In the Philippines, bacterial wilt caused by Ralstonia solanacearum is one of the most important diseases affecting vegetables and banana. In this study, 89 strains of R. solanacearum isolated from various hosts were screened for their biovar, phylotype, pathogenicity, and genetic diversity. Foreign strains were included for comparison with these Philippine strains. Results of the biochemical and multiplex-PCR tests divided the Philippine strains into five biovars (1, 2, 3, 4, and N2) and three phylotypes (I, II, and IV). Three potato strains belonged to biovar N2/phylotype IV. Pathogenicity tests divided the strains into five pathogenicity types based on their virulence in tomato, potato, eggplant, sweet pepper, and tobacco. Strains classified as biovar N2 were weakly pathogenic to potato (pathogenicity type III) and almost all strains isolated from banana were not pathogenic to the test plants except potato (pathogenicity type V). The results of AFLP analysis divided the strains into four clusters. Cluster 1 was composed of strains isolated from solanaceous crops, ginger (Zingiber officinale), and Morus sp. from the Philippines and other Asian countries. Cluster 2 grouped the potato strains (biovar N2) from the Philippines and Japan and blood disease bacterium strains from Indonesia. Cluster 3 contained the local and foreign strains isolated from potato (biovar 2) and banana (biovar 1). Cluster 4 consisted only of the tomato strain from the USA.  相似文献

The method of segregating nulliparous and parous females of Culicoides spp. based on the presence of burgundy-red pigment inside the abdominal wall of parous Culicoides midges, is used worldwide. Out of 320 females of Culicoides imicola trapped by emergence traps, set over an artificial breeding site for 10 and 24 days, 73 (22.8%) showed a red-pigmentation despite the fact that they were nulliparous. This finding indicated that 23% of the “parous” females that are examined for the presence of arboviruses and other pathogens or for age-grading purposes, are actually old nulliparous females, which had no chance of acquiring pathogens. This bias in parous rate distorts upward the calculation of vectorial capacity.  相似文献

The association between risk of seroconversion of sentinel cattle to bluetongue viruses and the number of Culicoides brevitarsis Kieffer and C. wadai Kitaoka caught by light traps was investigated using survival analysis. Eight sentinel herds that seroconverted to bluetongue viruses between 1990 and 1994, and for which insect-trapping data were available, were selected for inclusion in the study. These herds were located at six sites along the eastern coast of Queensland, Australia, from approximately latitude 10 °South to 25 °South. C. brevitarsis was detected at all locations where sentinel herds were maintained, whereas C. wadai was detected at only two locations in northern Queensland where four sentinel herds were maintained during the study period. The mean number of C. brevitarsis and C. wadai caught per month was 230 and 21, respectively. A significant (P = 0.05) positive association was found between the risk of seroconversion of sentinel cattle to bluetongue viruses and the number of C. wadai caught in the same month.  相似文献

The anti-inflammatory effect of a triamcinolone topical solution (TTS) was evaluated by use of induced cutaneous immediate-type hypersensitivity reactions in dogs. Healthy dogs ( n  = 3) were treated on one side of the lateral thorax with TTS, and on the contralateral side with vehicle solution. After 7 days of treatment, serial dilutions of inflammatory stimuli (histamine, compound 48/80, anticanine IgE, substance P, and morphine sulphate) were injected intradermally on both sides. Resulting immediate reactions were subjectively scored, their diameters were measured in millimetres, and microcirculatory blood flow at each site was measured using laser Doppler velocimetry. The magnitude of the differences between TTS and vehicle-treated sides varied with the type of measurement and the stimulus used. With compound 48/80, anti-IgE, substance P, and morphine sulphate-injected sites, for at least one concentration, the reaction scores and sizes were notably less on the TTS-treated sides. Punch skin biopsy specimens were obtained from selected sites 24 h after injection. A pathologist examined stained tissue sections and scored each section as to degree of late-phase dermal cellular infiltrate. Dermal infiltrate scores were generally lower at the TTS-treated sites; the composite score from all TTS-treated sites was 30% lower than that from the vehicle-treated sites. Observations suggest that TTS may be effective in decreasing some aspects of the inflammatory response in canine skin.  相似文献

Chili anthracnose, caused by Colletotrichum spp., is one of the major diseases to chili production in the tropics and subtropics worldwide. Breeding for durable anthracnose resistance requires a good understanding of the resistance mechanisms to different pathotypes and inoculation methods. This study aimed to investigate the inheritances of differential resistances as responding to two different Colletotrichum pathotypes, PCa2 and PCa3 and as by two different inoculation methods, microinjection (MI) and high pressure spray (HP). Detached ripe fruit of Capsicum baccatum ‘PBC80’ derived F2 and BC1s populations was assessed for anthracnose resistance. Two dominant genes were identified responsible for the differential resistance to anthracnose. One was responsible for the resistance to PCa2 and PCa3 by MI and the other was responsible for the resistance to PCa3 by HP. The two genes were linked with 16.7 cM distance.  相似文献

This laboratory study investigates the sub-adult developmental cycle of field collected Culicoides (Avaritia) imicola Kieffer (Diptera; Ceratopogonidae). The period required from blood-feeding field-collected females to the production of progeny adults occupied 34–56 days at 20 °C, 15–21 days at 25 °C and 11–16 days at 28 °C, demonstrating clear temperature dependence. When reared at 28 °C, C. imicola demonstrated higher variability in fecundity (between 2.4 and 20.6 eggs/female) and lower hatching rates (50.0–62.2%), although larval survival rates to pupation were low at all temperatures (20–30%). Similarly, the mean emergence rate from pupae was the highest at lower temperatures. These results highlight the difficulty in establishing and maintaining a laboratory colony of this species from field-collected material and results are discussed in reference to future research directions that may aid this process.  相似文献

This experiment was conducted to understand mosquito species, distribution of the main vector of Japanese encephalitis virus (JEV), JEV genotype and molecular characteristics in upper Pearl River region. We collected mosquito and Culicoides specimen in Shizong county, upper Pearl River region in July 2013, and these specimens were classified and identified. Meanwhile, JEV nucleotide were detected using PrM and E genes specific primers in 162 pools of mosquito and Culicoides specimen, and the positive specimens were sequencd and analyzed by bioinformatics software. Total 3 705 mosquitoes were collected from 4 collection spots, and the mosquitoes belong to 7 species in 4 generas: Culex, Anopheles, Aedes and Armigeres. In cattle pens, Anopheles Sinensis and Armigeres subalbatus were the dominant mosquito species. In sheep pens, Anopheles Sinensis and Armigeres subalbatus were the dominant mosquito species. And in pig pens, Culex tritaeniorhynchus, Anopheles Sinensis and Armigeres subalbatus were the dominant mosquito species. 4 of 162 pools mosquito and Culicoides were positive detecting by using PrM and E genes specific primers. Sequence analysis of JEV PrM and E genes results showed that 4 strains virus from Culex tritaeniorhynchus samples belonged to genotype Ⅰ JEV, and existing 15 amino acid difference sites between vaccine strain SA14-14-2 in the E gene protein, and 8 important sites which affected virus virulence did not mutant. These results suggested that there were many mosquito species in upper Pearl River region, and Culex tritaeniorhynchus, Anopheles Sinensis were dominant mosquito species in this region, and Culex tritaeniorhynchus was the main media of JEV in local region, genotypeⅠ JEV was popular in local media, and virulence of the pandemic strain did not reduce.  相似文献
