resistance exhibited
by the different landraces mainly conformed to those previously described in pea cultivars indicating various combinations
of the main resistance genes: R3, R2+4, R3+4 and R2+3+4. R3 was the most frequent R gene, being present in all landraces.
R4 was present in four and R2 in three of the landraces tested. Variation for resistance within landraces was limited except
for landrace accessions ZP-0102, ZP-0109 and ZP-0137 which also showed variation for morphological traits. The resistance
responses of landrace ZP-0109 were difficult to interpret, but suggested a genetic mixture with some evidence of less well
documented R genes, R5 and/or R6, and possibly some unknown resistance to race 6.

The use of bulked leaf samples from individual plants for amplified fragment length polymorphism (AFLP) analysis was evaluated
as a tool for assessment of genetic diversity in white clover (Trifolium repens L.). Bulking of leaf samples produced slightly simpler AFLP profiles compared to the combined profiles of individual plants
from the same cultivar. Approximately 90% of bands which were present in individual plants were present in bulked samples
of the same cultivar. The majority of those absent were rare bands, shared by less than 25% of individual plants. Replicate
bulk samples gave almost identical banding patterns, demonstrating the robustness of the bulked AFLP technique. Cluster analysis
of AFLP data derived from individual plants resulted in a phenogram similar to that produced from data derived from bulked
samples of the same plants. AFLP analysis of bulked samples detected significant amounts of genetic variability among 52 cultivars
and accessions with genetic similarity values ranging from 0.42 to 0.92. However, cluster analysis of AFLP data only partially
reflected the geographic origin of cultivars and accessions and was not congruent with cluster analysis based on variation
for morphophysiological characters. Bulked AFLP analysis provides a powerful tool for rapid assessment of genetic variability
in white clover and may also be used for cultivar identification.

Globe artichoke is a vegetable native to the Mediterranean basin. Its commercial production is mainly based on perennial cultivation of vegetatively propagated clones, which are highly heterozygous and segregate widely when progeny‐tested. The aim of this study was to measure genetic variation using random amplified polymorphic DNA (RAPD) in five populations of ‘Spinoso sardo’, the most widely grown and economically important cultivar in Sardinia (Italy). Analysis of molecular variance (amova ) gave highly significant differences between populations (28.1% of the total genetic diversity); substantial within‐population variation was detected (71.8% of total genetic diversity). The RAPD results show the need to apply clonal selection to narrow the broad genetic variability within the cultivar and to address important questions on the most suitable strategies for germplasm preservation. 相似文献

Amplified fragment length polymorphism (AFLP) analysis has been used to measure genetic diversity in perennial ryegrass (Lolium perenne L.) and to relate intra- and interpopulation variation to breeding history. Cluster analysis of AFLP data from contrasting
populations showed features consistent with the origins of these varieties. Significant differences in intrapopulation diversity
were detected and partial separation of different cultivars was observed. Restricted base cultivars, derived from small numbers
of foundation clones, were suitable for this type of study, allowing near complete discrimination of closely related cultivars.
Analysis of bulked samples was based on the pooling of genomic DNA from 20 individuals from 6 selected populations. Cluster
analysis of AFLP data from bulked samples produced a phenogram showing relationships consistent with the results of individual
analysis. AFLP profiling provides an important tool for the detection and quantification of genetic variation in perennial
ryegrass.

Summary Interspecific hybrids have been obtained in an otherwise incompatible cross betweenBrassica juncea × Brassica hirta through the in vitro culture of hybrid ovules and ovaries. The best response was observed from ovules and ovaries cultured 10–15 and 5–7 days after pollination respectively on a basal medium supplemented with indoleacetic acid, kinetin and casein hydrolysate. In some cases the basal cut end of the ovaries proliferated to form callus and shoots. The in vitro-derived hybrid seeds varied in their colour, size and shape, and the F1 plants in the field showed a large diversity in their morphological traits. Summary Various wild relatives of pigeonpea,Cajanus cajan, namely some species ofAtylosia andRhynchosia, possess desirable characteristics that could be utilized for effecting genetic improvement of this crop. In total 73 cross combinations among two cultivars ofC. cajan and one accession each of eightAtylosia species and one ofRhynchosia were attempted. Twelve hybrids were obtained. Seven of these were analysed for F1 fertility and their utility for agronomic improvement of theC. cajan. Fertility behaviour of the different F1 hybrids varied and indicated that potential of gene transfer between the two genera,Atylosia andCajanus, was as good as within the genusAtylosia. From F2 and F3 families ofC. cajan × A. scarabaeoides andC. cajan × A. albicans, plants were selected with greater physiological efficiency and agronomic superiority. The prospects of transferring pod borer resistance and higher seed protein content from someAtylosia species to pigeonpea are discussed. 相似文献
