Steroid receptor expression was assessed by immunohistochemistry in neoplastic, hyperplastic/dysplastic, and normal mammary tissue samples removed from 68 queens and 47 bitches, using monoclonal antibodies against human oestrogen-alpha (ER) and progesterone receptors (PR). Mammary lesions were classified according to World Health Organization (WHO) criteria, and all animals with invasive carcinomas were clinically followed for 2 years. Stromal and/or lymphatic invasion and histological grading were also recorded. In both species, ER expression was significantly higher in healthy tissues, hyperplastic/dysplastic lesions, and benign tumours than in carcinomas. The loss of ER expression was more marked in feline than in canine carcinomas. In queens, PR expression increased in dysplastic lesions and "in situ" carcinomas and decreased in invasive carcinomas, even if parts of these tumours were still PR-positive. In bitches no significant variation in PR expression was observed between normal tissue, dysplasias, and benign neoplasms, but was significantly lower in carcinomas. In both species ER and PR expression in invasive carcinomas did not correlate either with histological parameters or overall survival time. This study demonstrates several differences in steroid hormone dependency between the two species. The percentage of PR-positive feline carcinomas suggests a possible role of progesterone in promoting early tumour cell growth in queens. The low percentage of ER-positive invasive carcinomas further demonstrated the aggressive phenotype and behaviour of feline mammary tumours.  相似文献

The presence of metallothioneins (MTs) were demonstrated immunohistochemically using a monoclonal antibody (E9) against a conserved epitope of I and II isoforms in canine mammary tumours. In a semiquantitative analysis MT expression in the tumour cells was observed in 54/54 cases of benign and 32/40 malignant mammary neoplasms. A statistically significant difference at the level of P<0.01 was observed for MT expression between benign and malign mammary tumours in terms of immunoreactivity score. It is concluded that immunohistochemically demonstrated MT expression is significantly associated with benign canine mammary tumours.  相似文献

A yearling Irish Cob stallion was presented to De Morette Equine clinic for severe acute abdominal pain, nonresponsive to analgesics and sedatives. Rectal palpation and a limited transabdominal ultrasonographic examination led to the presumptive diagnosis of a strangulated obstruction of the small intestine. The painful behaviour of the pony did not allow conservative treatment and surgical exploration was denied by the owners for financial considerations. On necropsy, a large cystic mature teratoma was adhered to the ventral body wall, the mesenteric root, omentum and intestines. Teratomas are rare in man and animals, and an extragonadal intraperitoneal location has not previously been reported in the equine species.  相似文献

An investigation was undertaken to study two different types of farrowing pens, use being made of well characterized veterinary parameters for the well‐being of piglets. A housing system with a solid floor and straw‐bedding was compared with a partly metal‐slatted floor system. The veterinary parameters we used were mortality, morbidity, and external injuries in piglets. There were slight differences in mortality and morbidity rates between the two housing systems. There were big differences with regard tp the piglets” injury‐index’ of the two housing systems. The straw system was more favourable. The studies show that obervations of external injuries ('Method of Ekesbo') can be used to judge housing systems of piglets in the farrowing house.  相似文献

Summary

Two attenuated transmissible gastro‐enteritis (T. G. E.) virus strains were used for vaccination experiments in sows.

Four different experiments were carried out (see Table 1). In each experiment, 9 sows were vaccinated during pregnancy and 3 sows served as controls. They were kept together in one farrowing house. The sows were due to farrow at about the same time. The sows and their litters were challenged shortly after farrowing by exposing 3 piglets of 2 control litters to virulent TGE virus.

The following vaccination schedules were used (see Table 1): twice intramuscularly with TGE‐vac (a commercially available TGE‐vaccine), one oral administration followed by an intramuscular vaccination with an attenuated TGE Purdue (Pu) strain, twice orally with Pu strain in enteric coated capsules, and one direct intra intestinal administration followed by 2 intramuscular vaccinations or 3 intramuscular vaccinations with the Pu strain.

All sows, except most of those treated with enteric coated capsules, seroconverted demonstrably (Table 2). The geometric mean seroneutralization (SN) titer log 2 varied from 4.1 to 7.5 after the first vaccination and from 7.6 to 10 after the second vaccination.

None of the vaccination schedules resulted in an effective lactogenic immunity. The morbidity in the piglets was 100% within 3 to 5 days after challenge. The mortality rate varied from 44 to 80% in litters from vaccinated sows and from 71 to 100% in litters from control sows (see Table 3). Clinical signs were observed in 33,3% of the control sows and in 36% of the vaccinated sows.

No correlation was found between the titer of SN antibodies in the sera of the piglets and their survival rate (Table 4).

A rapid decrease in antibody concentration was observed, during the first week of lactation in milk samples collected from 4 orally and intramuscularly vaccinated sows (Table 5).  相似文献

During the onset of lactation, there is a dramatic increase in the expression of glucose transporters (GLUT) and a group of enzymes involved in milk fat synthesis in the bovine mammary gland. The objective of this study was to investigate whether the lactogenic hormones mediate both of these increases. Bovine mammary explants were cultured for 48, 72, or 96 h with the following hormone treatments: no hormone (control), IGF-I, insulin (Ins), Ins + hydrocortisone + ovine prolactin (InsHPrl), or Ins + hydrocortisone + prolactin + 17β-estradiol (InsHPrlE). The relative expression of β-casein, α-lactalbumin, sterol regulatory element binding factor 1 (SREBF1), fatty acid synthase (FASN), acetyl-CoA carboxylase α (ACACA), stearyol-CoA desaturase (SCD), GLUT1, GLUT8, and GLUT12 were measured by real-time PCR. Exposure to the lactogenic hormone combinations InsHPrl and InsHPrlE for 96 h stimulated expression of β-casein and α-lactalbumin mRNA by several hundred-fold and also increased the expression of SREBF1, FASN, ACACA, and SCD genes in mammary explants (P < 0.01). However, those hormone combinations had no effect on GLUT1 or GLUT8 expression and inhibited GLUT12 expression by 50% after 72 h of treatment (P < 0.05). In separate experiments, the expression of GLUTs in the mouse mammary epithelial cell line HC11 or in bovine primary mammary epithelial cells was not increased by lactogenic hormone treatments. Moreover, treatment of dairy cows with bovine prolactin had no effect on GLUT expression in the mammary gland. In conclusion, lactogenic hormones clearly stimulate expression of milk protein and lipogenic genes, but they do not appear to mediate the marked up-regulation of GLUT expression in the mammary gland during the onset of lactation.  相似文献

Background

The study aimed at examining hypoxia-inducible factor (HIF)1α expression in adenocarcinomas and adenomas in bitches in regard to tumour malignancy grade, proliferation, apoptosis and vascularisation. Therefore, paraffin sections of 15 adenomas and 64 adenocarcinomas sampled from 79 dogs aged 6 to 16 years were analysed.

Results

A significantly higher HIF-1α expression was noted in adenocarcinomas in comparison to adenomas (P < 0.0004). Moreover, HIF-1α expression in adenocarcinomas correlated positively with tumour malignancy grade (r = 0.59, P < 0.05), Ki-67 antigen expression (r = 0.43; P < 0.0005), TUNEL-positive cells (r = 0.62, P < 0001) and tumour vascularity measured by quantification of vessels characterized by the expression of von Willebrand Factor (r = 0.57, P < 0.05).

Conclusion

Results of this study indicate a similar biological role of HIF-1α in dogs and in humans, which may confirm suitability of the animal model in investigations on progression of tumours in humans.  相似文献
