用随机扩增多态性DNA(RAPD)技术对泰国产方斑东风螺养殖群体的遗传多样性进行检测,从100个随机引物中筛选出21个引物对方斑东风螺的DNA进行扩增,结果表明:21个引物共检测到222条清晰且重复性好的条带,每个引物可扩增出4~16条带,分子量在200~2200bp之间,其中多态位点为156个,占70.27%;群体的Shannon多样性指数为0.2818,Nei基因多样性指数为0.2491;个体间最大遗传距离为0.291,最小遗传距离为0.066。通过与其他贝类遗传多样性的研究结果比较,可初步判断泰国产方斑东风螺养殖群体的遗传多样性比较丰富。 相似文献

Genetic diversity of 54 accessions of Aegilops tauschii from five countries was assessed using sequence-tagged microsatellites (or simple sequence repeats, SSRs) and amplified fragment
length polymorphisms (AFLPs). In the case of AFLP analysis, a total of 256 amplification products obtained, 234 of them were
polymorphic across all the 54 accessions. A total of 224 fragments were obtained from the 24 SSR primers and 219 of fragments
were polymorphic across all the genotypes screened. Based on both AFLP and SSR markers, the highest percentage of polymorphisms
were obtained in Iranian and accessions of unknown origin. The highest polymorphic information content (PIC) value was observed
for SSRs (0.82) while the highest marker index (MI) value was for AFLPs (8.5) reflecting the hyper-variability of the first
and the distinctive nature of the second system. Principal co-ordinate analysis (PCO) revealed congruent patterns of genetic
relationships for both data sets, but did not group accessions strictly according to their geographical origins. Poor correlation
was found between AFLP and SSR marker loci. This low association may be due to low number of AFLP and SSR markers. These results
show that molecular markers can help to organize the genetic variability and expose useful diversity for breeding purposes. 相似文献

Total 65 lotus accessions in genus Nelumbo mainly collected from China, were subjected to random amplified polymorphic DNA (RAPD) markers to estimate the genetic diversity
and to test the genetic basis of the relationships between morphotypes and molecular markers. Seventeen primers generated
a total of 195 highly reproducible and discernible loci, among which 173 were polymorphic. Percent polymorphism varied from
66.7 to 100 with an average of 88.72, and five primers out of them, OPC05, OPG10, OPN20, OPP09 and OPS17, showed 100% polymorphism.
A relatively high genetic diversity was detected among all the samples with the similarity coefficient values ranging from
0.45 to 0.85, and Nei’s gene diversity (h) 0.30, and Shannon index (I) 0.46. The UPGMA dendrogram clustered 65 accessions in four clusters and the clustering pattern showed two groups, N. nucifera ssp. nucifera and those accessions related to the American lotus, and some special cultivars, landraces, hybrids and the American lotus.
Principal Coordinate Analysis (PCA) further indicated that the genetic diversity of Nelumbo accessions was not evenly distributed, instead, was presented by a clustered distribution pattern. Similar to the results
revealed by the dendrogram, two main groups representing the two subspecies of N. nucifera, as well as some special landraces, cultivars of Chinese lotus, the Japanese lotus and hybrids out of the two groups were
obtained. Neither the UPGMA dendrogram nor the PCA analysis exhibited strict relationship with geographic distribution and

Pistachio (Pistacia vera L.) is a strategic nut tree species in the Middle East which holds comparative advantage over other fruit trees in view of
its hardiness, income generation opportunities and benefits for the ecosystem. Yet pistachio cultivation depends on a very
narrow genetic base, in spite of the existence of many varieties still marginally exploited. Syria is an important center
of diversity for pistachio. A country wide ecogeographic survey in this country was carried out to determine the extent of
pistachio genetic diversity and its use. As a whole, 114 accessions were collected from 37 farms to assess diversity at morphological
and molecular level. Molecular evaluation was carried out using Amplified Fragment Length Polymorphism (AFLP) technique and
performed using seven primer pair combinations. Results from the studies allowed the identification of 25 pistachio female
varieties in Syria, some of which unique and described for the first time. Three groups of pistachio diversity were identified
by cluster analysis which provides useful information about the distribution of genetic diversity in Syria for enhanced use

There is a growing realisation world over that the introduction of modern agriculture has to be supplemented with measures
to conserve biodiversity in situ if yield gains are to be stabilized. Hence, there is a growing interest from agricultural development specialists and conservation
biologists for understanding the socioeconomic factors determining the conservation of biodiversity in situ. The present study was conducted with the objective of understanding the in situ (on-farm) conservation of agrobiodiversity in traditional agroecosystems taking the Urgam valley in north-western Himalaya
of India, as a case study. An inventory was made of traditional crops and wild economic species for subsistence, and the structure
of forest resource base, traditional knowledge related to resource management and use. Institutional and scientific challenges
for in situ (on-farm) management of crop diversity were studied and are discussed in this paper. Complementarity of in situ (on-farm) conservation with ex situ conservation together with crop improvement in such marginal areas are suggested. 相似文献

A total of 1027 genotypes of wild Lactuca spp. (L. serriola, L. saligna, and L. aculeata), originating from Israel, Jordan, East Turkey, Armenia, Kazakhstan, China, and various other countries in Europe were screened
for resistance to the lettuce downy mildew (Bremia lactucae Regel). The genotypes of L. serriola (N = 962); L. saligna (N = 43); and L. aculeata (N = 22) were tested at the seedling stage with two isolates of B. lactucae (race Bl-21 and Isr-74) isolated from cultivated lettuce in Italy and Israel, respectively. Altogether, the isolates possess
virulence against 17 out of 20 Dm genes/R-factors composing the established differential set of lettuce lines/varieties. They could therefore be used for preliminary
detection of the presence of new resistance factors. A total of 83 and three genotypes of L. serriola and L. saligna, respectively, were resistant to infection by each of the two isolates alone as well as to the infection by a mix of the
two isolates. These genotypes probably carry previously unknown resistance, and therefore could be suitable for exploitation
by breeders and further detailed research. The wild progenitor of lettuce, L. serriola, and related species harbor significant untapped genetic resources for lettuce improvement. 相似文献

The genetic relationships among 33 coconut germplasm accessions were analyzed using RAPD markers. The germplasm accessions were collected from various coconut growing regions viz. South Asia (SA), South East Asia (SEA), South Pacific (SP), Atantic and America, and Africa. Forty-five random primers produced a total of 399 polymorphic markers. The Polymorphism Information Content (PIC) ranged from 0.031 to 0.392 and the Marker Index (MI) ranged from 6.28 to 0.031 among the primers. Based on the MI a set of 5, 10 and 15 informative and reproducible primers were identified. The mantel matrix correlation was calculated to compare the similarity matrices of a set of reproducible informative primers and global primers. There was significant correlation among the similarity matrices (r ≥ 0.50). The similarity matrix based on 399 polymorphic markers was used to construct the dendrogram to show the genetic relationship among the accessions. Similarity values ranged between 0.573 and 0.846. There was less genetic similarity (based on Jaccard's coefficient) among South Pacific and South East Asian accessions. The clustering pattern obtained in the present study was in agreement with the earlier reports based on RFLP, SSRs and AFLPs. 相似文献

Lupinus angustifolius L. is a Mediterranean species, domesticated in the 20th century, representing an important grain legume crop in Australia
and other countries. This work is focused on the collection of wild germplasm and on the characterisation of morphological
and molecular diversity of germplasm accessions. It reports the collection of 81 wild L. angustifolius accessions from the South and Centre of Portugal, available at the ‘Instituto Superior de Agronomia Gene Bank’, with subsequent
morphological and molecular characterisation of a selection of these and other accessions. A multivariate analysis of morphological
traits on 88 L. angustifolius accessions (including 59 wild Portuguese accessions, 15 cultivars and 14 breeding lines) showed a cline of variation on wild
germplasm, with plants from Southern Portugal characterised by earlier flowering, higher vegetative development and larger
seeds. AFLP and ISSR molecular markers grouped modern cultivars as sub-clusters within the wider diversity of wild germplasm,
revealing the narrow pool of genetic diversity on which domesticated accessions are based. The importance of preserving, characterising
and using wild genetic resources for L. angustifolius crop improvement is outlined by the results obtained. 相似文献

As a part of an in situ survey of wild rice (Oryza rufipogon Griff.) in Myanmar (Burma), 16 strains of wild rice were collected, and analyzed for allelic diversity over 74 loci with
simple sequence repeat (SSR) markers to obtain a basic information for their conservation. Three each of indica and japonica cultivars were added for a comparison. In the six cultivars and 16 strains of wild rice, three to 15 alleles were detected
per locus with an average of 7.9. The wild rice revealed a large number of unique alleles throughout their chromosomes with
much wider ranges of variation than those detected in the six cultivars of O. sativa L.. The alleles found in the wild rice were classified into those specific to wild rice, common to wild rice and cultivars,
and those similar to indica or japonica cultivars. According to the classification, the genotype of each of the 16 strains of wild rice was schematically depicted.
The genetic variation among individual strains within a collection site was larger than the variation among the collection

As an oilseed crop, the cultivation of Ethiopian mustard (Brassica carinata) is restricted only to Ethiopia. Even though geographic diversity is a potent source of allelic diversity, the extent of genetic diversity among germplasm material of Ethiopian mustard from different countries has not been assessed. Forty-three accessions, comprising 29 accessions from eight different geographic regions of Ethiopia and 14 exotic accessions from Australia, Pakistan, Spain, and Zambia were analysed for their genetic diversity using random amplified polymorphic DNA (RAPD) technique. A set of 50 primers yielded a total of 275 polymorphic bands allowing an unequivocal separation of every Ethiopian mustard accession. The usefulness of the 50 RAPD primers in measuring heterozygousity and distinguishing accessions was variable such that polymorphic information content (PIC) varied from 0.05 to 0.40, band informativeness (BI) from 0.05 to 0.65 and primer resolving power (RP) from 0.15 to 6.83. Jaccard's similarity coefficients ranged from 0.44 to 0.87 indicating the presence of a high level of genetic diversity. On the average, Australian and Ethiopian accessions were the most similar while, Spanish and Zambian accessions were the most distant ones. Cluster analysis grouped the 43 accessions into four groups, which has quite a high fit (r = 0.80) to the original similarity matrix. With no prior molecular information, the RAPD technique detected large genetic diversity among the 43 accessions from five different countries and their grouping by dendrogram and principal coordinate analysis (PCoA) was inclined towards geographic differentiation of RAPD markers. Conversely, RAPD differentiation along geographic origin was not apparent within the Ethiopian accessions. 相似文献
