The discovery of Jenner in 1798 founded the science of immunology and eventually led to smallpox eradication from the earth in 1980 after a world-wide vaccination campaign with vaccinia virus (another poxvirus) and paradoxically, despite the eradication of smallpox, there has been an explosion of interest in vaccinia virus in the eighties. This interest has stemmed in part from the application of molecular genetics to clone and express foreign genes from recombinant vaccinia viruses. Vaccinia is also gaining renewed interest due to bioterrorism.
These recombinant viruses have multiple applications in research and vaccinology and led to the development of vectored vaccines, such as the recombinant vaccinia rabies vaccine used to eliminate rabies in Western Europe and, more recently, in the United States. Secondly, alternative poxvirus vectors, such as avipox viruses, were proved to be even safer and efficacious non-replicating vectors (suiciole vectors) when used in non-avian species. 相似文献
Bovine-leukosis virus (BLV; also termed ‘bovine-leukemia virus’) is a retrovirus that primarily affects lymphoid tissue of dairy and beef cattle. Our objective was to investigate the association between BLV infection and annual value of production (AVP) on dairy herds within the United States, as part of the USDA National Animal Health Monitoring System’s 1996 dairy study. 1006 herds (in 20 states) with at least 30 dairy cows were interviewed during 1996. The agar-gel immunodiffusion test was used to detect serum antibodies to BLV. 10–40 cows from each herd were tested and each tested cow was classified as negative or positive based on results of a single test.
A multivariable regression model was used with the 976 herds with complete data for analysis. When compared to herds with no test-positive cows, herds with test-positive cows produced 218 kg per cow (i.e. 3%) less milk. The average reduction in AVP was $ 59 per cow for test-positive herds relative to test-negative herds. For the dairy industry as a whole, BLV seropositivity was associated with loss to producers of $ 285 million and $ 240 million for consumers. Foot-and-mouth disease virus (FMDV) is an aphthovirus of the family Picornaviridae and the etiological agent of the economically most important animal disease. As a typical picornavirus, FMD virions are nonenveloped particles of icosahedral symmetry and its genome is a single stranded RNA of about 8500 nucleotides and of positive polarity. FMDV RNA is infectious and it replicates via a complementary, minus strand RNA. FMDV RNA replication is error-prone so that viral populations consist of mutant spectra (quasispecies) rather than a defined genomic sequence. Therefore FMDV in nature is genetically and antigenically diverse. This poses important challenges for the diagnosis, prevention and control of FMD. A deeper understanding of FMDV population complexity and evolution has suggested requirements for a new generation of anti-FMD vaccines. This is relevant to the current debate on the adequacy of non-vaccination versus vaccination policies for the control of FMD.

Le virus de la fièvre aphteuse est un aphtovirus de la famille des Picornaviridae et l'agent de la maladie animale la plus importante sur le plan économique. En tant que picornavirus typique, le virus de la fièvre aphteuse est nu, sous forme d'icosaèdre et son génome comprend un acide ribonucléique monobrin avec environ 8500 nucléotides et une polarité positive. L'acide ribonucléique de ce virus est infectieux et il se réplique par l'intermédiaire d'un brin d'ARN moins, complémentaire. La réplication de l'acide nucléique de ce virus conduit à des erreurs, de telle sorte que les populations virales comprennent un ensemble de mutants (quasi espèce) plutôt qu'une séquence génomique bien définie. Par suite, le virus de la fièvre aphteuse est génétiquement et antigéniquement varié. Ceci entraîne des difficultés importantes pour le diagnostic, la prévention et la maîtrise de la fièvre aphteuse. Une connaissance plus approfondie de la complexité et de l'évolution de la population de ce virus a conduit à des besoins pour une nouvelle génération de vaccines aphteux. Black-tailed prairie dogs (Cynomys ludovicianus) currently live in metapopulations in the parts of their range where plague, caused by the bacterium Yesinia pestis, has invaded. Prairie dogs are highly susceptible to the pathogen, with most animals within towns dying during Y. pestis outbreaks. A review of population genetic studies of prairie dogs demonstrates considerable differentiation between prairie
dog towns. Despite declines and fluctuations in size of prairie dog populations, they continue to harbor considerable genetic
variation. This results from continual dispersal and gene flow, likely along low-lying drainages that connect towns. When
combined with estimates of population size, the landscape genetic approach described here will provide precise estimates of
dispersal and gene flow, in addition to evaluation of long-term stability of prairie dog metapopulations. 相似文献
Mature green mango fruits of commercially important varieties were screened to investigate the levels of constitutive antifungal compounds in peel and to assess anthracnose disease after inoculation with Colletotrichum gloeosporioides. High-pressure liquid chromatography was used to quantify the levels of 5-n-heptadecenylresorcinol and 5-n-pentadecylresorcinol in the peel extracts. The fruit peel of the varieties ‘Kensington Pride’ and ‘Keitt’ were observed to have the highest levels of both 5-n-heptadecenylresorcinol (107.3–123.7 and 49.9–61.4 μg/g FW, respectively) and 5-n-pentadecylresorcinol (6.32–7.99 and 3.30–6.05 μg/g FW, respectively), and the fruits of the two varieties were found to have some resistance to postharvest anthracnose. The varieties ‘Kent’, ‘R2E2’, ‘Nam Doc Mai’, ‘Calypso’ and ‘Honey Gold’ contained much lower concentrations of resorcinols in their peel and three of these varieties were found to be more susceptible to anthracnose. Concentrations of 5-n-heptadecenylresorcinol were significantly lower at the ‘sprung’ and ‘eating ripe’ stages of ripening compared to levels at harvest. Concentrations of 5-n-pentadecylresorcinol did not differ significantly across the three stages of ripening. The levels of these two resorcinols were found to be strongly inter-correlated (P<0.01, r2=0.71), with concentrations of 5-n-heptadecenylresorcinol being an average 18 times higher than those of 5-n-pentadecylresorcinol. At the ‘eating ripe’ stage, significant relationships were observed between the concentrations of each type of alk(en)ylresorcinol and anthracnose lesion areas following postharvest inoculation, P<0.001, r2= 0.69 for 5-n-pentadecylresorcinol, and P<0.001, r2= 0.44 for 5-n-heptadecenylresorcinol. 相似文献
