Membranes from house fly heads were tested for the presence of mucarinic acetylcholine receptors using as a probe [3H]quinuclidinyl benzilate ([3H]QNB). Based on the presence of saturable and reversible high-affinity binding of [3H]QNB, which is inhibited by muscarinic drugs, it is suggested that these sites may be muscarinic receptors. However, these putative muscarinic receptors differ in several characteristics from the ones in mammalian brain. They have lower affinities for muscarinic drugs and lower stereoselectivity, a relatively higher affinity for the nicotinic antagonist d-tubocurarine, a lower Hill coefficient for binding of muscarinic antagonists, and a lower concentration relative to α-bungarotoxin binding sites in the same membranes. Also, unlike mammalian muscarinic receptors, they are sensitive to treatments with N-ethylmaleimide and 5,5′-dithiobis(2-nitrobenzoic acid). The effect of reduction of disulfide bonds by dithiothreitol or mercaptoethanol suggests that only the insect receptor has one or more disulfide bonds which are important to binding. On the other hand, the putative muscarinic receptors of both insect and mammalian brains have important SH group(s), whose alkylation by p-chloromercuribenzoate inhibits binding. Also, chlorobenzilate is equally effective in inhibiting [3H]QNB binding to muscarinic putative receptors of house fly and bovine brains.  相似文献

The actions of a range of formamidines have been investigated biochemically and physiologically on octopamine receptors in the locust Schistocerca gregaria. All the formamidines tested [chlordimeform (CDM), demethylchlordimeform (DCDM), amitraz, and UK 16353] mimicked the action of octopamine by (1) increasing the amplitude and relaxation rate of slow motorneurone tension in the extensor-tibiae muscle of the hind leg and (2) changing the levels of cyclic AMP in this muscle. UK 16353 was most effective in changing these parameters, followed by DCDM then amitraz and CDM. The formamidine-induced increase in cyclic AMP levels was reduced or completely blocked by phentolamine, an antagonist of insect octopamine receptors. The time course for the increase in cyclic AMP was followed for 30 min by incubating muscles in 10?5M DCDM. The increase was potentiated by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine. In the presence of this compound, the response peaked within 5 min, before declining to a lower plateau after 12 min. The response to 10?5M CDM was lower and the maximum increase occurred after 7 min, then rapidly declined. Both formamidines increased cyclic AMP in a dose-dependent way with a threshold of between 5 × 10?7 and 10?8M. The results are discused in terms of the relationship between the biochemical and physiological actions of the formamidines in this preparation.  相似文献

The effects of p,p′-DDT and four of its analogs on electrical activity in the central nervous system of the cockroach, Periplaneta americana (L.), were investigated. Cockroaches were injected intraabdominally with an organochlorine compound at LD50 96-hr doses (except for p,p′-DDE). Extracellular recordings were made from the central nervous system at 1 hr, 24 hr, or 3 weeks postinjection. p,p′-DDT, methoxychlor, and p,p′-DDD induced behavioral changes (tremors, jitters, hyperexcitability) and repetitive firing in the central nervous system prior to 1 hr postinjection. By 24 hr postinjection, most behavioral signs of poisoning had disappeared, though repetitive firing could still be readily elicited in the central nervous system. Cockroaches injected with o,p′-DDT, however, usually required about 48 hr before overt signs of poisoning became apparent. Cockroaches treated with p,p′-DDT or o,p′-DDT behaved normally at 3 weeks postinjection but still displayed a significant occurrence of repetitive firing in the central nervous system. A mechanism is proposed to explain how a cockroach might recover behaviorally from a sublethal dose of an organochlorine compound but still display repetitive firing in its central nervous system. A direct “cause and effect” relationship between repetitive firing in the central nervous system and mortality (and external signs of poisoning) is therefore questioned.  相似文献

Host-parasite relationships have been studied by electron microscopy using glutaraldehyde-OsO4-fixed pellets of lymphoid cultures infected in vitro by Theileria annulata and T. parva. Intracellular presence of the parasite resulted in a progressive and marked lymphoblastoid transformation. The schizont stage periodically provoked the formation of, and adopted an intimate association with, cytoplasmic annulate lamellae in the interphase cell. Annulate lamellae developed from the outer nuclear membrane of the host cell by a delamination process and were taken into the cytoplasmic matrix of the schizont by phagotrophy through the cytostome. Schizont nuclei themselves were seen to divide at the prometaphase stage of host cell mitosis, the division being characterized by the development of intranuclear spindle microtubules anchored in spindle pole bodies. A hypothesis is propounded that Theileria parasites, consequent on interiorization, provoke the blastoid transformation and the formation of annulate lamellae through the influence of components of their genomic material on host cell deoxyribonucleic acid (DNA) and that the annulate lamellae represent a species of messenger ribonucleic acid (mRNA) and serve as a monitoring device for the schizont, facilitating the accurate timing of the host cell cyclical events.  相似文献

Development and phenobarbital (PB) induction of microsomal cytochrome P-450, cytochrome P-450 reductase, two epoxidation, and two O-demethylation activities were examined in chronologically timed populations of insecticide-susceptible (NAIDM) and -resistant (Rutgers) house flies. Measurements of these enzymes started with the pharate adult stage and ended 5 days following eclosion. Untreated insects of both strains had comparable reductase levels, whereas cytochrome P-450 and associated monooxygenase activities were 1.5-fold or more higher in Rutgers. Maximum induction, as well as toxicity, occurred at a lower PB concentration in NAIDM than Rutgers. The drug caused consistently higher increases in enzymes and activities within 12 hr of starting treatment in both strains. When PB was withdrawn from treated flies (both strains) 48 hr after treatment began, specific activities (product min?1 mg protein?1) in all enzymes returned to control values in 24 hr while metabolic capacity (product min?1 insect?1) achieved control values within 48 hr. The changes in turnover numbers (pmol product min?1 pmol P-450?1), in conjunction with the differences in the monooxygenation of the four substrates, suggest that PB treatment induced both a quantitative and qualitative change in NAIDM monooxygenation but only a quantitative change in Rutgers monooxygenation.  相似文献

Alkaline-dissolved crystal δ-endotoxin from Bacillus thuringiensis var. israelensis (serovar H 14) was injected into mice and seven species of insects representing the orders Lepidoptera, Orthoptera, Coleoptera, Hemiptera, and Diptera. High in vivo toxicity, at 1 to 5 ppm (μg toxin/g body wet wt), was observed with mice and some insects, including some that are not sensitive to the toxin when administered orally. Neuromuscular effects were observed when the toxin was injected directly into the body cavity of the test animals. Biochemical studies suggested that different protein fragments within the crystal δ-endotoxin may be responsible for the majority of the mosquito larvacidal activity and the neurotoxic symptoms observed in larvae of Trichoplusia ni.  相似文献
