The primary structures of two rainbow trout growth hormone mRNAs (GH1 and GH2) have been deduced by direct sequencing of their respective cDNA clones and portions of the mRNA. Both GH1 and GH2 mRNA contain open reading frames comprised of 630 nucleotides and encode 210 amino acid residues of which 11 are variant. The translated regions of both mRNA are flanked by a short but rather conserved 5′-end, and a relatively long but highly diverged 3′-end. The differences at translated and 3′-untranslated regions suggest that the GH1 and GH2 mRNA originate from different loci. The GH1 and GH2 mRNA are likely transcribed from two distinct loci which were duplicated during tetraploidization of salmonid genome between 50 to 100 million years ago. The GH2 gene has been isolated and sequenced from a rainbow trout genomic library. This gene spans a region of approximately 4 kilobases. The trout GH gene is comprised of 6 exons and 5 introns, in contrast to 5 exons and 4 introns in mammals. The additional intron in the trout gene interrupts the translated regions that are analogous to the last exon of the mammalian counterpart. The alleged internally repeating sequences in mammalian GH, prolactin (Pr1) and placental lactogen (PL) are not observed in the predicted polypeptide sequence of trout GH. In addition, direct repeats that flank exons I, III and V of mammalian GH, Pr1 and PL genes are absent in trout gene. These findings indicate that the rainbow trout GH gene structure does not support the current hypothesis that internally repeated regions in GH, Pr1 and PL arose from a small primordial gene.  相似文献

单核苷酸多态性(SNPs)标记在大多数物种基因组中数量巨大且分布均匀,是许多水产物种遗传研究的理想标记,因此快速发展了几种高通量、快速的SNP标记分型平台。本研究利用Golden Gate分析技术对鲤Cyprinus carpio转录组测序的1 536个SNP标记合成了一张中等密度SNP芯片,对5个鲤群体的480份样本进行基因分型。结果表明:1 235个SNP标记成功分型,约占80%;915个标记至少在一个家系中呈现多态性,占74.1%;标记最小等位基因频率(MAF)介于0.05~0.5之间,平均为0.334,其中48.9%的标记最小等位基因频率(MAF)大于平均值;5个群体中SNP标记平均多态性信息含量(PIC)在0.3左右,为中度多态,杂合度为0.010~0.990,平均为0.5左右,暗示群体具有丰富的遗传多态性,育种价值及性状改良潜力很大。本研究分型的多态性SNP标记可广泛应用于遗传多样性研究、标记-性状关联分析以及分子标记辅助育种。  相似文献

为从分子水平研究巨[鱼丕](Bagarius yarrelli Sykes)的遗传多样性和系统进化关系,本实验对采集于怒江、澜沧江、元江3河流5种群中的60个个体进行12S rRNA和ND3基因序列的PCR扩增,同时与红河河口群体12个个体的12S rRNA和ND3基因序列进行比对分析,采用Mega5.02软件对碱基组成、Kimura-2 parameter遗传距离、可变位点等进行分析。应用DnaSP软件进行遗传多样性相关参数的计算。结果显示:12S rRNA和ND3基因序列长度分别为954 bp和346 bp(349 bp),分别定义了51个单倍型和42个单倍型,12S rRNA和ND3序列中的碱基平均含量分别为:T为20.8%、C为25.7%、A为32.1%、G为21.4%和T为29.5%、C为28.2%、A为28.1%、G为14.1%,表现出明显的A+T偏倚性;6个群体的群体内和群体间的遗传距离分别为0.003~0.284(12S rRNA),0.009~0.059(ND3)和0.004~1.062(12S rRNA),0.008~0.143(ND3);12S rRNA基因的51个单倍型和ND3基因的42个单倍型序列总的单倍型多样性(Hd)、核苷酸多样性(Pi)及核苷酸平均差异数(K)分别为0.972和0.937、0.035和0.079、31.414和27.176,均显示出丰富的遗传多样性。结合从GenBank中下载的12S rRNA和ND3基因同源序列的[鱼丕]科9属10种鱼类进行系统发育树的构建,结果表明巨[鱼丕]独自汇聚成一大单系群,怒江潞江坝群体、怒江三江口群体及澜沧江临沧群体间关系较近,澜沧江景洪群体可单独构成一个单系种群,红河河口群体、红河曼耗群体与其它巨[鱼丕]构成一单系种群后再同澜沧江景洪群体汇聚成一支。  相似文献

We estimated heritabilities of semen production traits and their genetic correlations with litter traits and pork production traits in purebred Duroc pigs. Semen production traits were semen volume, sperm concentration, proportion of morphologically normal sperms, total number of sperm, and total number of morphologically normal sperm. Litter traits at farrowing were total number born, number born alive, number stillborn, total litter weight at birth, mean litter weight at birth, and piglet survival rate at birth. Litter traits at weaning were litter size at weaning, total litter weight at weaning, mean litter weight at weaning, and piglet survival rate from birth to weaning. Pork production traits were average daily gain, backfat thickness, and loin muscle area. We analyzed 45,913 semen collection records of 896 boars, 6,950 farrowing performance records of 1,400 sows, 2,237 weaning performance records of 586 sows, and individual growth performance records of 9,550 animals measured at approximately 5 mo of age. Heritabilities were estimated using a single-trait animal model. Genetic correlations were estimated using a 2-trait animal model. Estimated heritabilities of semen production traits ranged from 0.20 for sperm concentration to 0.29 for semen volume and were equal to or higher than those of litter traits, ranging from 0.06 for number stillborn and piglet survival rate at birth to 0.25 for mean litter weight at birth, but lower than those of pork production traits, ranging from 0.50 for average daily gain to 0.63 for backfat thickness. In many cases, the absolute values of estimated genetic correlations between semen production traits and other traits were smaller than 0.3. These estimated genetic parameters provide useful information for establishing a comprehensive pig breeding scheme.  相似文献

Muskellunge, Esox masquinongy, is an important recreational freshwater fish native to North America. Since muskellunge populations are often maintained through stocking efforts, advances in muskellunge reproductive technologies are of direct relevance to fishery enhancement. We evaluated the efficiency of inbreeding through induced meiotic diploid gynogenesis. Eggs from six female muskellunge were manually stripped and activated using ultra violet‐irradiated yellow perch, Perca flavescens, sperm. Hydrostatic pressure shocking regimes (48 263 kPa) were then applied to the eggs to prevent second polar body expulsion producing unambiguous meiotic gynogens. Six female dams and samples of 12–20 of their gynogenetic progeny were genotyped at seven polymorphic microsatellite loci. Chromosomal recombination frequencies of microsatellite loci based on retention of heterozygosity among gynogens ranged from 0.043 to 0.839 (0.576 ± 0.237). There were no statistical differences in recombination frequency among females at any of the loci. The average inbreeding coefficient (F‐value) ranged from 0.581 to 0.979, equivalent to three to fourteen generations of full‐sibling crosses respectively. The average F‐value overall was 0.712, equivalent to between five and six generations of full‐sibling crosses. Centromere map distances of the seven microsatellite loci ranged from 2.15 to 41.95 cM and meiotic gynogenesis was useful in eliminating heterozygosity at loci proximal to the centromere, but not distal. Since the age at maturity of female muskellunge is approximately 5 years, gynogenesis may pose an expeditious alternative to traditional breeding strategies for creation of homozygous pedigrees for some loci that may be outcrossed to introduce positive heterozygosity effects in the offspring.  相似文献

本文应用RAPD技术,研究了中国虎纹蛙(Rana tigrina rugulosa)和泰国虎纹蛙(R.tigrina cantor)的种内种间遗传多样性及种质特异性分子标记,结果表明:中国虎纹蛙和泰国虎纹蛙多态位点比例(P)分别为53.77%和70.79%;遗传多样性指数(H)分别为0.1345和0.1780;种间的遗传相似系数(S)为0.4238,遗传距离(D)为0.6613。两种虎纹蛙遗传多样性都较丰富,但中国虎纹蛙的遗传变异程度较泰国虎纹蛙低。在OPA、OPF、OPP、OPZ几个系列随机引物的扩增中得到了可用于鉴别两种蛙的12条特异性谱带。  相似文献

采用PCR和DNA测序技术对4个泥鳅(Misgurnus anguillicaudatus)群体的线粒体DNA控制区序列进行比较,研究其遗传多样性和控制区的结构。结果显示,用于分析的线粒体DNA控制区片段序列为918~920bp。32个序列中共发现了56个多态位点,其中32个转换位点,19个颠换位点,5个转换与颠换同时存在的位点,定义了32种单倍型。同时对控制区结构进行分析,识别了其终止序列区(ETAS)、中央保守区(CD)和保守序列区(CSB)的关键序列。4个地理群体的单倍型多样性(Hd)、核苷酸多样性(Pi)和平均核苷酸差异数(K)分别为0.992、0.012和10.698。群体间的平均Kimura双参数遗传距离(Kimura 2-parameter distance,K2-P)、遗传分化指数(Fst)、基因交流值(Nm)和分子方差分析(AMOVA)均表明4个泥鳅群体具有较高的遗传分化,基因交流贫乏。利用核苷酸序列构建的NJ分子系统树揭示4个群体分为2个谱系,除范县群体外,其余各群体间相互交叉聚集。  相似文献

RNA aptamers are artificial nucleic acids that specifically bind to a wide variety of targets. They are an effective tool for pharmaceutical research and development of antiviral agents. Here, we describe four Hirame rhabdovirus (HIRRV)‐RNA aptamers (H1, H2, H3 and H4) that we obtained from an in vitro process called the systematic evolution of ligands by exponential enrichment (SELEX). The HIRRV‐RNA aptamers specifically bind to HIRRV. Hirame natural embryo (HINAE) cells treated with virus and the RNA aptamer showed a decrease in appearance of cytopathic effect when compared with control (treated only with virus). Rhodovulum sulfidophilum was transformed with genes for the RNA aptamers, and the aptamers were detected in the culture medium, indicating that they were secreted from the cells. Thus, the recombinant R. sulfidophilum might be a powerful tool for the prevention of HIRRV in aquaculture.  相似文献

采用RAPD技术对两个人工雌核发育系鲢近交F1遗传距离、遗传相似度及群体遗传多样性进行分析,用普通鲢和鲤作对照。结果表明:GSCⅠ、Ⅱ系近交F1系内个体间平均遗传相似度分别为0.9722、0.9807,高于对照鲢和鲤的0.9546、0.8727;两系近交F1的遗传多样性指数分别为0.0629、0.0529,低于对照鲢和鲤的0.1018和0.2132,表明GSCⅠ、Ⅱ系系内近交的F1保持了较高的纯度。LIPGMA和NJ系统树清晰反映了两个系近交F1与对照组个体问的相互关系。26个随机引物的RAPD扩增结果显示,5个引物OPG04、OPG17、OPP01、OPM11、OPM16可以扩增出区分两个系近交F1的特异标志带。  相似文献
