家蚕RAPD的多态性与稳定性分析

从家蚕品种大造和C108及其F1,F2的后部丝腺提取的基因组DNA通过的RAPD扩增,在477个随机引物中,85.9%的引物在家蚕基因组有稳定的扩增产物,总扩增片段5155条,每个引物扩增片段1-23条,平均为12.6条,多态性片段数共1496条,占两品种中总片段数的29.0%,经多次重复及反复验证发现,只要严格控制扩增反应条件及保证基因组DNA和试剂等不污染,则能检测到稳定的DNA多态性片段,从而保证其稳定性与重复性。     

高校设计专业“三大构成”课程教学模式改革与探索

三大构成课教学体系自包豪斯引入我国,对中国艺术设计教育产生了巨大影响。时代的发展和观念的更新,使得构成教学面临新的挑战。本文主要对高校设计专业三大构成课教学模式及存在的问题展开了调研分析,分别从课程设置、教学框架、教材选用、教学方式、学习方法、教学评价等方面提出了设计构成基础课教学新思路,以适应新时代发展的需要,使构成基础课与专业实践有效结合,充分发挥其在专业设计中的真正价值。     

三大因素引起我省粮食生产周期性波动

湖南粮食生产具有＂波动中增长＂特征。1978～2010年期间湖南粮食生产先后出现了7个波动周期,人均粮食产量先后出现8个波动周期,每个波动周期具有不可重复性和非对称性。     

标准体积管重复性的计算

介绍了体积管标准体积的计算方法,分析和比较了标准JJG 209～1980、JJG 209-1994和API MPMS 12.2.4-1997等关于体积管重复性的计算方法及相关规定,使用不同的计算方法计算了体积管的重复性.     

猪链球菌2型实时荧光定量PCR检测方法的建立与应用

试验根据猪链球菌2型荚膜多糖（CPS）抗原设计CPS2J基因特异性引物,建立猪链球菌2型实时荧光定量PCR检测方法。结果显示,该方法的标准曲线为y=-3.073x+36.87,r=0.995,熔解曲线只有单一的特异峰。敏感性试验显示,该方法可以检测出模板最低浓度为1.0×10¹拷贝/μL,是普通PCR的10倍;特异性试验显示,对猪链球菌2型具有良好的特异性,能够区分其他血清型猪链球菌和其他细菌;重复性试验变异系数为0.37%~0.63%,均低于2.5%。临床检测显示该方法的敏感性明显高于常规PCR方法和细菌分离的方法。以上结果表明,本研究建立的方法敏感性高、特异性强、重复性好,有利于对猪链球菌2型的快速检测。     

烟尘测试仪动压示值误差不确定度的评定

利用压力校准装置对烟尘测试仪动压进行了校准,评定了压力测量结果示值误差的不确定度。在测量900Pa动压时,示值误差扩展不确定度U=57Pa,提供了约为95%的置信概率。     

对当前兽药业发展的若干思考

兽药GMP重复性建设 亟待科学引导 目前,兽药产品生产线低水平重复建设问题显露。企业生产线“大而全”或“小而全”,每个省份甚至每个地市的GMP企业生产线类型、产品架构趋于雷同;而在兽药国家标准实施后,产品组方同质化严重,导致兽药生产企业和兽药产品鱼龙混杂,加剧了市场不良竞争。     

Establishment of Digital RT-PCR Assay for Detection of Swine Influenza Virus H3N2 Subtype

A new assay for the detection of swine influenza virus (SIV) was developed with a novel nucleic acid probe——Molecular beacon in this study. The specific primers and molecular beacon probes were designed according to the conserved region of H3 and N2 genes of SIV H3N2 subtype. A digital RT-PCR assay was developed for detection of SIV H3N2 subtype. The results showed that SIV H3N2 subtype could be identified simultaneously on this microarry with high sensitivity and reproducibility,which could reach to 10⁶ dilute viruse. The conclusion was that the digital RT-PCR method could analyze quantitatively the RNA templates.On the identification of H3N2 SIV,the digital RT-PCR method was much more scientific than Real-time quantitative PCR method.     

重复性过渡过程的分析

借助于两个具体电路，分析了在有规律的重复脉冲激励下，强制响应的形式。强制响应就是指电路进入稳态后网络对激励的反应，其求解依据换路定则—惯性量的初始值等于前一时间段内该物理量的终了值。     

Establishment and Application of Quantitative Real-time PCR Method to Detect Streptococcus suis Serotype 2

In this study,a quantitative Real-time PCR method using the specific primers according to CPS2J gene was established to detect Streptococcus suis serotype 2. The result showed that the equation of standard curve was y=-3.073x+36.87,r=0.995,which demonstrated that the assay had good linear relationship.The melting curve analysis showed that there was only specific peak.Sensitivity test showed that the method could detect the template at the lowest concentration of 1.0×10¹ copies/μL,which was 10 times higher than the ordinary PCR.The specific tests showed that this method could able to detect Streptococcus suis serotype 2 specially and had no cross-reaction with other serotypes or other bacteria from swine. The CV of repeatability test was 0.37% to 0.63%,lower than 2.5%. The clinical diagnosis showed this assay was more sensitive than ordinary PCR and bacteria isolation. All the results showed that the established method was sensitive,specific and reproducible,which could be used for the rapid diagnosis and quantitative detection of Streptococcus suis serotype 2.
