Methodological aspects of flow-cytometric evaluation of the phagocytic properties of equine neutrophils were elucidated. The kinetics of attachment and ingestion were studied, and the phagocytic process was more rapidly completed when serum-opsonized yeast cells were used than with use of IgG-opsonized yeast cells. Trypan blue was successfully used to quench fluorescence of non-ingested yeast cells. There were only minor differences in the kinetics of phagocytosis between quenched and un-quenched samples, indicating that attachment is rapidly followed by ingestion. Trypan blue quenching caused loss of cells with light scattering properties of granulocytes, although this did not affect the determined frequencies of truly phagocytic neutrophils. Aggregation of yeast cells proved to be a disturbance but not an obstacle to the determination of frequencies of actively phagocytic cells. Flow cytometry is well suited for studies of phagocytosis of yeast cells by equine neutrophils, and the trypan blue quenching provides a means of eliminating false-positive events due to aggregation of yeast cells. 选用1344尾平均体重为105 g的健康湘云金鲫鱼种。随机分为4个处理,每个处理2个重复。Ⅰ组投喂基础饲料,Ⅱ、Ⅲ、Ⅳ组在基础饲料中分别添加4 m g/kg的黄酶素及10、20 g/kg葡萄籽等植物提取物。30 d后测定湘云金鲫的生长性能和鱼体成分。结果显示:Ⅲ组与Ⅰ组、Ⅱ组、Ⅳ组比较,增重率分别提高53.77%、45.43%、20.16%(P<0.05),饵料系数分别降低33.06%、31.02%、16.44%(P<0.05);鱼体蛋白质含量显著提高(P>0.05),体脂肪含量无显著变化(P>0.05)。因此,葡萄籽等植物提取物作为饲料添加剂能显著提高湘云金鲫的生长性能、降低饵料系数、改善鱼体品质,本试验条件下,其适宜添加量为10 g/kg。 相似文献

为了开发新的食用蛋白质资源,本试验以“青大1号”紫花苜蓿新品系青干草为原料,采用酸化加热法配合加盐法提取叶蛋白.单因素试验设计,分别考察料水比、加盐量、絮凝时间、pH、预浸时间、絮凝温度6个因素对粗蛋白质提取率的影响,并以单因素试验结果为依据,采用四因素(料水比、加盐量、絮凝时间、pH)三水平L9(34)正交试验设计进行提取工艺优化.试验结果表明:影响粗蛋白质提取率的因素,从大到小依次为料液比、絮凝时间、pH、加盐量.当絮凝温度为70℃、料液比为1∶10、加盐量1%、絮凝时间5min、pH为4时是叶蛋白提取的最优提取条件. 相似文献

The study was conducted to determine the effects of different levels of selenium yeast(SeY, added as selenium content) on growth performance and some biochemical indexes in Jinding laying ducks under high temperature (29-35℃). 144 Jinding laying ducks of rearing period (10-16 weeks old) were divided into 4 groups, control group, experimental groups Ⅰ,Ⅱ and Ⅲ, supplemented with 0, 0.15, 0.45 and 0.75 mg/kg Se, respectively. The results showed that compared with the control group, the addition of SeY to the diets could significantly increase the average daily feed intakes (ADFI) of ducks at 14-16 weeks old (P<0.05); The body weights of ducks in group Ⅰ at 13-15 weeks old were significant higher than that in the control group (P<0.05),and the body weight at 16 weeks old was higher than the other three groups significantly (P<0.05),there was no significant difference between the body weight and standard body weight at 16 weeks old in group Ⅰ(P>0.05), but the body weights of other three groups were extremely significantly lower than the standard weight (P<0.01); There was no significant difference among the four groups for feed to gain ratios (F/G), while the group Ⅰ was the lowest; AST activities of group Ⅰ were significantly lower than group Ⅱ(P<0.05), LDH activities of groups Ⅰ,Ⅱ and Ⅲ were lower than the control group significantly (P<0.05), and LDH activities of groupⅠ were lower than groups Ⅱ and Ⅲ significantly (P<0.05). TCH concentrations of groups Ⅰ and Ⅱ were higher than group Ⅲ significantly (P<0.05). 本研究旨在探讨饲粮中添加酵母壁多糖对断奶仔猪肠道挥发性脂肪酸和微生物菌群的影响。试验采用单因素试验设计方法,选取180头遗传背景一致、健康状况良好、胎次和体重接近的21日龄断奶仔猪,随机分为4个组,每组5个重复,每个重复9头猪。4个组试验猪分别饲喂对照饲粮(未添加酵母壁多糖)、0.15%酵母壁多糖饲粮、0.30%酵母壁多糖饲粮和0.45%酵母壁多糖饲粮。试验期21 d。结果表明:1)与对照组相比,饲粮中添加0.15%、0.30%和0.45%酵母壁多糖显著提高了仔猪结肠乙酸的含量(P0.05);其中,0.30%和0.45%酵母壁多糖还显著提高了结肠丙酸、丁酸以及总挥发性脂肪酸的含量(P0.05),且二者之间差异不显著(P0.05)。2)与对照组相比,饲粮中添加0.15%、0.30%和0.45%酵母壁多糖显著降低了盲肠沙门氏菌和大肠杆菌数量(P0.05),且0.30%和0.45%酵母壁多糖组之间差异不显著(P0.05)。由此可知,酵母壁多糖可提高仔猪肠道挥发性脂肪酸含量,并改善肠道微生物菌群结构,根据回归方程预测,酵母壁多糖在仔猪饲粮中的适宜添加水平为0.31%~0.40%。 相似文献

The study compared the effects of an antibiotic growth promoter (AGP), yeast (Saccharomyces cerevisiae) and yeast cell wall (YCW) on performance, microbiology and histo-morphology of the small intestine and humoral immune responses in Ross 308 broilers. The treatments (eight replicates/treatment, n = 12/replicate) were negative control (NC, without AGP), positive control (PC, supplemented with bacitracin methylene disalicylate, 400 mg/kg), Y and YCW (supplemented with yeast and YCW, respectively, 1000 mg/kg). Live weight at 42 days improved (p = 0.086) in the PC, Y and YCW groups. Feed conversion ratio was better (p = 0.039) in the YCW group compared with the other groups. Antibiotic growth promoter in the PC group shortened the villi in duodenum (p = 0.044). Mucosal Escherichia coli number was higher in the PC group (p < 0.001), whereas in the digesta E. coli number was lower (p = 0.001) in the PC, Y and YCW groups in relation to the NC. Mucosal Salmonella populations increased (p = 0.0001) in the PC group, whereas in the digesta, all treatments reduced the Salmonella (p = 0.0001). Following oral challenge with Salmonella pullorum, YCW increased E. coli numbers on the mucosa (p < 0.001) whereas in the digesta the Y group had lower (p < 0.0001) number of E. coli. In the digesta, Salmonella count was lower in the YCW group compared with the other treatments (p < 0.01). Yeast cell wall -treated birds exhibited better (p < 0.05) humoral immune response against Newcastle disease which was far more persistent over time than in any other treatments. It was concluded that the yeast and the yeast cell wall may have effects identical to BMD on performance of broilers and thus may constitute an effective replacement strategy in the dietary regimens for broiler chickens. 相似文献

本试验旨在探讨固态酶制剂评估中酶的适宜提取液及固-液分离方法。采用4×3双因素完全随机设计,其中提取液分别为去离子水、乙酸-乙酸钠缓冲液(0.1 mol/L,p H 5.50)、磷酸盐缓冲液(0.05 mol/L,p H 6.00)和0.9%Na Cl溶液;溶液提取后的固-液分离方法分别为不分离、3 000 r/min离心3 min和中速滤纸过滤。每个处理5个重复,每个重复设2个平行,测定各个处理下酶的活性,并考察提取液的类型对酶制剂产品(α-半乳糖苷酶除外)溶解离心后溶液中溶质及蛋白质含量的影响。结果表明,磷酸盐缓冲液溶解木聚糖酶后活性最高(P0.05);乙酸-乙酸钠缓冲液、磷酸盐缓冲液及0.9%Na Cl溶液溶解β-葡聚糖酶后活性相当(P0.05),且均显著地高于去离子水(P0.05);去离子水溶解β-甘露聚糖酶后活性最高,其次为乙酸-乙酸钠缓冲液,两者都显著地高于磷酸盐缓冲液和0.9%Na Cl溶液(P0.05)。提取液类型对α-半乳糖苷酶活性无显著影响(P0.05)。酶制剂溶解后的固-液分离方法对木聚糖酶的测定活性无显著性影响(P0.05);提取液离心或过滤后β-葡聚糖酶活性最高(P0.05);提取液离心后β-甘露聚糖酶活性最高(P0.05);而提取液不分离时α-半乳糖苷酶的活性最高(P0.05)。提取液的种类和酶制剂溶解后的固-液分离方法对4种非淀粉多糖酶的测定活性有极显著的交互作用(P0.01)。乙酸-乙酸钠缓冲液对木聚糖酶制剂的溶解度最大(P0.05),去离子水和0.9%Na Cl溶液均对β-葡聚糖酶及β-甘露聚糖酶制剂的溶解度最大(P0.05)。然而,乙酸-乙酸钠缓冲液溶解木聚糖酶、β-葡聚糖酶、β-甘露聚糖酶后提取液中蛋白质的含量均最低(P0.05)。上述结果表明,乙酸-乙酸钠缓冲液溶解4种固态酶制剂可以最有效地将酶蛋白提取出来,α-半乳糖苷酶提取后不宜固液分离,而其他3种酶的提取液适宜进行离心分离。 相似文献

为初步探讨苦楝对捻转血矛线虫的作用,本试验通过观察苦楝提取物对虫卵和幼虫活性及形态变化的影响,评估其对捻转血矛线虫的驱虫活性,以期为家畜捻转血矛线虫病的防控及植物驱虫药物的研发奠定基础。试验对苦楝水提取物和乙醇提取物设置5个浓度(25、12.5、6.25、3.125、1.562 mg/mL)进行虫卵孵化试验和幼虫活性试验,对不同浓度药物处理的虫卵及幼虫进行计数,并观察药物作用后虫卵和幼虫的孵化状态及形态变化。结果显示,正常孵育的虫卵和幼虫活力良好,苦楝提取物作用48 h后,虫卵呈未孵化、半孵化状态,发育至桑葚期、蝌蚪期及幼虫期死亡,并且各浓度苦楝水提物和醇提物对虫卵均具有抑制作用,其中苦楝皮醇提物对虫卵孵化的抑制效果最佳,在浓度25 mg/mL时,对虫卵孵化的抑制率高达98.2%,在12.5 mg/mL时,苦楝子醇提物对虫卵的抑制率极显著高于苦楝皮醇提取物(P<0.01)。苦楝子提取物对感染性三期幼虫的致死效果较差,在50 mg/mL时对幼虫有较高致死率(82.6%)。苦楝皮水提取物在浓度为50 mg/mL时对幼虫的致死效果尤为突出,幼虫死亡率为97.0%,死亡的幼虫呈"直线型"或"弓型"。经方差分析,在12.5~50 mg/mL浓度范围内,相同浓度苦楝水提取物之间的致死率存在显著或极显著差异(P<0.05;P<0.01),同浓度苦楝醇提取物对幼虫的致死率存在极显著性差异(P<0.01)。综上所述,苦楝提取物对体外的捻转血矛线虫卵和感染性三期幼虫的活性均有抑制作用,其中25 mg/mL苦楝皮醇提物对虫卵孵化的抑制效果最佳,50 mg/mL苦楝皮水提物对感染性三期幼虫的致死作用最好。 相似文献
