The wheat-rye 1BL.1RS translocation chromosomes have been used widely around the world in commercial wheat ( Triticum aestivum L.) production because of the presence of several disease resistance genes and a yield enhancement factor on the rye ( Secale cereale L.) chromosome. However, the recent reports of the loss of complete effectiveness of the disease resistance genes on the most commonly used 1BL.1RS chromosome have highlighted the need to seek and deploy additional sources of disease resistance genes. Three new sibling wheat cultivars, 'CN12', 'CN17' and 'CN18', were developed carrying 1RS arms derived from the rye inbred line L155. Genomic in situ hybridization and C-banding analysis revealed that all the three cultivars contained the rye chromosome 1RS arm fused to the wheat 1BL wheat chromosome arm. The three cultivars displayed high yields and high resistance to local powdery mildew and stripe rust pathotypes. Fluorescence in situ hybridization analysis indicated the different structure of 1BL.1RS chromosome between 'CN18' and the other two cultivars. The present study provides a new 1RS resource for wheat improvement.  相似文献

Shotgun survey sequences of flow‐sorted individual rye chromosomes were data mined for the presence of simple sequence repeats (SSRs). For 787,850 putative SSR loci, a total of 358,660 PCR primer pairs could be designed and 51,138 nonredundant SSR marker candidates were evaluated by in silico PCR. Of the 51,138 SSR primer candidates, 1,277 were associated with 1,125 rye gene models. A total of 2,112 of the potential SSR markers were randomly selected to represent about equal numbers for each of the rye chromosomes, and 856 SSRs were assigned to individual rye chromosomes experimentally. Potential transferability of rye SSRs to wheat and barley was of low efficiency with 4.3% (2,189) and 0.4% (223) of rye SSRs predicted to be amplified in wheat and barley, respectively. This data set of rye chromosome‐specific SSR markers will be useful for the specific detection of rye chromatin introgressed into wheat as well as for low‐cost genetic and physical mapping in rye without the need for high‐tech equipment.  相似文献

Dry matter (DM) partitioning into root, leaf, stem, shoot dry weight plant?1 response in four cool season C3-cereals viz. wheat (Triticum aestivum L.), rye (Secale cereale L.), barley (Hordeum vulgare L.) and oats (Avena sativa L.) was investigated at 30, 60 and 90 days after emergence (DAE) under eight nitrogen, phosphorus and potassium (NPK) sources: S1 = 20-20-20, S2 = 20-27-5, S3 = 7-22-8, S4 = 10-10-10-20S, S5 = 11-15-11, S6 = 31-11-11, S7 = 24-8-16, and S8 = 19-6-12 in pot experiment at Dryland Agriculture Institute, West Texas A&;M University, Canyon, Texas, USA during winter 2009-10. A considerable variation in DM partitioning into various plant parts was observed in the four crop species at different growth stages and NPK source. At 30 DAE, 27% of the total DM per plant (TDMPP) was partitioned into roots and 73% into shoots (19% stems + 54% leaf). Only16 % of the TDMPP was partitioned into roots and 84% into shoots (18 % stem + 66 % leaf) at 60 DAE. At 90 DAE, 29% of TDMPP was partitioned into roots and 71 % into shoots (33 % stems + 38 % leaf) at 90 DAE. Percent DM partitioning into stems ranked first (33%) at 90 DAE > at 30 DAE (19%) > at 60 DAE (18 %). With advancement in crops age, DM partitioning into various crop parts increased. The root DM plant?1 (RDMPP) increased from 11.5–722 mg plant?1; stem DM plant?1 (STDMPP) from 8.3–889.0 mg plant?1; leaf DM plant?1 (LDMPP) from 23.1–1031.0 mg plant?1; shoot DM plant?1 (SHDMPP) from 31.3–1921 mg plant?1, and TDMPP increased from 42.9–2693.0 mg plant?1 at 30 and 90 DAE, respectively. Because of the higher N contents in S7 (24:8:16) and S6 (31:11:11) reduced the DM partitioning into various plants parts as well as TDMPP at all three growth stages. The adverse effects of S6 and S7 on DM partitioning was more on oats > rye > wheat > barley. The S4 with 10:10:10 (NPK) and :20S was not toxic at 30 DAE, but at 60 and 90 DAE it became toxic that adversely affected the DM partitioning as well as TDMPP probably may be due its high sulfur (20%) content which lacking in other NPK sources. The DM partitioning to various parts of barley and wheat was more than oats and rye at different growth stages (barley > wheat > rye > oats). Since the DM portioning values were determined on the average of five plants in pot experiment under organic soil at field capacity; in case of field experiments more research is needed on various crop species/varieties under different environmental conditions particularly under moisture stress condition.  相似文献

Field experiments were conducted to evaluate cover crop (none, winter wheat, or winter rye) and pre-emergence (PRE) applications of clomazone plus ethalfluralin alone or with halosulfuron [PRE or post-emergence (POST)] for smooth crabgrass [Digitaria ischaemum (Schreb. ex Schweig) Schreb. ex Muhl.] and redroot pigweed (Amaranthus retroflexus L.) control in no-tillage ‘Aspen’ jack-o-lantern pumpkin (Cucurbita pepo L.). At pumpkin harvest, cover crops had reduced smooth crabgrass density, but not redroot pigweed. Although PRE or POST applications of halosulfuron alone were more effective at reducing redroot pigweed density than clomazone plus ethalfluralin PRE, the combination of halosulfuron plus clomazone and ethalfluralin PRE reduced redroot pigweed density to the greatest extent. Pumpkin yields were not affected by cover crop, although average pumpkin sizes were greater with the inclusion of cover crop residues. Pumpkin fruit sizes and yields were the greatest with clomazone and ethalfluralin PRE in combination with halosulfuron applied PRE or POST. Overall, cover crop had relatively little influence on pumpkin yields compared with herbicide treatments. The addition of halosulfuron to clomazone and ethalfluralin provided greater broadleaf weed control resulting in greater jack-o-lantern pumpkin yields.  相似文献

将黑麦冬牧70幼穗切段和未成熟胚接种在附加不同激素浓度的改良MS培养基上培养,获得了能多次继代培养的愈伤组织,后者在分化培养基上再生植株.2,4-D和6-BA浓度明显影响愈伤组织质量和诱导率,幼嫩幼穗和较小的未成熟胚愈伤组织诱导率高.幼穗切段在改良MS 1.0 mg/L 2,4-D 200 mg/L谷氨酰胺的培养基上愈伤组织诱导率最高,质量较好;未成熟胚在改良MS 2.0 mg/L 2,4-D 0.1 mg/L 6-BA 200 mg/L谷氨酰胺的培养基上愈伤组织诱导率最高,质量最好.继代培养后的愈伤组织转移到附加200 mg/L谷氨酰胺、1.0 mg/L 2,4-D和0.1 mg/L TDZ的培养基上可分化出大量小芽.该体系可用于黑麦冬牧70遗传转化和细胞工程育种研究.  相似文献

在南京地区研究了不同前作(水稻Oryza satica和杂交狼尾草Pennisetum americanum×P. purpureum)对冬牧70黑麦Secale cereale var. Dongmu 70产量、再生情况和营养品质的影响.结果表明:水稻茬种植的冬牧70黑麦较杂交狼尾草茬种植的再生茎蘖数多,株高高,干物质积累多;水稻茬冬牧70黑麦,2次刈割鲜干比也明显比杂交狼尾草茬的高,第2次刈割的茎叶比较杂交狼尾草茬的低.因此,在水稻茬种植冬牧70黑麦产量较高,品质也较好.  相似文献

In CMS (cytoplasmic male sterility)‐based hybrid rye (Secale cereale L.) breeding, effective pollen‐fertility restoration is an essential prerequisite for achieving maximum grain yield on the one hand and for minimizing ergot (Claviceps purpurea) infestation on the other. Restorer genes for the CMS‐inducing ‘Pampa’ cytoplasm derived from landraces collected in Iran and Argentina are used by breeders for achieving this goal. Here, restorer genes from four germplasm sources (‘Altevogt 14160’, ‘IRAN III’, ‘Trenelense’ and ‘Pico Gentario’) were analysed by producing three‐way cross hybrids between an elite CMS single cross and pollinators with and without a given restorer gene. Materials were evaluated on large drilled plots for restorer index (RI), grain yield, plant height and other traits in six environments. In experiment 1, a restorer gene from ‘Altevogt 14160’ was used. Seven pairs of marker‐selected carrier and non‐carrier backcross lines served as pollinators. In experiment 2, the pollinators were 17 backcross line pairs from the other three germplasm sources. These lines were grouped as high (RI > 67%) and low restorers (RI < 30%), respectively, using testcrosses with a highly diagnostic CMS tester. Hybrids carrying an exotic restorer gene suffered from a significant grain yield reduction by 4.4% and 9.4% and were 9.3 and 4.8 cm taller in experiments 1 and 2, respectively. Thousand‐kernel weight was reduced, whereas quality traits were only slightly affected. For all traits, significant genetic variance existed among the testcrosses to the presence vs. absence of a given exotic restorer gene. This offers a chance for the breeder to reduce or ultimately overcome the presently observed performance reductions brought about by exotic restorer genes.  相似文献

Genetic analysis of three semi-dwarf genotypes of rye (Secale cereale L.)—‘Moskowskij Karlik’, ‘Gülzow kurz’ and ‘R 18’, which were shown to be insensitive to applied gibberellic acid (GA3), has been carried out by using a seedling test. It could be demonstrated that all of the three genotypes are carrying recessive alleles for GA-insensitivity. Whereas the alleles of ‘Moskowskij Karlik’ and ‘R 18’ seem to have the same locus on chromosome 5R, the GA-insensitivity of ‘Gülzow kurz’ is governed by a different gene, most probably located on chromosome 7R. The relationship between the genes (alleles) for GA-insensitivity and semi-dwarfness, including the symbolization of the Gai-genes as well as their utilization in rye breeding is discussed.  相似文献

The amount of Telomeric C-heterochromatin was investigated in 20 inbred lines of rye and their open-pollinated source cultivars. In this short communication only the banding pattern of the chromosomes in the lines is presented. The inbred lines had an amount of heterochromatin similar to the source cultivars. Both groups carried some chromosomes with small telomeric C-bands. The individual source cultivars were composed of 2—4 karyotypes having different amounts of telomeric heterochromatin. Most karyotypes were also identified in the respective inbred lines. Results suggest that the in breed in i: process was not related to a reduction of telomeric C-hetrerochromatin in rye chromosomes.  相似文献

根据已知植物抗病基因(resistance gene,Rgene)编码的蛋白质NBS-LRR保守结构设计了10对特异简并引物,对5份黑麦材料的基因组DNA进行抗病基因同源序列克隆,共获得45条抗病基因同源序列(resistance geneanalogs,RGAs)。同源性比较发现,RGAs序列之间的核苷酸相似性是41.9%~99.6%,其中16条具有连续开放阅读框(ORFs)。同时,利用MEGA 3.1将这16条序列与4个已知R基因进行聚类分析,发现有13条序列与RPM1亲缘关系近,2条与Pm3a亲缘关系近。本研究在黑麦中获得的RGAs既可用于筛选黑麦的抗病候选基因,同时也可以作为分子标记,用于作物分子辅助选择育种,且对进一步研究黑麦中NBS-LRR类R基因的起源和遗传进化提供参考和依据。  相似文献
