西尼罗热病毒RT-PCR检测方法的建立 总被引:3,自引:0,他引:3
参考Genebank发表的西尼罗热病毒(West Nile virus,WNV)E糖蛋白基因序列,自行设计合成一对引物,对WNV进行RT—PCR扩增,产物经琼脂糖电泳分析,呈现一条约400bp的条带,将其克隆入pMD18-T—Vector载体中,并进行序列测定,与已发表的WNV基因比较发现,核苷酸的同源性为99.7%,证实为WNV的E基因,通过对样品多次检测,都能扩增出一条约400bp的条带,表明该方法比较稳定。 相似文献

Liver metabolism is affected by nutrients. The aim of this study was to explore the effects of low‐protein diets (17% crude protein, CP) supplemented with branched‐chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile) and valine (Val), on hepatic amino acid profile and lipid metabolism in growing pigs. The ratio of Leu : Ile : Val in all groups was 1 : 0.51 : 0.63 (20% crude protein, CP), 1 : 1 : 1 (17% CP), 1 : 0.75 : 0.75 (17% CP), 1 : 0.51 : 0.63 (17% CP) and 1 : 0.25 : 0.25 (17% CP) respectively. Results revealed that compared to the positive control group (1 : 0.51 : 0.63, 20% CP), the low‐protein diets significantly augmented the concentrations of most essential amino acids and non‐essential amino acids (p < .05), with the greatest values observed in the 1 : 0.25 : 0.25 group. Moreover, relative to the control, the low‐protein diets with the Leu : Ile : Val ratio ranging from 1 : 0.75 : 0.75 to 1 : 0.25 : 0.25 markedly downregulated the mRNA abundance of acetyl‐CoA carboxylase (ACC), lipoprotein lipase (LPL) and fatty acid‐binding protein 4 (FABP‐4) (p < .05), and upregulated the mRNA expression of hormone‐sensitive lipase (HSL), peroxisome proliferator‐activated receptor‐g coactivator‐1α (PGC‐1α), uncoupling protein 3 (UCP3) and liver carnitine palmitoyltransferase 1 (L‐CPT‐1) (p < .05). Therefore, our data suggest that protein‐restricted diets supplemented with optimal BCAA ratio, that is, 1 : 0.75 : 0.75–1 : 0.25 : 0.25, induce a shift from fatty acid synthesis to fatty acid oxidation in the liver of growing pigs. Efficient red deer supplementary feeding depends on estimations of the nutritive value of offered feeds, frequently estimated with the use of equations derived from domestic ruminants. The aim of this study was to compare the 24‐hour in vitro true dry matter degradability (ivTD24), in vitro gas production (GP) kinetic parameters, GP in 24 hr of incubation (GAS24) and short‐chain fatty acid (SCFA) and microbial biomass (MBS) produced after 24‐hour incubation of feeds in inoculum prepared from sheep and red deer rumen fluid. Eleven feeds, frequently consumed by red deer in Slovenia, which occur either naturally (two fresh grasses, chestnut fruits and common and sessile oak acorns) or are fed as winter supplemental feeds (two grass hays, two grass silages, apple pomace, fresh sugar beetroot), were investigated. The in vitro GP kinetic parameters, GAS24 and ivTD24, did not differ between animal species. Amounts of SCFAs were greater (p < 0.05) when feeds were incubated in sheep inoculum, while molar proportions of acetic and propionic acids did not differ. Molar proportions of butyric acid produced during incubation of high fibre feeds did not differ between animal species, but were higher (p < 0.05) when feeds high in starch or sugar were incubated in red deer inoculum. Greater production of SCFA by sheep rumen microbes suggests better coverage of host animal with energy precursors, while greater production of MBS by red deer rumen microbes suggests better coverage of host animal with protein. Results also suggest that rumens of sheep and red deer are inhabited by different microbial communities, which did not affect the extent of in vitro GP and degradation of feeds used in the present experiment. However, the possibility exists that the divergent nutrient use could be a consequence of different priming by different feeds of the donor animal diets. 相似文献

In order to study and evaluate the feeding environment of the beef cattle farms in Xinjiang, the contents of heavy mentals in the soil from 8 beef cattle farms in Xinjiang Aksu, Yili and Tacheng regions were tested according to the Field of Livestock and Poultry Environment Quality Standard (NY/T 388—1999).The atomic absorption spectroscopic method was used to determine the contents of As, Hg, Pb, Cd, and Cr.The mean contents of As, Hg, Pb, Cd, and Cr in 8 beef cattle farms soil were 0.389, 0.568, 22.708, 0.573 and 149.449 mg/kg, respectively, and they were all below the upper limit of NY/T 388—1999.The comprehensive heavy metal pollution index of 8 beef cattle farms were 0.577 to 0.994, and the index were 0.7 to 1 of two observation points in Yili and one observation point in Tacheng.The soil environment quality of these 3 farms were at the level of slight cleaning.The comprehensive heavy metal pollution index of other five observation points were ≤0.7, the soil environment was in a clean level.The soil environment of beef cattle farms in these three regions conformed to the standards of livestock for environment quality and safe products. 相似文献

During the past two decades, food safety issues in China not only posed serious threats to Chinese consumers but also damaged the image of Chinese products internationally. In China, food safety is not only about scientific discoveries, advanced laboratories, and sanitation equipment; it is more about the role of different players in the food supply chain. The poultry meat supply chain is instrumental in the spread of the avian influenza A virus (H7N9), raising questions about how policymakers respond to such threats and whether industries need to be restructured to manage and control this epidemic so that it does not recur. As a short-term measure, to prevent the spread of this disease, government authorities enforced the closure of live bird markets (LBM) in disease-affected areas of China. However, in the long term, the poultry meat supply chain needs to be restructured. The aim of the current study was to analyze distribution channels for chicken meat in China and then describe arrangements in poultry meat sectors that incorporate small- and medium-scale producers into the supply chain while responding to shifts in LBMs. We also assessed the role of LBMs in spreading H7N9 and how these interventions affect the poultry meat supply chain in the Chinese market. 相似文献

对绿梗菜用睡莲(Nymphaea tetragona)及红梗菜用睡莲的15营养成分和4种重金属含量进行了测定与评价。结果表明,1与104~140种(品种)新鲜蔬菜已知数据比较,绿梗和红梗菜用睡莲均有较高含量的维生素C、钠及较低含量的干物质、蛋白质、钾、钙、镁、铜、锌、磷及硒;绿梗菜用睡莲有较高含量的粗纤维、锰及较低含量的铁,红梗菜用睡莲有较高含量的铁及较低含量的粗纤维。2菜用睡莲是一种含钠量较高的蔬菜。3绿梗菜用睡莲营养成分含量总体高于红梗菜用睡莲。4湖北产区菜用睡莲的铅、砷、镉及汞等重金属含量符合食品安全国家标准的规定。 相似文献

土壤重金属污染是全球普遍存在的问题,生物修复因其环境友好且成本效益高而得到广泛关注。但不同生物修复技术有其优势和局限性,充分了解每种修复技术的特点,才能更经济、有效地对污染土壤进行修复。本研究阐述对比了目前的土壤重金属生物修复方法,包括植物修复(植物挥发、植物固定和植物提取)、转基因植物提取、螯合辅助植物修复、微生物辅助植物修复等技术的机制、优势、局限性和适用性等方面的差异。综述提出有效的生物修复技术需要土壤化学、植物生物学、遗传学、微生物学和环境工程等多学科的有机结合。根据污染土壤的特点,结合具有相应改良特性的转基因植物,是实现污染土壤大面积修复的有效方法。同时,农艺措施对天然超级积累植物的生物量和重金属提取能力的刺激作用还需要进一步挖掘。植物修复可以与其他几种传统修复技术有效结合,利用转基因技术建立土壤+植物+微生物的组合是未来修复技术发展的最佳途径。 相似文献
