Genomewide association study (GWAS), which queries the association between loci and a particular trait by examining single nucleotide polymorphisms (SNPs) of the entire genome, is used in many fields of study. The development of next‐generation sequencing techniques has facilitated GWASs by decreasing the sequencing costs and time. In particular, genotyping by sequencing (GBS) is useful for sequencing many samples simultaneously and at a moderate price. Herein, we describe a potential GWAS using GBS, focused on the apple germplasm, with the goal of developing an effective apple breeding strategy through the identification of useful markers. From 308 Korean apple germplasm, SNPs were selected after GBS, and major traits were investigated. Proprietary individuals were confirmed and grouped by association through genetic diversity and population structure analyses of the selected SNPs. Genes highly associated with the target traits were identified, respectively. As the first GWAS report on the apple germplasm, these results will be useful as base data for GWASs on other apple populations and traits. 相似文献

The present investigation was undertaken to assess the antimicrobial susceptibility of a collection of 58 Gallibacterium isolates. All strains were tested by the broth dilution method using the veterinary fastidious medium. A total of 46 field strains were tested, whereof 23 were clinical isolates from 10 Mexican layer flocks and another 23 isolates originated from 13 clinically healthy poultry flocks in Denmark. In addition, 12 Gallibacterium reference strains that had been isolated some 30–40 years ago were included. 相似文献

Summary Studies on the efficacy in pigs of low level in‐feed medication with the anthelmintic thiophanate at a minimum intake of 6 mg/kg/day for fourteen days are reported. A trial was conducted to compare a group of medicated fattening pigs with a similar unmedicated group on premises known to have a high challenge of Ascaris spp. Daily growth rate was improved whilst feed conversion ratio and the liver condemnation rate were reduced in the treatment group. Routine medication of a whole herd using this regime contributed to a great improvement of the herd production when assessed by the above criteria. User studies in various geographical areas of the Netherlands involving 1500 adult pigs and 1200 fattening pigs medicated with thiophanate in‐feed for fourteen days demonstrated that the compound eliminated the faecal worm egg output and was readily accepted and tolerated by pigs. 相似文献

White spot syndrome virus (WSSV) is the most important pathogen in shrimp farming systems worldwide including the Mekong Delta, Vietnam. The genome of WSSV is characterized by the presence of two major ‘indel regions’ found at ORF14/15 and ORF23/24 (WSSV‐Thailand) and three regions with variable number tandem repeats (VNTR) located in ORF75, ORF94 and ORF125. In the current study, we investigated whether or not the number of repeat units in the VNTRs correlates with virus outbreak status and/or shrimp farming practice. We analysed 662 WSSV samples from individual WSSV‐infected Penaeus monodon shrimp from 104 ponds collected from two important shrimp farming regions of the Mekong Delta: Ca Mau and Bac Lieu. Using this large data set and statistical analysis, we found that for ORF94 and ORF125, the mean number of repeat units (RUs) in VNTRs was significantly lower in disease outbreak ponds than in non‐outbreak ponds. Although a higher mean RU number was observed in the improved‐extensive system than in the rice‐shrimp or semi‐intensive systems, these differences were not significant. VNTR sequences are thus not only useful markers for studying WSSV genotypes and populations, but specific VNTR variants also correlate with disease outbreaks in shrimp farming systems. 相似文献

TRAP是一种新的基于PCR的植物基因型标记技术,具有简单、稳定、效率高的特点。借助日益增长的庞大的生物序列信息,TRAP利用生物信息工具和EST数据库信息,产生目标候选基因区多态性标记。TRAP技术采用两个18核苷酸引物产生标记。一个为固定引物,依据EST序列设计;另一个为随机引物,针对外显子和内含子的特点,设计为分别富含GC或AT核心区的任意序列。PCR扩增前5个循环采用35℃的退火温度,后35个循环采用50℃的退火温度。对不同的植物种类,每一个PCR反应可产生多达50个可统计DNA片段。本文在阐述了TRAP的原理与流程后,对该技术的优势和应用情况进行了总结,并对其前景做了展望。 相似文献

Spinach (Spinacia oleracea L.) is an important leafy vegetable crop grown worldwide. Leaf traits, surface texture (smooth, savoy or semi‐savoy), petiole colour (different shades of green vs. purple) and edge shape (serrate vs. entire), are important commercial traits of spinach. Association mapping for the three traits was conducted on 323 USDA spinach germplasm accessions, originally collected from 33 countries and representing the entire USDA spinach germplasm collection. The majority of accessions were from Europe (36.3%), Asia (25.3%) and North America (15.8%). The majority of the spinach accessions (82.0%) were smooth (unwrinkled types), whereas the savoy and semi‐savoy types (wrinkled types) accounted for 18.0%. The collection contained 74.9% green petiole types, while the purple petioles consisted of 25.1%. The collection consisted of 27.2% serrated leaf types and 72.8% entire leaf edge types. Genotyping‐by‐sequencing (GBS) was used for single nucleotide polymorphism (SNP) discovery, and SNPs were used as genotypic data to conduct genetic diversity and association mapping of the three leaf traits. Five genetic subpopulations and principal components (PCs) were postulated by structure 2 and JMP Genomics 7 for this association panel. Five, seven and 14 SNPs were identified to be associated with surface texture, edge shape and petiole colour, respectively. This study provides us an approach to identify SNP markers through association analysis in spinach and thus leads to select these three leaf traits through marker‐assisted selection in spinach breeding programme. 相似文献

通过对中国436个马铃薯审定品种的系谱分析发现,马铃薯祖先亲本中‘疫不加’、‘竹板’、‘工业’、‘兰芽’和‘早玫瑰’应为第1代亲本,而其衍生系如‘男爵’、‘胜利’、‘燕子’、‘白头翁’、‘米拉’、‘多子白’、‘卡它丁’和‘小叶子’应为第2代亲本。在审定的436个品种中,自行选育的379个品种涉及423个亲本,分别来源于国内品种、国内品系、地方品种、新型栽培种、北美资源、欧洲资源、CIP资源和其他,共8类,且其相应的核遗传贡献分别为97.5、107、10.5、16、27.5、71、33和16.5,表明国内品种和国内品系一直是贡献最大的类型,而地方品种主要以母本为主,外来资源如新型栽培种、欧洲资源和北美资源多以父本为主。为进一步验证其准确性,利用多重引物系统,对278份国内育成品种(品系)和330份国外育成品种(品系)进行了细胞质分型。结果表明,T型和D型为主要细胞质类型,所占数量和比例分别为303个(49.8%)和243个(40%),其次为W、A、M和P型,分别为39(6.4%)、15(2.5%)、6(1%)和2(0.3%),但也发现部分育成品种的细胞质类型与其祖先母本不一致,未发现6种类型以外的其他细胞质类型。 相似文献
