The effects of lipid-rich bovine serum albumin (LR-BSA) on the development of porcine blastocysts produced
in vitro were examined. Addition of 0.5 to 5 mg/ml LR-BSA to porcine blastocyst medium
(PBM) from Day 5 (Day 0 = in vitro fertilization) significantly increased the hatching rates
of blastocysts on Day 7 and the total cell numbers in Day-7 blastocysts. When Day-5 blastocysts were cultured
with PBM alone, PBM containing LR-BSA, recombinant human serum albumin or fatty acid-free BSA, addition of
LR-BSA significantly enhanced hatching rates and the cell number in blastocysts that survived compared with
other treatments. The diameter, ATP content and numbers of both inner cell mass and total cells in Day-6 and
Day-7 blastocysts cultured with PBM containing LR-BSA were significantly higher than in blastocysts cultured
with PBM alone, whereas LR-BSA had no effect on mitochondrial membrane potential. The mRNA levels of enzymes
involved in fatty acid metabolism and β-oxidation (ACSL1, ACSL3,
CPT1, CPT2 and KAT) in Day-7 blastocysts were
significantly upregulated by the addition of LR-BSA. The results indicated that LR-BSA enhanced hatching
ability and quality of porcine blastocysts produced in vitro, as determined by ATP content,
blastocyst diameter and expression levels of the specific genes, suggesting that the stimulatory effects of
LR-BSA arise from lipids bound to albumin. 相似文献

The objective of the work was to evaluate the effect on the nutrient content of replacing fish meal (FM) with blood meal (BM) in fish supplement. Three isonitrogenous diets (35% crude protein) were formulated using FM as the main source of animal protein (BM0); 50% replacement of FM with blood meal (BM50); or 100% replacement of FM with BM (BM100). The chemical composition (ash, crude protein, crude fat and crude fibre) and amino acid composition were determined. Replacement of FM with BM did not affect the proximate composition of the diet apart from ash content which decreased with the level of substitution. Substituting fish meal with blood meal reduced the levels of methionine, lysine, isoleucine, leucine, proline, valine and increased the levels of arginine, phenylanine and alanine in the diet. Amino acid indices revealed that BM0 had more amino acids with the highest chemical scores followed by BM50 and BM100. In all the diets, methionine was the most limiting amino acid. The essential amino acid index of the diets reduced with the level of replacement of FM (0.94, 0.88 and 0.77). The study showed substitution of up to 50% FM with BM gave a useful protein diet and 100% gave almost a poor protein diet. 相似文献

通过对实验室自制标准物料-豆粕样品中氨基酸含量的多次测定,建立X-S质量控制图,开展水产饲料氨基酸分析日常质量控制。试验按照GB/T 18246-2000,通过微波水解-离子交换-柱后衍生-紫外检测法对豆粕样品氨基酸含量进行连续测定,并以FAPAS奶粉标准物质验证,利用偏度-峰度检验法对离群值进行判断,建立赖氨酸、甘氨酸、氨基酸总量质量控制图。结果显示,质量控制图中豆粕质控样赖氨酸、甘氨酸、氨基酸总量测定的RSD值均小于4%,FAPAS奶粉标准物质测定值落在参考范围内,建立的质量控制图能对日常氨基酸检测结果进行有效监控。结果表明,所建立氨基酸质量控制图直观有效,能对过程存在的异常因素起到预警作用,并可节约成本。 相似文献

Mycosporine-like amino acids (MAAs), a group of small secondary metabolites found in algae, cyanobacteria, lichens and fungi, have become ecologically and pharmacologically relevant because of their pronounced UV-absorbing and photo-protective potential. Their analytical characterization is generally achieved by reversed phase HPLC and the compounds are often quantified based on molar extinction coefficients. As an alternative approach, in our study a fully validated hydrophilic interaction liquid chromatography (HILIC) method is presented. It enables the precise quantification of several analytes with adequate retention times in a single run, and can be coupled directly to MS. Excellent linear correlation coefficients (R2 > 0.9991) were obtained, with limit of detection (LOD) values ranging from 0.16 to 0.43 µg/mL. Furthermore, the assay was found to be accurate (recovery rates from 89.8% to 104.1%) and precise (intra-day precision: 5.6%, inter-day precision ≤6.6%). Several algae were assayed for their content of known MAAs like porphyra-334, shinorine, and palythine. Liquid chromatography-mass spectrometry (LC-MS) data indicated a novel compound in some of them, which could be isolated from the marine species Catenella repens and structurally elucidated by nuclear magnetic resonance spectroscopy (NMR) as (E)-3-hydroxy-2-((5-hydroxy-5-(hydroxymethyl)-2-methoxy-3-((2-sulfoethyl)amino)cyclohex-2-en-1-ylidene)amino) propanoic acid, a novel MAA called catenelline. 相似文献

【目的】研究反刍动物蛙皮素样肽家族多肽及其受体的保守性。为不同动物,特别是反刍动物这2种蛙皮素样多肽及其受体蛋白抗原设计和活性多肽筛选等研究提供参考。【方法】采用生物信息分析学的方法,通过UniProt数据库比较牛的蛙皮素样肽家族胃泌素释放肽(Gastrin-releasing peptide, GRP)和神经介素B(Neuromedin B, NMB)2种多肽及其特异性受体GRP-R和NMB-R与其他动物氨基酸序列的差异性。【结果】13种动物成熟GRP多肽C-端的8个氨基酸序列完全相同,牛GRP氨基酸序列与绵羊、猪和豚鼠最高,分别为88.7%、77.8%和77.8%,与其他动物相似性低于74.1%。10种动物成熟NMB多肽C-端10个氨基酸序列完全相同。牛GRP-R与狗、马和猪等相似性最高,分别为96.1%、94.3%和94.0%,牛NMB-R与猪、人和马相似性最高,分别为92.3%、91.5%和91.0%;牛GRP-R与NMB-R相似性仅为62.2%。【结论】GRP和NMB的C-端氨基酸序列在不同动物之间均具有高度的保守性,而N-端为变化区域,且GRP-R和NMB-R氨基酸序列在动物之间保守性较高。 相似文献

The antibacterial potential of organic acids and essential oil components against Brachyspira
hyodysenteriae, the causative pathogen of swine dysentery, was evaluated. Minimum inhibitory
concentrations (MIC) of 15 compounds were determined at pH 7.2 and pH 6.0, using a broth microdilution assay.
In addition, possible synergism was determined. MIC values for the three tested strains were similar. For
organic acids, MIC values at pH 6.0 were lower than at pH 7.2. B. hyodysenteriae was most
sensitive to cinnamaldehyde and lauric acid, with MIC values <1.5 mM. Most antibacterial effects of binary
combinations were additive, however, for thymol and carvacrol, synergism could be observed. In
vitro results demonstrate the antibacterial action of certain essential oil components and organic

The effects of the severity and timing of leaf removal(LR) on the amino acids of Sauvignon Blanc grapes and wines were studied during the 2017 growing season. High-performance liquid chromatography(HPLC) was used to analyze the amino acids profiles of grape berries and wines. The basal leaves were removed at three time points(40, 56 and 72 days after flowering, named LR40, LR56 and LR72, respectively) at two severity levels(one at which the first, third, and fifth basal leaves of each shoot were removed(50% level); and another at which the first six basal leaves were removed(100% level)). The results showed that leaf removal had little impact on total soluble solids(°Brix), titratable acidity, pH or berry weight. The LR72-50% treated grapes had higher berry weight, titratable acidity and °Brix than those of the other treatments. The highest concentrations of total amino acids and of total amino acids except proline were detected in LR72-50% treated grapes(2 952.58 and 2 764.36 mg L~(-1), respectively); the lowest were detected in LR72-100% treated grapes(2 172.82 and 2 038.71 mg L~(-1), respectively). LR72-50% treatment significantly promoted the synthesis of aspartic acid, serine, arginine, alanine, aminobutyric acid and proline at both severity levels for grapes, the concentrations of all of these amino acids were increased relative to the control concentrations. The LR72-50%, LR40-100% and LR72-100% treated wines had higher total amino acids concentrations and higher concentrations of some individual amino acids, such as arginine, alanine and serine, than did the control wines. Of all the amino acids studied, glycine, tyrosine, cysteine, methionine and lysine were not significantly influenced by the timing or severity basal defoliation in grapes and wines.
