采用蛋鸡离体外翻肠段培养法,将小肠分为十二指肠、空肠前段、中段、后段和回肠五个部位肠段,洗净外翻。2种二肽培养液(Gly-Gly肽和Gly-L-Leu肽溶液)作为两组对照培养液,每种二肽培养液中添加肽酶抑制剂(Bestatin和Amastatin),作为试验培养液,39.5℃培养25min,每5min取样一次,测定培养液中的二肽含量。结果表明,培养5min后,含Gly-Gly肽的对照和试验培养液中均未能检测到完整Gly-Gly肽。而含Gly-L-Leu肽的对照培养液Gly-L-Leu肽的含量降低到16.75%,试验培养液则下降到43.40%。而试验培养液中Gly-L-Leu肽的含量10min后无显著降低(P>0.05)。各培养时段试验培养液Gly-L-Leu含量均显著高于对照培养液(P<0.05)。由此认为,离体肠囊在培养过程中能够迅速释放肠肽酶及一些生物活性物质,快速水解2种二肽。肽酶抑制剂(Bestatin和Amastatin)能抑制Gly-L-Leu的水解,但不能抑制Gly-Gly的水解。 相似文献

以耐寒性差异较大的杂交狗牙根品种运动百慕大、天堂419和普通狗牙根品种保定狗牙根为试验材料,分析了昼夜温度为适温(30 ℃/25 ℃)、亚适温(18 ℃/12 ℃)、冷害(8 ℃/4 ℃)和冻害(4 ℃/-4 ℃)4种温度处理下,低温对狗牙根生长速率和草坪质量、MDA含量、叶绿素荧光(Fv/Fm)、光合作用(Pn、Gs、Ci、Tr)、H2O2和O 2 · - 含量、抗氧化酶活性(SOD、POD、CAT、APX)、抗氧化酶及耐寒相关基因(CBF1、COR、LEA)表达的影响。结果表明,随着温度的降低,狗牙根草坪质量、生长速率、Fv/Fm和光合作用显著下降,但在耐寒性强的运动百慕大中下降幅度较小;低温下叶片MDA和H2O2含量及O 2 · - 产生速率显著升高,其中在耐寒性弱的保定狗牙根中升高程度更大;狗牙根叶片抗氧化酶活性及抗氧化酶基因的表达水平随着处理温度的下降而显著升高,其中在运动百慕大中更为明显;狗牙根CBF1、COR和LEA基因表达量随着温度的下降而显著升高,尤其是在耐寒狗牙根运动百慕大中更为显著。低温诱导的抗氧化酶和耐寒相关基因上调表达,增强了细胞内的抗氧化防御,有助于狗牙根植株维持较高的光合作用和细胞膜稳定性,延缓叶片的衰老,从而提高了狗牙根的抗寒能力。 相似文献

To support their combined use, the objective of the study was to evaluate the effects of benazepril and pimobendan on serum angiotensin‐converting enzyme (ACE) activity in dogs. A total of 48 healthy beagle dogs were randomized into four groups (n = 12 per group) in a parallel‐group design study: A (control, placebo twice daily (BID)); B (0.5–1.0 mg/kg benazepril once daily (SID) in the morning, placebo in the evening); C (0.25–0.5 mg/kg benazepril BID); D (0.25–0.5 mg/kg benazepril and 0.125–0.25 mg/kg pimobendan, both BID). The test items were administered orally for 15 days. Serum ACE activity was measured on days 1 and 15. Groups B, C and D had significantly lower average serum ACE activity compared to baseline and to the control group, on both days 1 and 15. There were no significant differences in average ACE activity between groups B, C and D. Noninferiority of group C to B was demonstrated. In conclusion, 0.25–0.5 mg/kg benazepril administered BID produced noninferior inhibition of serum ACE activity compared to 0.5–1.0 mg/kg benazepril dosed SID. Pimobendan had no significant effect on benazepril's action on serum ACE activity. The results support the use of benazepril BID in dogs and in combination with pimobendan. 相似文献

This study was to investigate the effects of in ovo feeding (IOF) L‐arginine (Arg) solution on the development of digestive organs, the duodenal mucosa of broiler embryos and hatchlings, and the growth performance of chicks during the first week post‐hatch. A total of 720 fertilized eggs with similar weight were randomly allocated to three groups, consisting of eight replicates of 30 eggs each. Three treatments were arranged as non‐injected control, diluent‐injected (0.75% NaCl solution) group and Arg solution‐injected group containing 1% Arg, dissolved in diluent. At 17.5 days of incubation, 0.6 ml of IOF solution was injected into amniotic fluid of each egg of injected groups. Results showed IOF of Arg solution increased (p < .05) the chick embryo weight at 19 days of incubation; the body weight gain of post‐hatch broilers during 1–7 days; the weights of liver, pancreas, proventriculus and gizzard; the concentrations of duodenal ghrelin, vasoactive intestinal peptide and glucagon‐like peptide 2; and the duodenum mucosal enzyme activities of alkaline phosphatase, maltase, sucrase and inducible nitric oxide synthase of 7‐day‐old post‐hatch broilers compared with other groups. The IOF of Arg solution also increased (p < .05) the villus height (VH) and the ratio of VH to crypt depth (CD) and decreased (p < .05) the CD in duodenum of broiler embryos and post‐hatch hatchlings, except for the CD at 19 days of incubation. In conclusion, IOF of 1% Arg solution into the amnion at 17.5 days of incubation could improve the development of digestive organs, the duodenal morphology, the releasing of gastrointestinal hormones and mucosal enzyme activities of broiler embryos and hatchlings and finally the growth performance of chicks during the first week post‐hatch. Therefore, IOF of appropriate Arg solution could be an effective technology for regulating early nutrition supply and subsequent growth development in poultry industry. 相似文献

Syngamus trachea is a pathogenic tracheal nematode that causes syngamiasis in wild and game birds, especially when birds are managed at high densities. Despite its pathogenic nature, very little is known about its epidemiology and relationship with ambient temperature and humidity. The spatial and temporal modelling of disease was undertaken on two pheasant estates within the South West of England from April 2014 to August 2014. Significant differences between the mean numbers of eggs per gram of soil were identified between pens at both site 1 and site 2 but did not differ significantly between sites. Egg abundance was significantly associated with soil moisture content, with greater egg survival between years in pens with higher average volumetric soil moisture content. Previous years stocking density and pen age were also associated with greater egg survival between years with more eggs being recovered in pens with greater stocking densities, and pens that had been sited longer. The greatest model to explain the variation in the numbers of eggs per gram of soil per pen was a combination of soil moisture content, stocking density and pen age.Larval recovery differed significantly between sites. Larval abundance was significantly and positively associated with temperature and relative humidity at site 1. Similarly, temperature and humidity were also positively and significantly associated with larval abundance at site 2. Rainfall did not influence larval recovery at either site 1 or site 2. The model with the greatest ability to explain larval abundance at both sites, was a combination of temperature, humidity and rainfall. Infection status (positive faecal egg counts) was significantly and positively associated with larval abundance at both sites, but rainfall was only positively associated at site 1. Temperature and humidity were positively associated with infection status at site 2, but not at site 1. The present study highlights the influence of climatic variables on both egg survival and larval abundance, and could therefore be used to develop more targeted treatment strategies around periods of higher disease risk. The frequent use of release pens is a clear factor in the epidemiology of syngamiasis, and it is recommended that pens be rested and/or rotated in order to reduce infection pressure in subsequent flocks. 相似文献

本研究通过野外调查取样与室内试验相结合,连续两年对黄土高原人工灌草系统不同坡向、坡位进行取样,以期对该区土壤种子库的物种组成、密度特征及其物种多样性进行初步了解。研究结果表明:该人工灌草生态系统土壤种子库共有9科,15种,其组成因坡位、坡向以及年份而异;坡向、坡位均显著影响土壤种子库物种多样性指数、丰富度指数与土壤种子库密度(P<0.05),土壤种子库密度为3 218~5 492粒·m-2,总体呈现为阴坡显著高于阳坡(P<0.05),下坡位显著高于上坡位(P<0.05);坡位和坡向均对物种均匀性系数无显著影响;年份对土壤种子库无显著影响。上述研究可为黄土高原人工灌草系统的可持续利用与恢复提供参考依据。 相似文献

‘Yandangxueli’ is a pear cultivar with predominant citric acid in the ripe fruit, different from most of pear cultivars such as ‘Gengtouqing’ in which malic acid is the predominant acid type. It was found that ‘Yandangxueli’ accumulated citric acid for three times against that in ‘Gengtouqing’ at fruit ripening stage. To investigate the mechanism of citric acid accumulation in ‘Yandangxueli’, organic acids content, gene expression and enzyme activity were studied in both cultivars. Five genes, Pp:mtCs, Pp:cyAco, Pp:cyIdh, Pp:mtMdh and Pp:cyMe which encoded citric synthase (CS), cytosolic aconitase (cyACO), NADP-dependent isocitrate dehydrogenase (NADP-IDH), NAD-dependent malate dehydrogenase (NAD-MDH) and NADP-dependent malic enzyme (NADP-ME) respectively, were identified from pear fruit. Their expression profiles and the corresponding enzyme activities were determined throughout fruit development in both cultivars. Results from these enzymes indicated that there were no strict relationship between gene expression, enzyme activity and citric acid accumulation. Expression analysis for two Py:vVAtp genes encoding vacuolar H+-ATPase A subunit and one Py:vVpp gene encoding Vacuolar H+-pyrophosphatase showed that they were all with up-regulated expression at the later development stage of ‘Yandangxueli’ but with down-regulated expression in ‘Gengtouqing’. Therefore, it is concluded that the different ability in citric acid transportation and storage might be involved in the high citric acid content in ‘Yandangxueli’. 相似文献

In Central Europe, various plant species including large-grain legumes and their mixtures are grown as catch crops, particularly between grains harvested early and subsequent summer crops. This article investigates the question of how soil structure in the topsoil is influenced when catch cropping with large-grain legumes (experimental factor A: without catch crop, with catch crop) under different ploughless tillage conditions during catch crop seeding (experimental factor B: deep tillage/25–30 cm, shallow tillage/8–10 cm). Five one-year trials were performed using standard machinery at various sites in Germany. Soil core samples extracted from the topsoil in the spring after catch crop cultivation served to identify air capacity, saturated hydraulic conductivity and precompression stress. The above-ground and below-ground biomass yields of the catch crops were also determined at most of the sites. In addition, the soil compaction risk for the working steps in the experiments was calculated using the REPRO model.The dry matter yield of the catch crops varied considerably between the individual trial sites and years. In particular, high levels of dry matter were able to form in the case of early seeding and a sufficient supply of precipitation. The soil structure was only rarely affected positively by catch crop cultivation, and catch crops did not contribute in the short term to loosening already compacted topsoils. In contrast, mechanical soil stresses caused by driving over the ground and additional working steps used in cultivating catch crops often led to lower air capacity in these treatments. This is consistent with the soil compaction risks calculated using the REPRO model, which were higher in the treatments with catch cropping. Catch crop cultivation also only resulted in improved mechanical stability at one location. The positive effect of deep ploughless tillage on air capacity and saturated hydraulic conductivity, however, became more clearly evident regardless of catch crop cultivation. In order for catch crop cultivation with large-grain legumes to be able to have a favourable impact on soil structure, it is therefore important that cultivating them does not result in any new soil compaction. In the conditions evaluated, deep tillage was more effective at loosening compacted topsoil than growing catch crops. 相似文献
