This study uses non-invasive evoked surface electromyography (SEMG) to investigate postnatal muscle development in pigs, and to assess any correlation between recorded signal parameters and muscle fibre types in two different skeletal muscles. Four litters (n = 43) of Large White × Landrace pigs were used. Evoked SEMG measurements were taken on days 2, 5, 14, 26, 60 and 151 post partum from m. Longissimus dorsi (LD) and on days 14, 26, 60 and 151 post partum from m. Biceps femoris (BF). A third of each litter was slaughtered at days 27, 61 and 153 post partum. Biopsy samples for LD and BF were taken to categorize muscle fibre types. For LD there was a significant increase in compound muscle action potential (CMAP) parameters from day 2 to day 5 post partum, whilst for BF significant increases occurred from days 14 to 26 post partum. Fibre type development in both muscles showed a significant decrease in type IIA fibre number (P < 0.001) and an increase in type IIB fibre number (P < 0.001). Analysis of CMAP parameters in relation to fibre type percentages showed significant positive correlations between percentage of type I fibres in BF and CMAP Area (r = 0.71; P = 0.05), SlopeL (r = 0.79; P = 0.02) and Corr Peak (r = 0.78; P = 0.02) and a negative correlation with SlopeT (r = 0.89; P = 0.003) at day 151. A greater CMAP Area and Corr Peak as well as steeper leading and trailing slopes were on the other hand correlated to fewer type IIB fibres.

It is concluded that 1) changes in the evoked CMAP are muscle-specific during early postnatal development in the pig, and 2) in spite of a small sample size, the correlations between CMAP signal parameters and fibre type percentages warrant further investigation.  相似文献

The aim of the current study was to investigate the effects of a porcine growth hormone releasing hormone (pGHRH) gene plasmid injection in piglets on growth performance and whole body protein turnover. Sixty male Canadian Landrace × Chinese Taihu piglets were assigned to an intramuscular injection of 0 (control), 0.25, 0.5, 1 and 2 mg. All pigs were fed with the same diet (crude protein: 239.8 g/kg, digestible energy: 14.28 MJ/kg) at ad libitum intake. Protein turnover was determined on the 22nd day with a three-pool model by using a single-dosage, end-product analysis method with 15 N-glycine as a tracer. Injection of the pGHRH gene plasmid increased the piglets' growth rate, altered feed intake and decreased feed conversion ratio. It increased plasma growth hormone releasing hormone (GHRH), growth hormone (GH), insulin-like growth factor-I (IGF-I) and somatostatin but reduced serum urea and triglyceride. It reduced the urinary nitrogen excretion and led to higher nitrogen retention as well as the efficiencies of nitrogen retention and digestible N utilization. It increased the rates of protein synthesis, protein breakdown and net protein gain. Excretion of endogenous urinary nitrogen was reduced and nitrogen reutilization rate was improved. Conclusions: Injection of the pGHRH gene plasmid in skeletal muscle stimulated GHRH, GH and IGF-I excretion in piglets. Protein deposition was increased by an increase in protein synthesis and a smaller increase in protein breakdown, which was accompanied by reducing amino acid oxidation and increasing nitrogen reutilization.  相似文献

ABSTRACT:   This study is the first to report on the high occurrence of agglutinated pelvic fin membrane deformities in hatchery-reared black rockfish Sebastes inermis . For 5 years, this symptom was marked in hatchery-reared fish, with 58.7% of fish deformed on average (varying between 46.7 and 72.0%). The deformity was a peculiarity in hatchery-reared fish, but is negated in wild fish, and was not related to whether the fish broodstock originated from the wild or from a hatchery. Mark–release experiments showed that deformed fish were almost the same as normal hatchery-reared fish in growth and survival rates, and, theoretically, the recapture numbers of hatchery-stocked fish, estimated by deformity, almost coincided with actual recapture numbers, confirmed by otolith tagging. The results of the present study indicate that deformity in hatchery-reared black rockfish is useful as a stock separation tool.  相似文献

为了研究哺乳期磺胺间甲氧嘧啶(SMM)暴露对仔鼠骨骼肌蛋白质代谢及mTOR信号通路的影响,以0、10、50、200 mg/(kg·d)剂量对哺乳期ICR小鼠灌胃给药直至出生后21 d;第22天断乳时剖杀部分仔鼠,采集腓肠肌,BCA法测定其腓肠肌总蛋白含量,氨基酸分析仪测定其游离氨基酸水平,RT-PCR法检测其mTOR信号通路关键基因表达水平;另一部分仔鼠以性别分笼喂养至出生后63 d,并每周称重1次。与对照组比较,结果表明仔鼠体重无统计学差异(P0.05);中剂量组仔鼠腓肠肌谷氨酸(Glu),甘氨酸(Gly),丙氨酸(Ala),瓜氨酸(Cit),蛋氨酸(Met),组氨酸(His)和鹅肌肽(Ans)含量显著升高(P0.05);高剂量组的Cit和Ans含量也显著升高(P0.05),而低剂量组的Cit含量显著降低(P0.01);仔鼠腓肠肌Mtor,Pi3k3ca,Pi3k3cb,Akt1,Eif4ebp1和Rps6kb1等基因表达无统计学差异(P0.05);腓肠肌总蛋白含量无统计学差异(P0.05)。哺乳期SMM暴露对仔鼠骨骼肌氨基酸代谢有一定的改变作用;对mTOR信号通路并未产生显著影响,为临床上探讨生命早期SMM暴露是否引起代谢性疾病问题提供了毒理学风险评估依据。  相似文献

AIM: To observe the effect of endogenous nitric oxide synthase(NOS) inhibitor asymmetric dimethylarginine(ADMA) and its signaling pathways on NO levels and skeletal muscle contractility in 4-week running rats. METHODS: The 4 weeks running rat model was established. The twitch tension, tetanic tension and the fatigue test of soleus muscle induced by electrical stimulation ex vivo were detected. The ATP content, mitochondrial DNA levels and the mRNA expression of peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α), nuclear respiratory factor(NRF) were measured to reflect the mitochondrial function and biosynthesis in the skeletal muscle. Serum ADMA concentration was detected by high performance liquid chromatography. The endogenous ADMA enzymes PRMT1 and 2 subtypes of ADMA metabolism enzyme DDAH, 3 subtypes of NOS protein expression in the skeletal muscle were determined by Western blot. NOS activity and nitric oxide(NO) content were analyzed by colorimetric method. RESULTS: Compared with normal control group, the twitch tension, tetanic tension and the anti-fatigue capability of soleus muscle in running group were significantly enhanced, ATP content, mitochondrial DNA content and the mRNA expression of PGC-1α and NRF were significantly increased(P<0.01). In addition, the protein expression of constitute type NOS(cNOS) and NOS activity were significantly increased(P<0.01), but the increase in NO content was relatively smaller in soleus muscle in exercise group(P<0.05). Moreover, serum ADMA concentration in running group was increased, while the DDAH2 expression in skeletal muscle was decreased.CONCLUSION: Short-term endurance exercise enhances the twitch tension, tetanic tension and fatigue resistance of soleus muscle. The mechanism may be that increased cNOS expression feedbacks to increase ADMA concentration, thus maintaining the increase in NO synthesis at a relatively low level, and resulting in promoting skeletal muscle mitochondria biosynthesis and mitochondrial function.  相似文献

Background

Mucopolysaccharidoses (MPS) are common lysosomal storage disorders causing typically progressive skeletal and ocular abnormalities.

Objectives

To describe the clinic features, metabolic profile and a unique mutation in a domestic shorthair (DSH) kitten with MPS VII.

Animals

Affected kitten and 80 healthy cats.

Methods

Serum lysosomal enzyme activities and urinary glycosaminoglycan (GAG) accumulation were assessed. Exons of the β‐glucuronidase gene (GUSB) were sequenced from genomic DNA and genotyping was conducted.

Results

A 3‐month‐old DSH cat was presented for stunted growth, paresis, facial dysmorphia, multiple skeletal deformities, and corneal opacities. Evaluation of blood smears disclosed metachromatic granules in leukocytes and a urinary mucopolysaccharide spot test was positive. The proband had no GUSB activity but normal or increased activities for other lysosomal enzymes. Sequencing of the GUSB gene from the proband and comparison to the sequence of 2 healthy cats and the published feline genome sequence demonstrated 2 unique single base transitions (c.1421T>G and c.1424C>T) in exon 9, altering 2 adjacent codons (p.Ser475Ala and p.Arg476Trp). These amino acid changes are in a highly conserved domain of the GUSB protein and nontolerable to maintain function. Moreover, the p.Arg476Trp mutation previously has been identified in human patients. None of the other clinically healthy cats had these mutations.

Conclusions and Clinic Importance

The diagnostic approach to MPS disorders is delineated. This is only the second mutation known to cause MPS VII in cats. Similarly, 2 different mutations have been described in MPS VII dogs, thereby showing the molecular heterogeneity of MPS VII in companion animals.  相似文献

This is a study of vertebral deformities in ornamental koi based on computed radiography and skeletons cleaned by dermestid beetles (Dermestes maculatus). All koi developed gradual onset of swimming abnormalities as adults. Extensive intervertebral osteophyte formation correlated with age of fish and was associated with hindquarter paresis in one koi. Vertebral compression and fusion were the most common spinal deformities occurring at multiple sites, similar to findings in other farmed fish. Site‐specific spinal deformities were thought to develop due to differences in swimming behaviour and rates of vertebral growth. One koi had offspring with spinal deformities. Spinal deformities are significant problems in both European and Australian food fish hatcheries. The heritability of vertebral deformities in farmed fish is reportedly low unless there is concurrent poor husbandry or nutritional deficiencies. The specific aetiologies for vertebral deformities in koi in this study could not be ascertained. Current knowledge on spinal deformities in the better studied European food fish species suggests multifactorial aetiologies. Future research should include prospective longitudinal studies of larger numbers of koi from hatch and consideration of all potential risk factors such as husbandry, nutrition, temperature, photoperiod and genetics.  相似文献
