Colostrogenesis during an induced lactation in dairy cattle

Colostrum immunoglobulin G (IgG) is of major importance for the newborn calf because epitheliochorial placentae do not provide transport in utero. The formation of colostrum occurs in the later stages of pregnancy. Our objectives were to induce lactation in non‐pregnant dairy cows and (i) to determine the changes of IgG in serum and mammary secretions during the induction process and (ii) to establish α‐lactalbumin (αLA) and prolactin (Prl) alterations to monitor the changing mammary epithelial tight junction status and development pattern. Estradiol‐17β (E2) and progesterone (P4) injections in a 1–7 days series were combined with a 3‐day injection series (day 21–23) of dexamethasone (DEX). Blood and both front quarter secretion samples were collected daily. Milking started 24 days after the start of the experiment. Results show that the mammary secretory IgG1 was increased at >7 days after the start of steroid injections and depicted a bimodal pattern reaching a high of 16 mg/ml at 21 day compared with 3.2 mg/ml in the serum. There was a small increase in secretory IgG2 that did not correlate with tight junction status, but never reached the serum concentration. The injections of DEX resulted in constriction of tight junctions. Secretory αLA was immediately increased with steroid injections, dropped precipitously after 7 days and then began a steady increase until the start of milking. Changes in serum αLA are related to mammary tight junctions while serum Prl gradually increased from 30 to >60 ng/ml after the steroid injections stopped. These results provide insights into the mechanisms and timing of colostrogenesis during an induced lactation protocol.     

猪血浆IgG提取物对小白鼠免疫功能的影响

研究灌胃猪血浆IgG提取物对小白鼠免疫功能的影响。采用腹腔注射环磷酰胺制造试验小白鼠的免疫功能抑制模型,选用齐墩果酸为阳性对照组,猪血浆IgG提取物为试验组,预饲5d后,然后将猪血浆IgG提取物按每千克体重45、90和180mg3种剂量给小白鼠连续灌胃10d后测定小白鼠免疫水平的变化。小白鼠灌胃猪血浆IgG提取物后,对抗由环磷酰胺引起的小白鼠免疫器官萎缩有一定的保护作用,并对环磷酰胺所致动物DTH反应低下有一定的对抗作用;能显著增加小白鼠血清中IgG的含量和免疫球蛋白的含量(P<0.05),能显著增加小白鼠血清中球蛋白的含量(P<0.05),灌胃猪血浆IgG提取物能显著提高小白鼠的免疫功能。     

Interaction of bovine immunoglobulin gamma 2 (IgG2) with staphylococcal protein A: evidence for IgG2a and IgG2b sub-subclasses in the bovine

The reactivity of bovine IgG with protein A is confusing with respect to which of the bovine IgG class and subclasses are reactive. We have, therefore, re-examined the interaction of bovine immunoglobulins with protein A. The results presented in this paper indicated that at pH 8.0 protein A binds only immunoglobulin of the IgG2 subclass. The bound IgG2 can be readily recovered from an immobilized protein A column at pH 5.0. Furthermore, the antigenic IgG2 eluted demonstrated two charged species which could readily be separated by ion-exchange chromatography. These results indicate that IgG2 in the bovine exists in two sub-subclasses, IgG2a and IgG2b. The two sub-subclasses of IgG2 could be rapidly isolated with a good yield in two-steps namely protein A affinity chromatography followed by ion exchange chromatography.     

通城猪及其杂种猪若干免疫性状的研究

通过对通城猪、长大通、大长通杂种猪红细胞数(RBC)、白细胞数(WBC)、血红蛋白浓度(HGB)、红细胞压积(HCT)、平均血细胞容积(MCV)、平均血细胞血红蛋白量(MCH)、平均血细胞血红蛋白浓度(MCHC)、IgG含量、ANAE阳性率(ANAE^ )、迟发型皮肤超敏反应(DTH)的测定,发现通城猪DTH、WBC、RBC、HGB、HCT、MCV、MCHC性状的平均值均高于长大通和大长通杂种猪。对影响各性状的因素进行了分析,结果表明DTH、WBC、RBC、HGB、HCT、MCV、MCHC性状与品种有关,WBC、MCHC与性别有关。对各免疫性状间的相关关系进行了分析,探讨了所测免疫性状指标对机体免疫力的代表性。     

感染鸡传染性贫血病毒的雏鸡新城疫疫苗免疫后局部体液免疫球蛋白含量变化

本试验对感染鸡传染性贫血病毒（ＣＩＡＶ）雏鸡新城疫（ＮＤ）疫苗免疫及其强毒攻击后，其泪液、气管液、肠液和胆汁的免疫球蛋白ＩｇＧ、ＩｇＭ、ＩｇＡ含量的动态变化进行了检测。结果发现，感染雏鸡泪液的ＩｇＧ、ＩｇＡ和气管液的ＩｇＧ及胆汁的ＩｇＧ、ＩｇＭ、ＩｇＡ含量均于接种ＮＤ疫苗后７～２８天明显低于未感染ＣＩＡＶ的免疫对照雏鸡；泪液的ＩｇＭ和气管液的ＩｇＡ含量在ＮＤ免疫后１４～２８天明显降低；肠液的ＩｇＡ和ＩｇＭ、ＩｇＧ含量分别于免疫后７～１４和７、１４天也显著减少。表明ＣＩＡＶ感染雏鸡对ＮＤ疫苗免疫的局部体液免疫应答能力降低。ＮＤ强毒攻击后，ＣＩＡＶ感染ＮＤ免疫雏鸡泪液、气管液、肠液和胆汁的ＩｇＧ、ＩｇＡ、ＩｇＭ含量及免疫保护率，均明显低于未感染ＣＩＡＶ的免疫对照雏鸡。     

免疫球蛋白G(IgG)三种提取方法比较

比较3种提取猪血清中免疫球蛋白IgG方法的提取效果。分别用辛酸沉淀法,硫酸铵分步沉淀法,辛酸-硫酸铵沉淀法粗提猪血清免疫球蛋白IgG,并通过SDS-PAGE凝胶电泳、AlphaEaseFC凝胶成像分析软件、Bradford蛋白浓度测定法等比较3种方法的提取纯度和得率,同时比较3种方法的时效性和经济性。辛酸沉淀法提取IgG所用时间最短,只需1h,得率较高但纯度不高;硫酸铵盐析法提取的IgG纯度较高、得率也高,但时间较长;辛酸-硫酸铵分步沉淀法所得IgG纯度很高,但所用时间较长,得率也较低。3种提取方法各有特点,可根据不同的实验条件和目的选择不同的提取方法。     

牛初乳IgG的特性及检测方法

主要介绍了牛初乳中IgG的理化特性和生理功能及其在胃肠内的抗降解特性,同时介绍了牛初乳IgG的提取及常用检测方法,并对牛初乳IgG作为保健产品和医疗药品基料的趋势作出展望。     

Isolation and Purification of Bovine Colostrum sIgA and IgG

To further utilize bioactive substance such as bovine colostrum sIgA and IgG,sIgA and IgG were isolated and purified simultaneously by salting out,ultrafiltration and gel chromatography,etc.The analysis of results were showed quantitatively by non-hydrogenized SDS-PAGE,and quanlities of sIgA and IgG were respectively detected by Western Blot.The results showed that the purity and yield of bovine colostrum sIgA were 85.3%and 42.8%,respectively,while the purity and yield of bovine colostrum IgG were respectively 97.2%and 64.4%.This preparative method provides theoretical and experimental foundation for sIgA industrial production.     

大肠杆菌MM—3基因工程活菌苗免疫母猪血清及乳清中抗K_(88ac)及LTB抗体动态

以MM-3基因工程活菌苗口服免疫妊娠73d、83d、103d母猪各4头,口服及肌注免疫妊娠92d母猪各5头,用间接BA-ELISA法检测血清及乳清中IgG、IgA和IgM型抗K_(88ac)及LTB抗体的动态变化.结果表明,无论口服还是肌注免疫母猪,抗K_(88ac)及抗LTB抗体均极显著增高.肌注以IgG增加为主,口服以IgA增加为主.肌注免疫IgG型抗K_(88ac)抗体滴度极显著地高于口服免疫(P<0.01);口服免疫IgA型抗LTB抗体滴度显著高于肌注免疫(P<0.05).口服及肌注免疫乳清中抗K_(88ac)及抗LTB抗体滴度很高,达1:3200～204800,但在1周内即降低,约减少一半(约3～5个滴度),2～3周内各型抗体下降缓慢,维持在对照组周内水平,4周后达最低水平.妊娠不同时期口服免疫,其血清中抗K_(88ac)抗体的动态变化相似,但乳汁中抗体则差异非常明显,妊娠83d和92d免疫组IgG、IgA和IgM均显著增加,且高于103d组(P<0.01)和73d组(P<0.05).口服免疫的最佳时间为分娩前26～30d,肌注免疫最佳免疫时间为分娩前21d.
