Electrical stunning in water using a high frequency (1000 Hz) power source is a humane and practical method for killing trout, which results in damage free carcasses. However, the electrical power requirement can be high, particularly with high conductivity water. The equipment needed for the high frequency power supply significantly adds to the capital cost of the equipment and can make such equipment impractical, particularly for use in high conductivity water. A two-stage approach to stunning has been demonstrated which reduces the electrical power required by over 80%. The approach is based on the observation that the electric field strength needed to induce rapid insensibility is greater than the electric field strength required to maintain stunned fish in a state of insensibility until they are beyond the point of recovery. It is further proposed that since the electric field required for the prolonged second stage is quite low, it might be possible to use a 50 Hz power source without causing carcass damage.  相似文献

Insulin-receptor binding in skeletal muscle of trout   总被引:2,自引:0,他引:2  
Two hundred rainbow trout (Oncorhynchus mykiss) age 0 +, weight range 11.3 – 11.5 g, were distributed randomly in two groups and maintained for five weeks on either 10% dextrin, or 20% dextrin diet. The fish were sampled 3–5 h and 18–20 h after the last feeding and insulin binding to partially purified insulin receptors in white and red skeletal muscles and to liver plasma membranes was assessed. Plasma insulin, plasma glucose, and liver glycogen content were analyzed in the same fish.Fish fed a diet with higher carbohydrate content (HC) had elevated insulin and glucose levels in peripheral blood, but lower liver glycogen contents compared to the fish fed a diet with lower carbohydrate content (LC). No growth retardation was observed in the fish from HC group.Three to five hours after the last feeding, insulin-receptor binding in white skeletal muscles was higher in HC group of fish, mostly because of an increase in number of high affinity binding sites. Eighteen to twenty hours after the last feeding this difference disappeared. In contrast, the specific binding of insulin to the liver plasma membranes appeared to be lower in the HC group of fish. The lower insulin binding to the liver plasma membranes observed 3–5 h after feeding, could be attributed to the lower quantity of binding sites, while the same phenomenon 18 h after feeding was likely a result of affinity changes. We conclude that higher glycemic levels observed in trout fed a HC diet as compared to LC group of fish, are not a consequence of impaired binding of insulin to its receptors in skeletal muscles.Presented in part at the Annual Meeting of American Society of Zoologists, December 26–30, 1989, Boston, U.S.A. The objective was to compare water quality and fish growth and mortality in a pilot scale recirculating system (RS) and a control tank in flow through system (FTS). The RS was designed after the Danish Model Trout Farm and operated with a make up water renewal rate of 9 m3 kg-1 of fish produced. RS water quality did not decrease significantly with water flow rate decrease in the RS. During the experiment, the RS water treatment system presented solids removal efficiency of 59.6 ± 27.7% d−1, ammonia oxidation of 45 ± 32 g m−3 d−1, oxygenation yield of 392 ± 132 g of O2 kWh−1 and CO2 degassing of 23.3 ± 11.9% pass−1. In the RS, nitrite concentration was 0.15 ± 0.07 mg l−1, close to the toxicity threshold; a N2 supersaturation phenomenon was measured, probably due to the air injection depth. The biofilter and sedimentation area management has to be improved to avoid organic matter decomposition and release of dissolved elements. Even if no N2 over-saturation apparent effect on fish performance and aspect were detected, the airlift depth has to be modified in the case of industrial development of the RS. Some improvements of the water treatment system, especially on the airlift and sedimentation area, are suggested.Concerning fish growth, no significant differences were observed between the RS and the FTS. No pathologies were detected and cumulative mortality rates (0.1%) were similar to the farm's usual data. There were no significant effects of water flow rate decrease in the RS on fish performance and energy savings were recorded to be 0.7 kWh kg−1 of fish produced between RS1 and RS2. The global energy cost of the RS was 3.56 kWh kg−1 of fish produced (0.107 € kg−1 of fish produced). Even if the energy consumption of the water treatment system can be improved, the results confirm that recirculating system can be used for industrial trout on growing, without fish performance deterioration.  相似文献

The aim of the present study was to assess the prevalence of the flavobacteria within farmed trout and to quantify their bacterial burden. A total of 61 fish were sampled from seven farms, and were distributed in two groups: (1) visibly diseased fish suffering from the rainbow trout fry syndrome or the bacterial cold water disease caused by the bacteria Flavobacterium psychrophilum and (2) normally appearing fish. F. psychrophilum cells were titered by qPCR, targeting a specific area of the 16S rRNA gene in skin, muscle, gills, liver, spleen and kidney from all fish. The pathogen was detected in these organs whatever the health status, with titers ranging from 104 to 6 × 107 bacteria/g of tissue in normally appearing fish, thus showing they were bacterial carriers. Two organs allowed differentiation between diseased and normally appearing fish: spleen and kidney, with titers ranging from 106 to 107 bacteria/g of tissue in normally appearing fish vs 1011 to 1012 bacteria/g of tissue in diseased fish. No relationship was found between immunoglobulin M-like titer in plasma and health status. Gene expression analysis in fish organs revealed two genes that were markers of the bacterial infection: mt-a and il-1β genes encoding the metallothionein A and the interleukin1-β, respectively. These genes were both over-expressed in gills, liver, spleen and kidney of diseased fish. Four genes encoding immunity markers were down-regulated in spleen (a key organ implicated in immunity) of diseased fish: tgf-β, cd8-α, mhc2-β and igt, suggesting a weakened immune system in diseased fish.  相似文献

Off flavor characterization and origin in French trout farming   总被引:1,自引:0,他引:1  
The results of a study on off-flavor problems in four French trout (Onchorynchus mykiss) farms are presented. Methodological aspects on sensory analyses and volatile compounds quantification in fish are discussed. Detection of odorous compounds by gas chromatography–mass spectrometry shows that significant concentrations of geosmin (up to 18 μg/kg in fat) are found in trout. The sensory evaluation, the presence/absence of geosmin in the flesh and the identification of off-flavor compounds producers demonstrate the implication of Microcoleus in the appearance of earthy/musty off-flavors. The presence of this particular cyanobacterium is linked to the deterioration of water quality during the water recirculation period. Correlations between chemical and sensory detection in the flesh indicate that the taste evaluation enables the differentiation of the four categories “non-tainted”, “slightly-tainted”, “tainted” and “strongly-tainted”. However, the average concentrations of geosmin found for these different intensities are relatively limited, from 0.2 to 4.9 μg/kg. Finally, recommendations are made to allow a more effective control of off-flavor occurrences.  相似文献

The magnitude of the cortisol response to a standardised stressor is a heritable trait in salmonid fishes, and selection for stress responsiveness induces differences in both behaviour and neuroendocrine function. For instance, in laboratory studies, fish with a high cortisol response show a greater propensity for stress induced anorexia. Some authors have, however, commented that controlled studies encompassing relatively small groups of fish hold little or no relevance to practical aquaculture. This notion may be flawed, since understanding the mechanisms behind the behaviour of individuals is a proviso to predict behaviour in groups even with the caveat that some behaviors may be modified by group size. As an example, optimal feeding regimes should be easier to predict in a population consisting of individuals whose appetite is relatively less affected by external stressors. In a fluctuating and potentially stressful environment, such a population should also generate less feed waste, if kept on fixed rations. In the present experiment, we tested this hypothesis by monitoring feed waste and feed conversion efficiency in lines of rainbow trout selected for a low (LR) or high (HR) cortisol response to stress. The study was carried out after fish had been transported between rearing sites in the United Kingdom and Norway. There was significantly more feed waste from rearing units containing HR fish, and these fish also showed lower feed efficiency (growth per unit feed consumed). The difference in feed waste became more apparent with increasing time after transport, as rations increased. Simultaneously, size was more variable and growth was slower in HR rearing units. These results suggest that there are several potential benefits of selection for low stress responsiveness in aquaculture.  相似文献

Brook Trout (Salvelinus fontinalis) is an important fish species in Ontario, Canada, supporting recreational fisheries that contribute significantly to local economies. Hydroelectric dams disrupt the river continuum, altering downstream conditions and impacting riverine fish populations. Specifically, Brook Trout activity has been found to increase during hydropeaking periods, when dam operators rapidly increase river discharge to meet electricity demands. Higher energetic outputs driven by hydropeaking may decrease the energy available to allocate towards fish growth and condition, negatively impacting Brook Trout. We investigated the impact of two different hydropeaking regimes on resident Brook Trout populations downstream from a 15‐MW dam used for hydropeaking, compared to a population in a nearby naturally flowing river. Length‐at‐age as determined by otolith back‐calculations was higher in the regulated river relative to the naturally flowing river. Muscle tissue caloric content and weight–length relationships did not differ between rivers. Field metabolism, as inferred from fish otolith δ13C values, was higher in the regulated river relative to the naturally flowing river and was significantly positively related to time spent hydropeaking. Higher metabolic outputs in the regulated river were likely offset by an increased food supply, allowing for higher Brook Trout length‐at‐age. The opposing and complicated impacts of river regulation on Brook Trout highlight the need for studies to consider multiple indicators of fish health when characterising the response of fish populations to river regulation.  相似文献

European water legislation enforces increasingly restrictive measures with regards to reduction of water consumption and waste emission in order to minimise the potential environmental impact of the agro industry sector. Fish farms are particularly concerned, but legislation covering effluent discharge varies significantly from country to country. However, recommendations and directives from institutional, national or regional bodies suggest the enforcement of increasingly strict waste reduction measures and the development of waste treatment. Before treatment, it is necessary to evaluate waste production in terms of composition and quantity. The waste quantification methods used today for fish culture systems are either based on direct measurements of nutrient and suspended solid fluxes or on indirect evaluation based on the digestibility coefficients of the feed constituents. The objective of the present study is to evaluate the waste of a freshwater flow through farm using both approaches and to discuss their applicability, drawbacks and advantages from the viewpoints of fish farmers and control authorities. Waste production on the farm was monitored during several 24 hour cycles in order to characterise the effluents of the system. The predictions and measurements for the total nitrogen (TN) parameter were well correlated, but measured and predicted suspended solids (SS) and total phosphorus (TP) values presented a weaker correlation coefficient. The hydrobiological method gives details on the N and P forms of waste but this method is heavy and it is difficult to obtain representative samples and flow rate measurements. The nutritional method is the simplest to use, provided that feed data are available.  相似文献
