AIM: To investigate the effects of oxidized high-density lipoprotein (oxHDL) on membrane fluidity and the expression of lymphocyte function associated antigen(LFA-1) of monocyte. METHODS: The membrane fluidity of THP-1 cells was assayed by fluorescence anisotropy with DPH (1,6-dipheny-1,3,5-hexatriene), a fluorescent probe; The LFA-1 expression on THP-1 cells were assayed by flow cytometry with indirect immunofluorescence.RESULTS: The membrane fluidity of THP-1 cells was reduced by 45% and 52% respectively (P<0.01) after incubation with 100 μg protein/mL oxHDL and oxLDL for 20 h; oxHDL could stimulated the expression of LFA-1 on THP-1 cells, the positive cell rate and mean fluoresence intensity (MFI) increased by 39% and 45% respectively versus control group (P<0.01) after incubation with cells for 24 h. CONCLUSIONS: By increasing the expression of LFA-1 on monocyte, oxHDL can promote the monocyte-endothelium adhesion; oxHDL also can reduce the membrane fluidity of monocyte, that will limit the returning of monocytes from subendothelium back to the circulation. This suggested that oxHDL may play an important role in atherogenesis.  相似文献

AIM To investigate the effects of cytochrome P450 (CYP450) epoxygenase/epoxyeicosatrienoic acid (EET) pathway on insulin resistance in obese mice, and to explore the possible mechanisms. METHODS High-fat diet-induced obesity model was established in C57BL/6Cnc mice, and the obese mice were randomly divided into 3 groups, including obesity group (treated with saline; n=10), EET group (treated with 11,12-EET; n=10) and EET inhibitor 14,15-epoxyeicosa-5(Z)-enoic acid (EEZE) group (n=10). Normal C57BL/6Cnc mice (n=10) treated with saline served as control. Protein expression of CYP2J2 (one of CYP450 epoxygenases) and hypoxia-inducible factor-1α (HIF-1α) was measured by Western blot. Vessel-like structure was detected by immunofluorescence staining. The serum levels of insulin, tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and monocyte chemoattractant protein-1 (MCP-1) were measured by ELISA. RESULTS In obese mice, homeostasis model assessment of insulin resistance (HOMA-IR) values were increased, the protein level of CYP2J2 was reduced, and the protein level of HIF-1α was increased in adipose tissues as compared with the controls (P<0.05). The serum levels of MCP-1, IL-1β, IL-6 and TNF-α were also significantly increased in obese mice (P<0.05). After treatment with 11, 12-EET, the HOMA-IR values were decreased compared with vehicle-treated obese mice, HIF-1α expression levels were decreased in the adipose tissue, and the serum levels of MCP-1, IL-1β, IL-6 and TNF-α were reduced (P<0.05). Immunohistochemical results of adipose tissue from vehicle-treated obese mice showed a marked decrease in vessel-like structures (CD31-positive) compared with normal control mice (P<0.05). EET treatment significantly increased the newly formed vessel-like structures in the visceral adipose tissues of obese mice as compared with vehicle-treated obese mice (P<0.05). CONCLUSION High-fat diet-induced obesity and insulin resistance are closely related to the CYP450 pathway. Exogenous EETs effectively decrease obesity-induced insulin resistance possibly through pro-angiogenesis and attenuation of hypoxia and inflammation.  相似文献

MNX1 antisense RNA 1 (MNX1-AS1) is a newly discovered long non-coding RNA (lncRNA), which is highly dysregulated in various carcinomas and its expression level is closely related to the overall survival and prognosis of patients. MNX1-AS1 regulates the occurrence and development of carcinomas by endogenous competitive adsorption of miRNA, regulating cell cycle, inducing epithelial mesenchymal transformation and activating multiple signaling pathways. The in-depth study of the carcinogenesis of MNX1-AS1 is useful for the early diagnosis, targeted therapy and prognostic assessment of relevant carcinomas. 平椒3号是以自交系99101为母本,自交系9949为父本配制的早熟一代杂种。果实羊角形,青熟果深绿色,果纵径17.7cm,果肩径3.0cm,果肉厚0.25cm,平均单果质量29.5g,肉质脆嫩,辣味浓且有香味,商品性好。田间对疫病、炭疽病、病毒病的抗性明显优于猪大肠。露地栽培每667m2青椒产量为3700kg左右,适宜西北地区及四川、山西、湖南等嗜辣地区露地栽培。  相似文献

Compound 1 (SMTP-7, also FGFC1), an isoindolone alkaloid from marine fungi Starchbotrys longispora FG216 and fungi Stachybotrys microspora IFO 30018, possessed diverse bioactivities such as thrombolysis, anti-inflammatory and anti-oxidative properties, and so on. It may be widely used for the treatment of various diseases, including cerebral infarction, stroke, ischemia/reperfusion damage, acute kidney injury, etc. Especially in cerebral infarction, compound 1 could reduce hemorrhagic transformation along with thrombolytic therapy, as the traditional therapies are accompanied with bleeding risks. In the latest studies, compound 1 selectively inhibited the growth of NSCLC cells with EGFR mutation, thus demonstrating its excellent anti-cancer activity. Herein, we summarized pharmacological activities, preparation of staplabin congeners—especially compound 1—and the mechanism of compound 1, with potential therapeutic applications.  相似文献

芝麻是我国重要的特色优质油料作物。提高芝麻单产水平是实现我国粮油安全生产的重要举措。为建立芝麻高产高效栽培模式,本研究以耐密植黑芝麻新品种豫黑芝1号为试验材料,在3个产区(漯河、信阳和三门峡),开展了不同栽培密度(12万株/hm2、15万株/hm2、18万株/hm2、21万株/hm2)下豫黑芝1号产量及产量相关性状变化分析。结果显示,豫黑芝1号在不同产区的生育期差异明显;种植密度对豫黑芝1号的株高、有效果节数、主茎果轴长、单株蒴果数、千粒重和单株产量等性状变化程度不同;对小区产量及单株产量影响显著。在21万株/hm2密度下,豫黑芝1号单产水平最高达到1463.33 kg/hm2(三门峡)。研究为芝麻高产高效生产提供了技术支持。  相似文献

 通过TAIL-PCR 染色体步移技术,从甘蓝(Brassica oleracea L. var. capitata L.)基因组中克 隆到胞质雄性不育(OguCMS)相关基因BoMF1 翻译起始位点上游521 bp 的启动子序列。软件分析预测 表明,该启动子序列中存在多个顺式作用元件,包括TATA-box、CAAT-box、MYB 结合位点、植物激 素响应单元等。为了研究该启动子的表达特性,亚克隆了BoMF1 转录起始位点上游521 bp 序列,将其置 换pBI121 中的CaMV35S 启动子,驱动其下游的GUS 基因,构建植物表达载体pBI121-BoMF1P,以pBI121 空载体作为阳性对照,通过农杆菌(LBA4404)介导法转入拟南芥。结果表明,甘蓝BoMF1 启动子序列 能驱动GUS 基因在拟南芥花药发育晚期的花药和花粉中特异表达,表达具有组织特异性。  相似文献

AIM:To observe changes of serum asymmetry dimethylarginine (ADMA) and endothelin-1 (ET-1) levels,which reflect blood vessel endothelial function,after therapy of enhanced external counterpulsation(EECP) in patients who had taken percutaneous transluminal coronary angioplasty and stent.METHODS:Fifty one coronary heart disease patients (all of them had taken percutaneous tranluminal coronary angioplasty and stent) were distributed into two groups by matching them with ratio of 1∶2,17 patients in EECP group and 34 patients in control group.Both of two groups were given conventional medicine,in addition,EECP group was undertaken three courses of treatment of EECP.ADMA was detected by HPLC-fluorescence method,and ET-1 was detected by radio-immunity method.RESULTS:In EECP group ,compared with prior treatment,ADMA and ET-1 levels was obviously reduced (P<0.05).In control group,compared with prior treatment,ADMA had no significant difference (P>0.05) and ET-1 was higher (P<0.01).Compared between two groups,extent of decrease in ADMA and ET-1 levels in EECP group were more obvious than control group (P<0.01).Improvement of angina and decrease of frequency of angina was more obvious in EECP group than control group.Besides,decrease of ADMA level was positive correlation with them (r=0.85,0.70,respectively P<0.01).CONCLUSION:EECP reduces serum ADMA and ET-1 levels by increasing shear stress to vessel endothelium in coronary heart disease patients who had taken percutaneous tranluminal coronary angioplasty.It hints that EECP can improve endothelial function and provides experimental evidence for the combined treatment of coronary heart disease.  相似文献
